Comparison between the Effects of Intraperitoneal Injection of LDL and Intravenous Injection of LDL on Arterial Endothelial Cells Apoptosis

To observe the effect of oxidized low density lipoprotein (OxLDL) on arterial endothelial cells apoptosis in vivo, we established a model in which Sprague-Dawley rats were given intraperi-toneal and intravenous injection of unmodified LDL (8 mg/kg every day) via the tail vein. Seven days after the injection, the aortic endothelial cells specimens were prepared by an en face preparation of rat aorta. The apoptotic cells were identified and counted by in situ nick and labelling (TUNED method and light microscopy. The numbers of the apoptotic cells were 12. 52±4. 71/field in the in-traperitoneal injection control group, 11. 41±2. 94/field in the intravenous injection control group, 22. 98±8. 01/field in the intraperitoneal injection LDL group and 103. 8±11. 5/field in the intravenous injection LDL group, respectively. The difference was significant between injection LDL group and control (P<0. 01), and the difference was also significant between two LDL injection groups (P<0. 01). These findings suggest that injection of LDL can induce apoptosis in arterial endothelial cells and the effect is especially significant with intravenous injection LDL. After injection, oxidative modification of LDL may occur in local arteries and causes injury to the endothelial cells.

An algorithm for solving the graph isomorphism problem with 3-D DNA structures is proposed in this paper. The k-armed branched junction molecules are used to code k-degree vertices. Double stranded molecules are used to code edges. Then the molecules are mixed in a tube to be ligated. The result can be detected by gel electrophoresis. The time complexity of the algorithm is O(n2), where n is the number of vertices of the graph.

By using Fura-2/AM, the effects of magnesium (Mg 2+) on the glutamate-induced increase of intracellular free calcium ([Ca 2+]i) in the cultured hippocampal neurons and the features were investigated by integrated photoelectric detecting system. The experiments were designed to three groups (The drug was spit to the cells for 20 s): Group A receiving 1×10 —5 mol/L glutamate; Group B receiving 1×10 —5 mol/L glutamate and1×10 —5 mol/L Mg 2+ simultaneously; Group C receiving 1×10 —5 mol/L glutamate again after [Ca 2+]i in group B back to the baseline. The results showed that in group A, [Ca 2+]i was obviously increased. In group B, the changes in [Ca 2+]i and the peak value were significantly decreased. Moreover, the elevation of Phase 1 was slowed down and Phase 2 was shortened to some extent, and the plateau phase between them was relatively prolonged. In group C, calcium oscillation similar to that in group A occurred, but both the Phase 1 and Phase 2 were shortened and the △[Ca 2+]i was slightly decreased. It was suggested that Mg 2+ could quickly inhibit the rise of [Ca 2+]i induced by glutamate in the cultured hippocampal neurons in rats.

Changes in Serum Leptin Levels during r-hGH Treatment in Growth Hormone-Deficient Children

To observe the effect of growth hormone on serum leptin levels, serum leptin concentrations were measured by enzyme immunoassay in 12 prebutal children with growth hormone deficiency 1, 3 and 6 months before and after the treatment with recombinant human growth hormone (r hGH). For comparison, 34 normal prepubertal children were also investigated. Relationship between leptin levels and body mass index (BMI) was observed at the same time. Our results showed that serum leptin level in normal prepubertal children was 1.22±0.34 ng/ml; the pretreatment serun leptin levels in GHD children was 3.08±2.41 ng/ml, which was significantly different from those 1, 3 and 6 months after GH treatment (i.e. 1.64±1.37 ng/ml,1.57±1.40 ng/ml and 1.35±0.89 ng/ml respectively) (all P <0.001). Our results suggested that r hGH has a suppressive effect on leptin expression.

In order to increase the positive detection rate of HCV RNA in the patients with chronic hepatitis C , RT PCR was used to synchronously detect HCV RNA in the plasma and peripheral blood mononuclear cells of 583 CHC patients with a continuously elevated level of ALT for more than one year. The results showed that the positive detection rate of HCV RNA in the plasma of the CHC patients was 19.2 %, while 24.5 % in PBMC. It was demonstrated that the positive detection rate for HCV RNA in PBMC was obviously higher than that detected in plasma. To synchronously detect HCV RNA in PBMC by using RT PCR can increase the positive detection rate of HCV RNA in the CHC patients.

Suramin Inhibits the In Vitro Expression of Encephalitis B Virus Proteins NS3 and E

In this study, the mechanism by which Suramin inhibits the replication of epidemic encephalitis B virus was explored to provide a theoretical basis for its further application in clinical practice. After viral infection of HepG2 and IMR 32 cells, different concentrations of Suramin were added to the culture media, and then the cultural supernatants and infected cells were collected 48 h later. For the evaluation of the curative effect, cytopathic effect (CPE), virus titers, the expression of viral protein and viral RNA were determined by Western blot, RT PCR and in vitro RNA synthesis, respectively. At the concentration of 50 μg/ml of Suramin, HepG2 and IMR 32 infected with epidemic encephalitis B virus decreased by 51.8 % and 0.03 % respectively, as compared with controls. It was suggested that expression of encephalitis B virus proteins NS3 and E was notably reduced by Suramin. This is especially true of E protein. At RNA level, however, no difference in RNA virus was found between Suramin treated virus and non treated cells. Our results suggest that Suramin can inhibit viral replication by blocking the production of viral proteins.

6-OHDA Induces Cycle Reentry and Apoptosis of PC12 Cells through Activation of ERK1/2 Signaling Pathway

This study investigated the effect and mechanism of cell cycle reentry induced by 6-hydrodopamine （6-OHDA） in PC12 cells. By using neural differentiated PC12 cells treated with 6-OHDA, the apoptosis model of dopaminergic neurons was established. Cell viability was measured by MTT. Cell apoptosis and the distribution of cell cycle were assessed by flow cytometry. Western blot was used to detect the activation of extracellular regulator kinasel/2 （ERK1/2） pathway and the phosphorylation of retinoblastoma protein （RB）. Our results showed that after PC12 cells were treated wtih 6-OHDA, the viability of PC12 cells was declined in a concentration-dependent manner. Flow cytornetry revealed that 6-OHDA could increase the apoptosis ratio of PC12 cells in a time-dependent manner. The percentage of ceils in G0/G1 phase of cell cycle was decreased and that in S phase and G2/M phase increased. Simultaneously, ERK1/2 pathway was activated and phosphorylated RB increased. It was concluded that 6-OHDA could induce cell cycle reentry of dopaminergic neurons through the activation of ERK1/2 pathway and RB phosphorylation. The aberrant cell cycle reentry contributes to the apoptosis of dopaminergic neurons.

Aim:To investigate the effect of the antisense otigonucleotides（ASODN）spe-cific for human telomerase RNA（hTR）on radio sensitization and proliferationinhibition in human neurogliocytoma cells（U251）.Methods:U251 cells weretransfected with hTR ASODN or nonspecific oligonucleotides（NSODN）.Beforeand after irradiation of60Co-γray,telomerase activity was assayed by telomericrepeat amplification protocol（TRAP-PCR-ELISA）,and DNA damage and repairwere examined by the comet *** classical colony assay was used to plotthe cell-survival curve,to detect the ***:hTR antisense oligo-nucleotides could downregulate the telomerase activity,increase radiation in-duced DNA damage and reduce the subsequent ***,it could in-hibit the proliferation and decrease the D0value which demonstrates ***,telomere length was unchanged over a short period ***:These findings suggest that an ASODN-based strategy may beused to develop telomerase inhibitors,which can efficiently sensitize radiotherapy.
