Cloning, sequencing and identification of single nu-cleotide polymorphisms of partial sequence on the porcine CACNA1S gene

CACNA1S gene encodes the α1 subunit of the calcium channel. The mutation of CACNA1S gene can cause hypokalemic periodic paralysis (HypoKPP) and maliglant hyperthermia synarome (MHS) in hu-man beings. Current research on CACNA1S was mainly in human being and model animal, but rarely in livestock and poultry. In this study, Yorkshire pigs (23), Pietrain pigs (30), Jinhua pigs (115) and the second generation (126) of crossbred of Jinhua and Pietrain were used. Primers were designed ac-cording to the sequence of human CACNA1S gene and PCR was carried out using pig genome DNA. PCR products were sequenced and compared with that of human, and then single nucleotide poly-morphisms (SNPs) were investigated by PCR-SSCP, while PCR-RFLP tests were performed to validate the mutations. Results indicated: (1) the 5211 bp DNA fragments of porcine CACNA1S gene were ac-quired (GenBank accession number: DQ767693 ) and the identity of the exon region was 82.6% be-tween human and pig; (2) fifty-seven mutations were found within the cloned sequences, among which 24 were in exon region; (3) the results of PCR-RFLP were in accordance with that of PCR-SSCP. Ac-cording to the EST of porcine CACNA1S gene published in GenBank (Bx914582, Bx666997), 8 of the 11 SNPs identified in the present study were consistent with the base difference between two EST frag-ments.

Tsallis model-based separation of overlapped peak signals

A simple scheme for separating overlapped peak signals (OPS) is proposed, with the Tsallis distribution as the model for OPS processing and fractional-order differentiation as tool for the signal analysis, the relationships of the maxima and zero-crossing values of the Tsallis peak signal, separately, to the order of differentiation are established, leading to two types of parameter estimators, which are utilized, by adjusting the peak-shape parameter, to calculate the parameters of position, height and width of the Gaussian, Lorentzian and Tsallis distribution. The obtained characteristic parameters can be employed to fit the peak signals to be treated, thereby fulfilling the OPS decomposition.

By introducing the distribution probability of structural units in austenite contain- ing alloying elements and considering its effects on phase transformation, this paper establishes a calculation model of distribution probability of structural units. A new valence electron structure (VES) parameter-transformation effect coefficient of alloying elements (HL), is defined and then studied both theoretically and ex- perimentally. The relationship between the parameter HL and the multiplying factor (the quenching capability factor) of alloying elements is studied. The results indi- cate that the two parameters (HL and the quenching capability factor) have the same variation characteristic and substance feature. Therefore, the multiplying factor virtually expresses the relative quantity of structural units in the alloying elements-containing austenite.

Geochemistry of Trace and Rare Earth Elements in Red Soils from the Dongting Lake Area and Its Environmental Significance

Trace element(TE) and rare earth element(REE) contents in red soils from the Dongting Lake area of china were determined to understanding the provenance and weathering characteristics of the red *** results showed similar REE distribution patterns among red soils from the Dongting Lake area,Xiashu loess from Zhenjiang,loess and Pliocene red soil from the Loess *** patterns implied a similar provenance from dust-storms,except for red soil R5 which formed by bedrock weathering and had significant light REE(LREE) enrichment and heavy REE(HREE) depletion due to longer weathering periods and higher intensity of *** element,especially the Rb/Sr and Li/Ba ratios,and REE,especially the LREE/HREE ratio and δEu could trace weathering *** Rb/Sr,Li/Ba,and LREE/HREE ratios and negative Eu anomalies were present in the red soils from the Dongting Lake *** weathering intensity was in the decreasing order of R5 in the Dongting Lake area > red soils from the Dongting Lake area(including reticulate red soil,weak reticulate red soil,and homogeneous red soil formed by dust storms) > Xiashu loess from Zhenjiang > loess-paleosol and Pliocene red soil from the Loess *** in the TE and REE contents of soil could be effectively used to study the provenance and the weathering intensity.

The problem of fault estimation for a class of non-uniformly sampled-data systems is investigated from the time delay point of view in this ***,the output delay approach is employed to model the sampled-data system as a continuous-time one with time-varying delay ***,based on the analysis of the inapplicability of the adaptive fault diagnosis observer in such class of time-delay systems,a novel augmented fault estimation observer design method is proposed to guarantee the exponential convergence of the estimation ***,an extension to the case of time varying fault estimation for the noisy sampled-data systems is ***,simulation results of a flight control system are presented to demonstrate the effectiveness of the proposed method.

The role of aerial algae in the formation of the landscape of the Yunnan Stone Forest,Yunnan Province, china

Aerial algae on the surface of carbonate rocks at the Stone Forest, Shilin County, Yunnan Province, china, and their bioerosion were investigated in the field and studied in the laboratory in detail. Through the observation, identification and statistics of more than one hun- dred algal samples and rock samples with the optical microscopes (stereomicroscope, biological microscope) and the scanning electronic microscope (SEM), the relationships between erosional forms on the surface of the Stone Forest and algae and/or algal communities and the genetic mechanism for the formation of erosional forms were analyzed. It is suggested that aerial algae play an active role in bioerosive processes that may affect the formation of karst erosional forms. These effects include both direct and indirect ones. The direct effect is the initiative control (algal shape-controlling role) of algae on the formation of karst forms of various scales, mostly mi- cro-scale (3m) and minor-scale (10-3-10-1m) erosional forms. The algal shape-controlling roles can be divided into the algal individual shape-controlling role and the algal community shape-controlling role. The former mostly controls the formation of micro-scale erosional forms, while the latter mostly controls the formation of micro-scale and smaller minor-scale erosional forms. The indirect effect refers to the promoting role of algae in the formation of karst forms, which may affect the formation of karst forms of all types and scales. The bioerosion of algae accelerates the weathering process of the whole Stone Forest karst landforms.

Distributed GEP query optimization on grid service

In order to better solve the problem of distributed query optimization, a query optimization algorithm on gene expression programming （GEP）（QO-GEP） is presented. On the basis of QO-GEP, distributed GEP query optimization on grid service （DGEPQO-GS） is proposed which combines grid service. Simulated experiments show that with the increment of the number of query relation, query time which QO-GEP carries out query decreases apparently. Meanwhile, with the increase of the number of grid nodes, the average querying success rate of DGEPQO-GS increases significantly.

Inducement of chromosome translocation with small alien segments by irradiating mature female gametes of the whole arm translocation line

Haynaldia villosa Schur. (syn. Dasypyrum villosum Candargy, 2n=14, VV) has been proved to be an important genetic resource for wheat improvement. The development of translocation with small alien chromosome segments, especially interstitial translocation, will be helpful for better utilization of its useful genes. Up to now, most of the reported Triticum aestivum – H. villosa translocation lines are involved in a whole arm or large alien fragments. In this paper, we report a highly efficient approach for the creation of small chromosome segment translocation lines. Before flowering, the female gametes of wheat-H. villosa 6VS/6al translocation line were irradiated by 60CO-γ ray at 160 Rad/M dosage rate and three dosages (1600, 1920, 2240 Rad). Anthers were removed from the irradiated florets on the same day and the florets were pollinated with normal fresh pollens of T. aestivum cv. Chinese Spring after 2-3 days. Genomic in situ hybridization (GISH) at mitosis metaphase of root-tip cell of M1 plants was used to detect the chromosome structural changes involving 6VS of H. villosa. Among the 534 M1 plants screened, 97 plants contained small segment chromosome structural changes of 6VS, including 80 interstitial translocation chromosomes, 57 terminal translocation chromosomes and 55 deletion chromosomes. For the 2240 Rad dosage treatment, the inducement frequencies of interstitial translo-cation, terminal translocation and deletion were 21.02%, 14.01%, and 14.65%, respectively, which were much higher than those previously reported. The M2 seeds were obtained by backcrossing of 74 M1 plants involving 146 chromosomes structural changes of 6VS, and it was found that the structural aberrations in the M1 plants could be transmitted to their progenies. Irradiating mature female gametes of whole arm translocation is a new and highly efficient approach for creation of small segment chromosome struc-tural changes, especially for interstitial translocations.
