Anti-nociceptive role of neuropeptide Y in the nucleus accumbens in rats with inflammation, an effect modulated by mu- and kappa-opioid receptors

recent study in our laboratory showed that neuropeptide Y (NpY) plays an antinociceptive role in the nucleus accum-bens (NAc) in intact rats. The present study was performed to further investigate the effect of NpY in nociceptive modulation in the NAc of rats with inflammation, and the possible interaction between NpY and the opioid systems. Experimental inflammation was induced by subcutaneous injection of carrageenan into the left hindpaw of rats. Intra-NAc administration of NpY induced a dose-dependent increase of hindpaw withdrawal latencies (HWLs) to thermal and mechanical stimulations in rats with inflammation. The anti-nociceptive effect of NpY was significantly blocked by subsequent intra-NAc injection of the Y1 receptor antagonist NpY28-36, suggesting an involvement of Y1 receptor in the NpY-induced anti-nociception. Furthermore, intra-NAc administration of the opioid antagonist naloxone significantly antagonized the increased HWLs induced by preceding intra-NAc injection of NpY, suggesting an involvement of the endogenous opioid system in the NpY-induced anti-nociception in the NAc during inflammation. Moreover, the NpY-induced anti-nociception was attenuated by following intra-NAc injection of the μ-opioid antagonist β-funaltrexamine (β-FNA), and κ-opioid antagonist nor-binaltorphimine (nor-BNI), but not by S-opioid antagonist naltrindole, indicating that μ- and K-opioid receptors, not S-opioid receptor, are involved in the NpY-induced anti-nociception in the NAc in rats with inflammation.

Synthesis and Efficiency of a SphericalMacroporous Epoxy-dicyandiamide Chelateresin for preconcentrating and SeparatingTrace Au, Hg, pd and ru from Samples

A novel spherical macroporous epoxy-dicyandiamide chelate resin was synthesized simply and rapidly from epoxy resin and used for the preconcentration and separation of trace amounts of Au (Ⅲ ), Hg (Ⅱ ), pd (Ⅳ) and ru (Ⅲ) ions from solution samples. The analyzed ions can be quantitatively concentrated by the resin at a flow rate of 2. 0 mL/min at pH 4, and can also be desorbed with 15 mL of 4 mol/L HCl + 0. 3 g thiourea from the resin column with recoveries of 96. 5%-99. 0%. After the chelate resin was reused for 7 times, the recoveries of these ions were still over 92%, and 400-1 000 times of excess of Fe(Ⅲ ), Al(Ⅲ ), Ni( II), Mn( Ⅱ ), Cr (Ⅲ ), Cu ( Ⅱ ), Cd (Ⅱ ) and pb (Ⅱ ) caused little interference with the determination of these ions by an inductively coupled plasma optical emission spectrometer (ICp-OES ). The capacities of the resin for the analytes are in the range of 0. 35~0. 92 mmol/g. The rSDs of the proposed method are in the range of 1. 1 % ~4. 0% for each kind of the analyzed ions. The recoveries of a standard added in real solution samples are between 96. 5% and 98. 5%, and the results for the analyzed ions in a powder sample are in good agreement with their reported values.

Nitrogen-Fixing Inhibition by KCl in A Blue-Green Alga Anabaena cylindrica

Growing in the medium containing 0.10 mol/L KCl for three days, the nitrogen-fixing activity ofAnabaena cylindrica decreased by 95%. Extent of the inhibition was enchanced with increased KCl concentrations. The biggest decrease of nitrogen-fixing activity was between 0.02–0.05 mol/L KCl. The same decline on growth was between 0.05–0.10 mol/L KCl. The longer treating-time was, the lower nitrogen-fixing activity was. A short time of KCl-treatment (for 4 h) resulted in 66.4%decrease of the activity. Under effecting of KCl for three days, the frequency of heterocyst differentiation decreased from 7.2%to 2.1%.

ranalisma rostratum (Alismataceae) is an endangered aquatic plant, and now there is only one population existing in china. In its natural habitat, Huli marsh in Chaling, Hunan province, the best growing plants have been observed at microsites where the water depth is 5～10cm, and the plants grow in emergent form.r. rostratum reproduces asexually or sexually. The recruitment of asexual propagules into population is a very important way to maintain the natural population size. Although seed production does not appear to limitr. rostratum, restricted dispersal of propagules and seeds, low seed viability, low germinability, slow growth of seedlings under all conditions, and habitat loss due to agriculture appear to be more important reasons for the endangerment of this plant.

Self-assembly of peptides into fibrils and other morphologies has attracted much attention inmany fields,especially in nanofabrication,pathology and *** this paper,self-assembly of GAV-9peptide in organic solvents,ethanol and acetone,was investigated using atomic force microscopy(AFM)and attenuated total reflectance-Fourier transform infrared spectroscopy(ATr-FTIr).The results indicatedthat GAV-9 self-assembled into various nanostructures in both solvents after deposited and evaporated *** with β-sheet conformation were observed in both solvents when the peptide concentrationwas higher than 280 μ***,ordered fibrils with β-sheet conformation were formed in ethanol,butnot in acetone,with a peptide concentration ranging from 7 μM to 28 μ*** attribute the formation ofvarious nanostructures to the different physicochemical properties of the polar organic solvents on the self-assembly of GAV-9 peptide.

peplomycin induces G1-phase specific apoptosis in liver carcinoma cell line Bel-7402 involving G2-phase arrest

AIM: To investigate the mechanism of peplomycin (pEp)-induced apoptosis in liver carcinoma cell line (Bel-7402). METHODS: Growth inhibition by pEp was analyzed using 3- 4,5-Dimethylthiazol-2-yl-2,5-diphenyltetrazolium bromide (MTT) assay. Apoptotic cells were detected using Hoechest 33258 staining, and confirmed by flow cytometric analysis and DNA fragmentation analysis. The expression of cyclin A and B1 were determined by flow cytometry and Western blot. Annexin V assay was measured by flow cytometric analysis. rESULTS: pEp induced apoptosis and then inhibited cell proliferation in liver carcinoma cell line Bel-7402. Cells treated with pEp 50 μmol/Lfor 15 h were arrested in G2-phase with dramatical expression of cyclin A and a little change in cyclin B1. Almost all the apoptosis occurred in cells undergoing the G1-phase after treatment for 24 h. CONCLUSION: peplomycin induced G1-phase specific apoptosis in Bel-7402 involving G2-phase arrest.

The clonal diversity of Clintonia udensis *** *** detected by ISSr markers among 16 populations,and its correlation with ecological factors was analyzed as well in this *** showed that individuals(clonal ramets)per genotype were 1.12 and 1.149 at population and species levels,respectively,and that the 16 populations were all *** detected genotypes were localized,without exception,within populations but demonstrated relatively high clonal differentiation among *** clonal diversity of the studied populations was high,with the average Simpson's index of 0.975,while the genets showed a clonal architecture of"guerilla".The population genetic diversities revealed by genet were consistent with those by ramet,further confirming their genetic differentiation among *** its genotype diversity within populations probably resulted largely from the frequent seedling regeneration and *** addition,the correlation analysis further revealed that,among the ecological factors,Simpson's index of *** had a significant positive correlation(ppH values in the soil but not others.

IrC analysis of methanol carbonylation reaction catalyzed by rhodium complex

In the reaction cycle for methanol carbonylation catalyzed by rh complex, the structure geometries of the reactant, intermediates, transition states and product of each elemental reaction have been studied by using the energy gradient method at HF/LANL2DZ level, and the changes of their potential profiles have also been calculated. Through IrC analyses of the transition states for each elemental reaction, it is confirmed that the various structure geometries obtained are stationary points on the cycle reaction pathway of methanol carbonylation catalyzed by rh complex, and the changes are given in energies and structure geometries of the reactant molecules along the reaction pathway of lowest energy. It has been proposed that the geometrical conversions of intermediates play an important role during the cycle reaction. Through analyses of structure geometries, it has been suggested that, in addition to cis- and transstructure exchange linkage of catalysis reactive species, the two pathways, cis- and frans-catalyzed cycle reactions, can also be linked through geometrical conversion of intermediates, of which the activation energy is 49.79 kJ/mol. Moreover, the reductive elimination elemental reaction may be neither c/s-cycle nor trans- one, showing that the cycle reaction can be achieved through various pathways. However different the pathway, the oxidative addition elemental reaction of CH3I is the rate-controlling step.
