Aim:To simultaneously investigate the contribution of the alpha 1 chain of col-lagen type 1(COLlAl)and alpha2-HS-glycoprotein(AHSG)genes to the varia-tion of bone geometric parameters in both Caucasians and ***:Sixhundred and five Caucasian individuals from 157 nuclear families and 1228 Chi-nese subjects from 400 nuclear families were genotyped at the AHSG-Sacl,COLIA1-PCOL2 and Spl polymorphisms using polymerase chain reaction(PCR)-restric-tion fragment length polymorphism(RFLP).5 FN bone geometric parameters werecalculated based on bone mineral density and bone area of femoral neck(FN)measured by dual energy X-ray *** stratification,totalfamily association,within-family association,and linkage tests were performed bythe quantitative transmission disequilibrium test ***:The t-testshowed the significant differences of all bone geometric phenotypes(except ED)between Caucasians and Chinese in the offspring using both unadjusted andadjusted(by age,height,weight,and gender)*** Caucasians,we foundsignificant within-family association results between the COLlAl-Spl polymor-phism(rs1800012)and cross sectional area(CSA),cortical thickness(CT),endocortical diameter(ED),buckling ratio(BR)(P=0.018,0.002,0.023,and 0.001,respectively);the COLlAl-Spl polymorphism also detected significant linkagewith BR(P=0.039).In the population of China,the within-family associationsbetween the COLlAl-PCOL2 polymorphism(rs1107946)and CT,BR were signifi-cant(P=0.012 and 0.008,respectively).Furthermore,evidence of linkage wereobserved between the AHSG-Sacl polymorphism(rs4918)and CT,BR(P=0.042and 0.014,respectively)in Caucasians,but not in ***:Ourresults suggest that the COLlAl gene may have significantly association withbone geometry in both Caucasians and Chinese,and the AHSG gene may belinked to bone geometry in Caucasians,but not in *** study representsour first efforts on investigating the importance of the COLlAl and A

Sox2, one of genes which express in embryonic stem (ES) cells, plays an important role in self renewal of ES cells, and is used to make induced pluripotent stem (iPS) cells. In this study, we subcloned mouse (Mus muscules) full-length Sox2 cDNA and inserted Sox2 into pET-41a expression vector. Then the recombinant vector was transformed into Escherichia coli BL21 (DE3) cells to express Sox2 fusion protein. The expressed products were identified by Western blot using anti-GST antibody. The result indicated that the size of Sox2 fusion protein was in 61 kD. And the protein was recovered from the SDS-PAGE. And we used the sox2 fusion protein to prepare polyclonal antibody. The Dot blotting result showed that the anti-Sox2 antiserum had 1∶12,800 titers. The antibody will be useful for studying the function of Sox2 and its role in ES cell self-renewal.

Tumor necrosis factor (TNF) is a proinflammatory cytokine that plays a critical role in diverse cellular events, including cell proliferation, differentiation and apoptosis. TNF is also involved in many types of diseases. In recent years, the molecular mechanisms of TNF functions have been intensively investigated. Studies from many laboratories have demonstrated that the TNF-mediated diverse biological responses are achieved through activating multiple signal- ing pathways. Especially the activation of transcription factors NF-κB and AP-1 plays a critical role in mediating these cellular responses. Several proteins, including FADD, the death domain kinase RIP and the TNF receptor associated factor TRAF2 have been identified as the key effectors of TNF signaling. Recently, we found that the effector mol- ecules of TNF signaling, such as RIP and TRAF2, are also involved in other cellular responses. These finding suggests that RIP and TRAF2 serve a broader role than as just an effector of TNF signaling.

It is well documented that the glycosylation of E-cadherin is correlated with cancer metastasis, but whether E- cadherin could be core fucosylated remains largely unknown. We found that E-cadherin was core fucosylated in highly metastatic lung cancer cells while absent in lowly metastatic lung cancer cells. Since α-1,6 Fucosyltransferase (α-1,6 FucT) is known to catalyze the reaction of core fucosylation, we investigated the biological function of core fucosylation on E-cadherin by α-1,6 FucT targeted RNAi and transfecting α-1,6 FucT expression vector. As a result, calcium dependent cell-cell adhesion mediated by E-cadherin was strengthened with the reduction of core fucosylation on E- cadherin after RNAi and was weakened with the elevated core fucosylation on E-cadherin after α-1,6 FucT over expression. Our data indicated that α-1,6 FucT could regulate E-cadherin mediated cell adhesion and thus play an important role in cancer development and progression. Computer modeling showed that core fucosylation on E-cadherin could significantly impair three-dimensional conformation of N-glycan on E-cadherin and produce conformational asym- metry so as to suppress the function of E-cadherin. Furthermore, the relationship between the expression of core fucosylated E-cadherin and clinicopathological background of lung cancer patients was explored in lung cancer tissue of patients. It turns out to demonstrate that core fucosylated E-cadherin could serve as a promising prognostic indicator for lung cancer patients.

BACKGROUND: Hepatocellular carcinoma (HCC) is one of the most common malignant tumours worldwide. Te-lomerase is reactivated in various types of malignant tumors and may contribute to the development of HCC. To evaluate the role of telomerase in formation and development of HCC, we analyzed its expression status in different parts of HCC tissues and peripheral blood mono-nuclear cells (PBMCs) , and explored its clinical implications for diagnosis of HCC. METHODS: Total RNAs and telomerase were extracted from HCC and their non-cancerous tissues, and both relationships were analyzed between them. The expression of telomerase reverse transcriptase (hTERT) mRNA and telomerase activities in liver tissues and PBMCs were detected by RT-PCR and telomeric repeat amplification protocol (TRAP)-ELISA, respectively. The diagnostic values of telomerase in PBMCs were investigated in the diagnosis and differentiation of HCC. RESULTS: The specific activities of telomerase were 18.25 ±15.02 A/μg RNA in HCC tissues, and significantly higher than those in their non-cancerous tissues (8.16 ± 6.22 A/μg RNA, P 34 μg/mg wet liver and lower than 53.77 ± 52.02 μg/mg wet liver in their non-cancerous parts (P < 0.01). The different telomerase levels could be detected in PBMCs from patients with chronic liver diseases and control group. The enzyme activities were significantly higher in HCC than in any other groups (P <0.01). However, circulating telomerase levels were more obviously decreased in HCC patients after transcatheter arterial embolization treatment than before the treatment (P <0.01). The analysis of combined serum alpha-fetoprotein level and PBMCs telomerase was more sensitive and specific for the diagnosis of HCC. CONCLUSION: The abnormal expression of telomerase in HCC tissues and circulating PBMCs could be a useful marker to the diagnosis and prognosis of HCC.

In order to establish the novel high throughput, high efficiency and low cost technological platform for the research of N-glycoproteomics, to resolve the significance of characteristic expression profile of glycoprotein and to find the proteins with biological functional importance, the glycoproteins with high-mannose core and the two antennary types were purified and enriched by the Con A affinity chromatography. Con A affinity protein expression profiles of normal human liver tissue were gener- ated by using SDS-PAGE, two-dimensional electrophoresis (2-DE) followed by fast fluorescence stain- ing based on multiplexed proteomics (MP) technology. 301 visible protein spots on the gel were de- tected and 85 of glycoproteins were further successfully identified via peptide mass fingerprinting (PMF) by a matrix assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF- MS/MS) and annotated to IPI databases. Identified glycoproteins definitely take part in the regulation of cell cycle and metabolic processes. The glycosylation sites were predicted with NetNGlyc 1.0 and NetOGlyc 3.1 software, meanwhile they were classified according to the geneontology methods. The construction of Con A affinity glycoprotein database of normal human liver tissue would contribute to the subsequent research.

The diploid Oryza species with C-genome type possesses abundant genes useful for rice improvement and provides parental donors of many tetraploid species with the C-genome(BBCC,CCDD).Despite extensive studies,the phylogenetic relationship among the C-genome species and the taxonomic status of some taxa remain *** this study,we reconstructed the phylogeny of three diploid species with C-genome(Oryza officinalis,***,and ***) based on sequences of 68 nuclear single-copy *** obtained a fully resolved phylogenetic tree,clearly indicating the sister relationship of *** and ***,with *** being the more divergent *** phylogenies of the C-genome species found in previous studies might result from lineage sorting,introgression/hybridization and limited number of genetic markers *** further applied a recently developed Bayesian species delimitation method to investigate the species status of the Sri Lankan and African *** of two datasets(68 genes with a single sample,and 10 genes with multiple samples) support the distinct species status of the Sri Lankan and African *** addition,we evaluated the impact of the number of sampled individuals and loci on species *** simulation suggests that sampling multiple individuals is critically important for species delimitation,particularly for closely related species.
