Breast cancer is a common malignant tumor,threatening the women's health *** of malignant tumor is the main cause of tumor related death,which is a complicated biologic *** activation of estrogen receptor alpha(ERα) expression is an early event in breast cancer *** evidence supports the existence of a plasma membrane *** proteins constitute important structural components of caveolae,participate in signal transduction,and Caveolin-1 could inhibit the anchorage-independent cell *** study the relationship between Caveolin-1 and ER in breast cancer,we transcripted the siRNA into normal epithelial cells(MCF10A) to silence the expression of *** we examined the expression of Caveolin-1,ERαand associated proteins by Western *** results indicate that Caveolin-1 was silenced by 85%at 96h,whereas we found that the expression of protein ERα66 and ERα36 was up-regulated. With the Caveolin-1 down-regulation, the phosphorylation of ERK1/2 was increased. These data shows that the down-reg- ulation of Caveolin-1 could activate MAP kinase, including the phosphorylation of ERK1/ 2,which played critical roles in the control of cell proliferation,differentiation,homeostasis, and survival via the expression of ERαwhich is residing in the ***,monolayer culture of MCF10A,MCF10A-ST1, MCF10A-7SD8 and MCF-7 were scrape wounded and allowed to grow for 24h,the distance traveled was measured as the distance between the wound edge and mirgrating *** we also obtained the evidence that migrant distance were difference in the four cell lines, 50%in MCF10A-7SD8 and 30%in MCF10A-ST1 comparing to MCF-7,little in MCF10A by wounded *** results indicate that Caveolin-1 could significantly mediate membrane-initiated estrogen signaling pathway, it played an important role in mammary tumorigensis and may effect on cell metastasis in *** evidences will greatly advance the progress in prevention

Breast cancer is one of the most common cancers among women in the *** early detection is continuing to improve the survival rate,the prognosis is poor after the tumor has ***,it is urgent to identify the genes that drive breast cancer metastasis and develop new ***-1,a 21-24 kD membrane protein,is an essential structural molecule of Caveolae,participating in Caveolae formation,cell trafficking, cholesterol homeostasis,signal transduction,and Caveolin-1 could inhibit the anchorage-independent cell growth. The matrix metalloproteinases (MMPs) are a family of zinc-dependent endopeptidases that play a key role in both normal and pathological processes involving tissue remodeling events. The expression of these proteolytic enzymes is highly regulated by a balance between extracellular matrix (ECM) deposition and its degradation, and is controlled by growth factors, cytokines, hormones, as well as interactions with the ECM macromolecules. Furthermore, the activity of the MMPs is regulated by their natural endogenous inhibitors, which are members of the tissue inhibitor of metalloproteinases (TIMP) family. In the normal mammary gland, MMPs are expressed during ductal development, lobulo-alveolar development in pregnancy and involution after lactation. Under pathological conditions, such as tumorigenesis, the dysregulated expression of MMPs play a role in tumor initiation, progression and malignant conversion as well as facilitating invasion and motastasis of malignant cells through degradation of the ECM and basement membranes. ＜正＞ Since 1977, numerous studies have indicated the existence of a putative membrane-based estrogen receptor. However, the identity of this membrane-based estrogen receptor has not been established. Through an extensive homology search in Gene Bank, we recently identified and cloned a novel variant of ER-αwith a predicted molecular weight of 35.7 kDa. INTRODUCTIONClinical factors contributing to the therapeutic challenge of hepatocellular carcinoma (HCC) are manifold :tumors arise often in patients with compromised liver function ,therefore limiting therapeutic options ;symptoms develop only at later stages of tumor progression ,and tumors tend to invade normal stuctures or occur in multiple locations simultaneously.

NIH 3T3 cells, a mouse fibroblast cell line used as routine target cells for transfection experiments, undergo spontaneous transformation in our experiments after they form a confluent sheet in medium containing fetal bovine serum (FBS) or lower coneentration of calf serum (CS). The transformation takes the form of foci of multiplying cells among the surrounding cells which have stopped cell division. However, no focus of transformed cells could be seen in medium containing high concentration (10％) of CS. Further experiments indicated that the frequency of transformation is highly dependent on the concentration of serum and the transformation in CS is changeable when the cells are passaged in FBS. 3~H-thymidine autoradiography has been proved to be a sensitive measurement indicator for focus formation. Our results suggest that the high frequency of transformation and its dependence on confluency as well as on medium composition are characteristics of cell differentiation rather than mutation. The role of the NIH 3T3 cell line as a cancer-initiated cell population and its accelerated transformation by ras oncogene might be considered as a form of tumor promotion is discussed.

Here,we report that the expression of ERα-36 and contribution of caspase-3 mediated by ERα-36 to the MPP-induced apoptosis in PC12 cells by using Western ***,we transfected ERα-36 shRNA plasmid and established the ERα-36-downregulated PC12 cells model(PC12-36L1).And then,flow cytometry and Heochst33342 staining methods were used for apoptosis ***,we examined caspase-3 signal proteins related to cell *** were pretreated with estrogen(E2,1 h) followed by treatment with MPP(48 h).The results showed that(1) ERα-36 was expressed in PC12 cells,(2) Both PC12 cell and PC12-36L1 cell showed apoptosis after treatment with MPP(48 h).The expression of caspase-3 in PC12-L1 cell was lower than that of PC12 cells (P<0.05).Compared with control group,pretreatment with E2 can decrease the expression of caspase-3 in PC12- L1 and PC12 cells,and in PC12 cell the expression of caspase-3 was higher than that
