BACKGROUND: Previous studies reported that frontal-temporal-parietal-occipital pathological changes and diseased range in the right cerebral hemisphere were closely correlated with neglect. But studies on the correlation of neglect with diseased region and area in patients who suffer from initial attack of single focus of cerebral infarction (CI) in left and right cerebral hemispheres are few. OBJECTIVE: To observe the status of neglect in patients who suffer from single focus of CI in cerebral hemisphere, and analyze the correlation of neglect with diseased region and area of CI. DESIGN: Case analysis. SETTING: Treatment Center for Cardiocerebrovascular Disease, Second hospital of Xiamen city; department of neurology, First hospital affiliated to Baotou medical college. PARTICIPANTS: All the CI patients hospitalized in the department of neurology, First hospital affiliated to Baotou medical college from June 1998 to May 2001 were retrieved. Inclusive criteria: ① Patients who suffered from initial attack of CI, which was confirmed by skull CT or MRI within 24 hours after onset and presented single focus in cerebral hemisphere. ② be conscious and could cooperate in the examination. ③ did not receive formal education, but could do accounts and some simple writing and reading. ④Patients with homonymous hemianopia were excluded through the examination of perimeter. ⑤ Informed consents were obtained from all the patients. Among 67 patients who met the inclusive criteria, 33 suffered from CI in the left cerebral hemisphere and 34 in the right cerebral hemisphere. METHODS: ① Patients received neglect supplement examination and Chinese aphasia examination within 2.5 to 3 months after the attack of CI . AIM: To investigate the effect of tanshinone (Tan) on the neuropathological changes induced by amyloid β- peptide1-40(Aβ1-40) injection in hippocampus in rats. METHODS: Aβ1-4010 μg was injected bilaterally into the dorsal blade of the dentate gyrus in the hippocampus. The level of acetylcholinesterase (AChE) in hippocampus was evaluated by histochemistry. The expressions of neuronal nitric oxide synthase (nNOS) and inducible form of NOS (iNOS) were detected by immunohistochemistry and Western blot. Aβ1-40-injected rats were treated ig with Tan, the major active ingredient from Salvia miltiorrhiza of Chinese herb extract. RESULTS: The level of AChE positive fibers of each subfield in Aβ1-40-injected hippocampus decreased significantly compared with those of control (P2 toCA3 (CA2-3),and dentategyrus (DG)subfields (P<0.01). CONCLUSION: Tan can protect the neuropathological changes induced by Aβ1-40 injection in hippocampus.

Synthesis of cystine C_(60) derivative and its protective effects on hydrogen peroxide-induced apoptosis in PC12 cells

Oxidative stress has been considered as a major cause of cellular injuries in a variety of clinical abnormalities, especially prominent in neural diseases. One of the usable ways to prevent the reactive oxygen species (ROS)-mediated cellular injury is dietary or pharmaceutical augmentation of some free radical scavenger. Water-soluble amino-fullerene is a novel compound that behaves as a free radical scavenger with excellent biology consistent. In the present study, we have synthesized and characterized a novel cystine C60 derivative for the first time, and investigated the effects on hydrogen peroxide-induced oxidative stress and apoptotic death in cultured rat pheochromocytoma (PC12) cells. PC12 cells treated with hydrogen peroxide underwent apoptotic death as determined by MTT, PI/Hoechst 33342 staining and flow cytometry analysis. These results suggested that cystine C60 derivative has the potential to prevent oxidative stress-induced cell death and has no evident toxicity.

NMDA and AMPA receptors mediate intracellular calcium increase in rat cortical astrocytes

AIM: To study the effect of glutamate on the intracellular calcium signal of pure cultured rat astrocytes and the roleof N-methyl-D-aspartate (NMDA) and alpha-amino-3-hydroxy-5-methyl-4-isoxazole-propionic acid (AMPA) re-ceptors in the procedure. METHODS: The fluorescence of calcium was measured by Fura-2/AM (F345/F380).RESULTS: L-Glutamate induced [Ca2+]i increase in most of the cells in concentration- and time-dependent *** 50 mmol/L induced the fluorescence increase by almost three to four times, while the effect of AMPA 50mmol/L was just half of that of D-(-)-2-amino-5-phosphonopentanoic acid (D-AP-5; a selective antagonist of theNMDA receptor). 6-Cyano-7-nitroquinoxaline-2,3-dione (CNQX, a selective antagonist of the AMPA receptor)abolished the effects of NMDA and AMPA, respectively. D-AP-5 and CNQX simultaneously or respectively attenu-ated the effect of L-glutamate at different degrees, but could not abolish it entirely. CONCLUSION: Glutamatemodulated intracellular Ca2+ of pure cultured rat astrocytes through different pathways. The activation of NMDAand AMPA receptors took part in the complex mechanisms.

BACKGROUND: Prostaglandin E2 (PGE2) can directly regulate toxic injury of hippocampal neurons through participation by its receptor. Increase of excitability of hippocampal membrane and long-term synaptic elasticity are closely related to PGE2, PGD2 and PGF2A. This suggests that PGE2 may be a key molecule of neuronal signal passage and regulate the existence of neurons through its receptor. However, which isoforms of PGE2 receptor expressing in hippocampal neurons is still unclear. OBJECTIVE: To research the subtype expression of PGE2 receptor in hippocampus of rats through mRNA transcription and protein interpretation. DESIGN: Animal studies with random, control and operator and designer double-blind methods. SETTING: University of South Carolina, Animal Center. MATERIALS: Sprague-Dawley rats, aged 12 weeks, weighing 200 g, females 48 and males 48, were selected from Animal Center in South Carolina University. Tri ReagentTM kit was provided by Molecular Research Center, USA. METHODS: The experiment was carried out in Animal Center in South Carolina University from January to December 2005. The expression of the PGE2 receptors was profiled and compared in rat hippocampus using real-time RT-PCR and Western-blot techniques. MAIN OUTCOME MEASURES: Expression of PGE2 receptors in various isoforms of hippocampal neurons of rats. RESULTS: mRNAs of all four EP1-4 subtypes were detected in the hippocampus. Western-blot data showed consistently detectable bands at approximately Mr 50 000 of EP1, Mr 40 000 and Mr 52 000 of EP2, Mr 45 000, Mr 57 000 and Mr 105 000 of EP3, and Mr 46 000 of EP4. CONCLUSION: Identifying four subtypes of EPs heterogeneously expresses in the hippocampus.

This study aimed to investigate the effects of arsenic trioxide(As2O3) on the mitochondrial DNA(mtDNA) of acute promyelocytic leukemia(APL) *** NB4 cell line was treated with 2.0 μmol/L As2O3 in vitro,and the primary APL cells were treated with 2.0 μmol/L As2O3 in vitro and 0.16 mg kg-1 d-1 As2O3 in *** mitochondrial DNA of all the cells above was amplified by PCR,directly sequenced and analyzed by Sequence Navigatore and Factura *** apoptosis rates were assayed by flow *** DNA mutation in the D-loop region was found in NB4 and APL cells before As2O3 use,but the mutation spots were remarkably increased after As2O3 treatment,which was positively correlated to the rates of cellular apoptosis,the correlation coefficient:rNB4-As2O3=0.973818,and rAPL-As2O3=*** mutation types include transition,transversion,codon insertion or deletion,and the mutation spots in all samples were not constant and *** is revealed that As2O3 aggravates mtDNA mutation in the D-loop region of acute promyelocytic leukemia cells both in vitro and in *** DNA might be one of the targets of As2O3 in APL treatment.

Rapid Inhibition of the Glutamate-induced Increase of Intracellular Free Calcium by Magnesium in Rat Hippocampal Neurons

By using Fura-2/AM, the effects of magnesium (Mg 2+) on the glutamate-induced increase of intracellular free calcium ([Ca 2+]i) in the cultured hippocampal neurons and the features were investigated by integrated photoelectric detecting system. The experiments were designed to three groups (The drug was spit to the cells for 20 s): Group A receiving 1×10 —5 mol/L glutamate; Group B receiving 1×10 —5 mol/L glutamate and1×10 —5 mol/L Mg 2+ simultaneously; Group C receiving 1×10 —5 mol/L glutamate again after [Ca 2+]i in group B back to the baseline. The results showed that in group A, [Ca 2+]i was obviously increased. In group B, the changes in [Ca 2+]i and the peak value were significantly decreased. Moreover, the elevation of Phase 1 was slowed down and Phase 2 was shortened to some extent, and the plateau phase between them was relatively prolonged. In group C, calcium oscillation similar to that in group A occurred, but both the Phase 1 and Phase 2 were shortened and the △[Ca 2+]i was slightly decreased. It was suggested that Mg 2+ could quickly inhibit the rise of [Ca 2+]i induced by glutamate in the cultured hippocampal neurons in rats.

Association between apolipoprotein E promoter-219G/T polymorphism and total cholesterol level in patients with Alzheimer disease

BACKGROUND: Many researches have suggested that apolipoprotein E (APOE) and total cholesterol metabolism are closely related with dementia. In the supposed theory, 219 site of APOE promoter region is near gene coding region, so its polymorphism may result in the abnormality of APOE gene and protein expression, and finally lead to dementia. OBJECTIVE: To observe the association between APOE promoter-219G/T polymorphisms with serum total cholesterol in patients with Alzheimer disease, and compare it with non-dementia people. DESIGN: Case-control, comparative observation. SETTING: department of neurology, Fengtian hospital of Shenyang medical college. PARTICIPANTS: Fifty-five dementia patients including 27 males and 28 females aged (66±3) years and treated in the department of neurology, Fengtian hospital were selected from January 2002 to December 2005 as the Alzheimer disease group. They all diagnosed according to the DSM-Ⅳdiagnostic criteria of Alzheimer disease instituted by American Psychiatry Association in 1994. Meanwhile, 44 none-dementia patients including 21 males and 23 females aged (66±3) years were selected from other clinical departments of Fengtian hospital as control group. All the participants were informed the detection and agreed. METHODS: Genomic DNA was extracted from the peripheral blood of all subjects, then 'NEST'PCR, DNA sequence and enzyme digestion were adopted to detect the expression of APOE promoter-219 polymorphism, following by biomedical statistics analysis based on the clinical total cholesterol level. MAIN OUTCOME MEASURES: Polymorphism of APOE promoter-219 G/T and total cholesterol level. RESULTS: All 55 dementia patients and 44 non-dementia ones were involved in the result analysis. ①Allele and genotype frequency: The T allele frequency of the Alzheimer disease group was significantly higher than that in the control group [88.2% (97/110), 54.5% (48/88)], while G allele frequency was remarkably lower than that in the control group [1
