Regulation of Propylene and 1-Methylcyclopropene on Expressions of ACS and ACO Genes in Persimmon Fruits

The regulation of postharvest treatment with propylene and 1-MCP on ethylene release rate and expressions of 1- aminocyclopropane-l-carboxylic acid synthase （ACS） and 1-aminocyclopropane-1-carboxylic acid oxidase （ACO） genes in Fuping Janshi persimmon （Diospyros kaki L.） fruit were investigated. Fruits were treated with propylene and 1-MCP, then stored at 20℃, ethylene release rate of the treated fruits was measured at regular intervals and RNA was extracted for Northern blotting analysis. The results suggested that treatment with propylene accelerated the expressions of ACS and ACO genes and then enhanced the ethylene biosynthesis, while treatment with 1-MCP inhibited the expressions of two genes and their ethylene biosynthesis. Furthermore, different effects on expressions caused by treatments with propylene and 1-MCP existed in various fruit tissues, the inhibitory effect on ACS and ACO genes by 1-MCP was the strongest in pericarp, followed by pulp and core tissues, in the area near fruit stalk, the inhibitory effect was the weakest. While the enhanced effect on ACS and ACO genes by propylene increased from pulp, core, and pericarp to the area near fruit stalk. Expression of each member of ACS and ACO families in various tissues was also completely different, in control and propylene treatment, DKACS3 gene just expressed in the area near fruit stalk and did not express in other tissues, but DKACS2 gene expressed in four tissues by treatment with propylene.

Difference of a citrus late-ripening mutant (Citrus sinensis) from its parental line in sugar and acid metabolism at the fruit ripening stage

‘Fengjiewancheng’(FW) (Citrus sinensis), a bud sport of‘Fengjie 72-1’navel orange (FJ), ripens one month later than its parental line. Differences in sugar and acid content and the transcript level of su-crose- and citric-metabolic enzymes for the two cultivars were investigated during fruit ripening. Re-sults showed that both sugar and acid metabolisms of the mutant were affected by the mutation. In the pulp of FW, sugar content was significantly lower than that in FJ before 227 DAF (days after flowering) and higher at 263 DAF; the mutant’s gene expression of one isoform of citrus sucrose synthase (CitSS1) was delayed, and its gene expression of citrus acid invertase (CitAI) was stronger than that in its pa-rental cultivars at 207 and 263 DAF. In the peel, only the sucrose content in FW was significantly lower than those in FJ at the early periods of fruit ripening (165 and 187 DAF); however the transcripts of the sucrose-cleaving enzymes in the mutant were higher than those in FJ at different ripening points. As regards acid accumulation in the two cultivars, it was observed that in the pulp of the mutant, the malic acid content was significantly lower than that in its parental cultivars from 187 to 263 DAF, and in the peel, remarkably higher during the whole fruit ripening period. The citric acid content in both the pulp and the peel of FW was higher than that in those of FJ during the early ripening period and lower during the late ripening period, which were correspondingly associated in part with the higher transcript level of citrus mitochondrial citrate synthase (CitCS) and with lower or undetectable transcript level of citrus cytosolic aconitase (CitAC). Hence, it could be concluded that the mutation in FW affected sugar and acid metabolism, which might be related with other late-ripening phenotypes.

Mitochondrial(mt) genome sequences are very useful for investigating population genetic structures,systematics,and phylogenetics of *** the present study,genetic variation among isolates of Spirometra erinaceieuropaei spargana from different geographical locations in hunan province, china,were characterized by three mt genes encoding cytochrome c oxidase subunit 3(cox3),NADH dehydrogenase subunits 1 and 4(nad1 and nad4),*** three mtDNA regions were amplified separately from genomic DNA of individual sparganas by polymerase chain reaction(PCR) and the amplicons were sequenced *** sequences of pcox3,pnad1,and pnad4 were 541,607,and 847 bp in length,*** A+T contents of the sequences were 68 39-68.76%(pcox3),63.76-64.91% (pnad1),and 67.18-67.77%(pnad4),*** variations between *** from different regions were 0.0-1.5%for pcox1,0.0-2.8%for pnad1,and 0.0-2.7%for *** analysis using neighbor joining(NJ),maximum likelihood(ML),and maximum parsimony(MP) methods inferred that spargana isolates in hunan province clustered into two main clades:isolates from Yiyang and Shaoyang clustered together in one clade,with a high bootstrap value(>50%),and those from other origins grouped together in another *** findings demonstrated the usefulness and attributes of the three mtDNA sequences for population genetics studies of ***,the results of the present study also have implications for the diagnosis and control of *** infections in human and animals.

The complete mitochondrial gonome of three Taenia species was amplified in two overlapping fragments and then *** sizes of the entire nat genome are 13700 bp for ***,13489 bp for *** and 13647 bp for ***,*** contains 36 genes(consisting of 12 genes for encoding proteins,2 genes for rRNA and 22 genes for tRNA),but a gene for ATP synthetase subunit 8 is missing,which is consistent with all other species of cestode studied to *** genes are transcribed in the same direction and have a nucleotide composition high in A and T,but low in G and *** contents of A+T of the complete genomes are 71.3%for T. multiceps,70.8%for *** and 73.0%for *** AT bias had a significant effect on both the codon usage pattern and amino acid composition of *** the noncoding region,*** and *** have two noncoding regions,but *** only one the location of the noncoding *** include: a short non-coding region(SNR,73nucleotides) and a long non-coding region(LNR,176nucleotides),*** include:a short non-coding region(SNR,64nucleotides) and a long non-coding region(LNR,124nucleotides),T. taeniaeformis include:non-coding region 414 *** are substantial differences when compared with T. saginata,***,*** and Echinococcus multilocularis and other of non-coding *** analyses based on concatenated amino acid sequences of 3 protein-coding genes(cox1,cox2,nad6),revealed that the newly described species *** was more closely related to the others of Taenia genus, consistent with results of a previous classification based upon their morphology.

Estimation of Cucumber Evapotranspiration in Solar Greenhouse in Northeast china

The solar greenhouse without heating system has been widely used for vegetable production in Northeast china since the 1980s. The experiment was conducted in the solar greenhouse to study the relationship between evapotranspiration （ET） of cucumber and climatic factors. Reference evapotranspiration （ET0） and pan evaporation （Ep） were applied to determine cucumber evapotranspiration by regression methods. Results showed that the ET of cucumber increased with the increasing of solar radiation, air temperature and vapor pressure. The solar radiation was the most important factor that influences ET among environmental parameters. The determination coefficient （R2） was 0.865 between ET and Ep, higher than that between ET and ET0 （application of a constant vegetation coefficient, R2 = 0.46）. The pan coefficient （kp, determined from the ratio of ET to Ep） and the crop coefficients （kc, determined from the ratio of ET to ET0） showed considerable variability and no obvious systematic pattern could be described throughout the study period. Ep was linearly related to ET and 20 cm pan can be well used to determine the water requirement of cucumber under subsurface drip irrigation in this type of solar greenhouse.

Development of EST-SSR primers and their practicability test for Laminaria

Laminaria （Saccharina） is one of the economic important seaweeds. Through sequence analysis of 4 099 ESTs from Laminaria （971 were generated from our own cDNA laboratories and 3 128 were downloaded from updated EST databases） with SSRIT software, two hundred and fifty-four SSRs in 206 ESTs were found. From the 254 SSRs, sixty-three SSR primer-pairs were designed. In order to test their practicability, the 63 primer-pairs were tested in commonly used SSR reaction conditions using 12 Laminara DNA samples as templates. The results show that 23 SSR primer-pairs gave good amplification patterns on most （more than 80%） of the 12 Laminara DNA templates. Genetic diversity study of 12 Laminaria lines, which were widely used in breeding and economic cultivation in china, was performed based on the obtained SSR data.

Sequence variability in two mitochondrial DNA(mtDNA) regions and internal transcribed spacer of rDNA, namely cytochrome c oxidase subunit 1(cox1) and NADH dehydrogenase subunit 4(nad4),among and within tapeworm,***,***,*** and *** from different geographical origins was examined by a mutation scanning approach.A portion of the cox1 gene(pcox1),nad4 genes(pnad4) and ITS were amplified separately from individual tapeworm by polymerase chain reaction(PCR).Representative samples displaying sequence variation were subjected to sequencing in order to define genetic markers for their specific identification and *** the intra-specific sequence variations within each of the four tapeworm species were 0-0.7% for pcox1,0-1.7%for pnad4 and 0.1-3.6%for ITS,the inter-specific sequence differences were significantly higher,being 12.1-17.6%for pcox1,18.7-26.2%for pnad4 and 31-75.5%for ITS,*** analy- ses based on the combined sequences of peox1 and pnad4 revealed that the recently described species *** was more closely related to *** than to *** findings provided mtDNA evidence for the validity of *** and also demonstrated clearly the usefulness and attributes of the mutation scanning-sequencing approach for studying the population genetic structures of these and other tapeworm of socio-economic importance.
