Orthogonal design for optimization of pigment extraction from surface sediments of the Changjiang River Estuary

Using a suitable solvent for extracting pigments from sediment for high performance liquid chromatography（HPLC） analysis is critical for obtaining qualitative and quantitative estimates of phytoplanktonic and benthic algal biomass,as well as community composition. Five methodological factors（sample dehydration,extraction solvent,extraction duration,number of extractions,and ratio of solvent volume：sample weight） were studied using an L 9（3 4 ） orthogonal design in a sedimentary pigment extraction experiment on samples collected from the Changjiang large-river delta-front estuary（LDE）,using HPLC analysis. The results show that the optimal extraction method for sedimentary pigments should include freeze-drying samples prior to extraction. The effects of different factors on sedimentary pigment extraction were separated by the L 9（3 4 ） orthogonal design experiments and showed that the extraction solvent was the most important,with extraction duration the second most important,and numbers of extraction and ratio of solvent volume：sample weight was the least important. The mixed solvent treatment comprised of acetone,methanol and water（80：15：5,by volume） was best for polar pigment extraction,with 100% acetone better for apolar pigments. For most pigments employed in this study（i.e.,peridinin,fucoxanthin,alloxanthin,diatoxanthin,zeaxanthin,pheophytin-a and β-carotene） ,3 h was found to be enough time for extraction from these deltaic sediments. However,for chlorophyll-a,the most important pigment used for estimating algal biomass,12.h was needed. A small amount of solvent（3 ml） with duplicate extractions obtained the greatest amount and diversity of pigments. Unfortunately,no extraction method was found to be suitable for all pigments in sediments. The choice of extraction procedure should be made in accordance with the objective of each study,taking into consideration the properties of sediments and pigments in question.

A new algorithm for retrieving wind vector using dual polarization imagery of advanced synthetic aperture radar (ASAR) is developed and tested. Based on the combination of co-polarization and cross-polarization, this new algorithm effectively eliminates the 180° ambiguity which occurs when using single imagery of the ASAR to retrieve wind vector. This algorithm also solves the problem that the retrieval results will break down on very small spatial scales. The results retrieved from dual polarization imagery of ASAR show that the wind speed and direction are in agreement with the data from Quikscat and buoy measurements. The root mean square errors of wind direction and speed between the retrieved results and data from Quikscat are 2.2.° and 0.53 m/s, respectively. Comparisons between the new method and the traditional method show that the new methods fit the buoy data and Quikscat data a little better than the traditional method in this image.

Hydrothermal convection in the upper oceanic crust has been inferred to be a common and important process. Under the simplified conditions of planar boundaries, permeability provides a strong constraint on the pattern of circulation, the dimensions of convective cells and flow field of hydrothermal circulation. By applying an advanced numerical modeling method, to our knowledge, it is the first time to investigate convection as it is influenced by different strata permeability structures, formational anisotropy, fracture zone and cooling intrusion. The simplified geological model is com-posed of 3 layers, sedimentary layer, high permeable basement layer and low permeable basement layer from top to bottom. When permeability in high permeable layer is 10 times larger than that in sedimentary layer, convection occurs in high permeable layer. The pattern of hydrothermal circulation and flow velocity of hydrothermal fluid are strongly influenced by strata permeability structures, changes of permeability in high permeable basement layer, fracture zone and cooling intrusion. However, formational anisotropy relatively exerts weak influence on hydrothermal circulation, with the ratio up to 1.5 of vertical permeability to lateral permeability in high permeable layer. Fracture zone existing in basement is the most important factor affecting the circulation field. The effects of a local intrusion are limited to convection intensity above the intrusion and have little impact on the fluid flow on a regional scale. As the result of numerical modelling, key factors affecting the hydrothermal cir-culation are good permeable zone and long-term heat source, not including fluid source.

The Kuroshio frontal instable processes （KFIP） in the East china Sea （ECS） not only have a great impact on the hydrologic characteristics,the pollutants drift,the distribution of seafloor sediment and the ships navigation of the ECS,but also are closely related to the climate changes of the coastal areas of the *** the frequency and area of occurrence of the KFIP have not been studied fully and *** of its high spatial and temporal resolution,MODIS data is a kind of very good data source for surveying and researching the KFIP in the *** aim of this study is to detect the KFIP in the ECS by using MODIS data,and to study the frequency and region of occurrence of the KFIP in the *** selection has coverage of level 2.data of MODIS SST and Kd490 ranging from July 1,2.02.to June 30,2.09 of the ECS when there was no cloud impact or *** using of the data,the minimum standard of the Kuroshio temperature fronts and the diffuse attenuation coefficient （Kd490） fronts of the ECS are *** on these standards and the curvature distinguish methods,the standard of curvature distinguish for the KFIP in the ECS are put *** making use of this standard,we study a total of 2.73 satellite-derived images,and discover that as long as there is no cloud impact from January to May and October to December,the KFIP in the ECS are surely found in MODIS satellite *** June to September,the frequency of occurrence can also reach to 82.9% at ***,it is obtained that there are three source regions of these instability processes,namely,（2.°N,12..5°E） nearby,（2.°N,12.°E） nearby and （30°N,12.°E） *** differences of the characteristics of these instability processes which are generated in different regions are analyzed in the present study.

Three-dimension reconstruction from serial sections has been used in the last decade to obtain information concerning three-dimensional microstructural ge-ometry. One of the crucial steps of three-dimension reconstruction is getting compact and fairing grain contours. Based on the achievement of closed raw con-tours of ceramic composite grains by using wavelet and level set, an adaptive method is adopted for the polygonal approximation of the digitized raw contours. Instead of setting a fixed length of support region in advance, the novel method computes the suitable length of support region for each point to find the best es-timated curvature. The dominant points are identified as the points with local maximum estimated curvatures. Periodic closed B-spline approximation is used to find the most compact B-spline grain boundary contours within the given tolerance. A flexible distance selection approach is adopted to obtain the common knot vector of serial contours consisting of less knots that contain enough degrees of freedom to guarantee the existence of a B-spline curve interpolating each contour. Finally, a B-spline surface interpolating the serial contours is generated via B-spline surface skinning.

A new model is proposed to predict the dynamic subsidence of ground surface above salt cavern gas storage during the leaching and storage, which takes into account the creep of rock salt. In the model, the extended form of Gaussian curve is adopted to figure out the shape of subsidence areas. The corresponding theoretical formulas are derived. In addition, parameters are studied to investigate the surface subsidence as a function of the salt ejection rate, internal pressure, buried depth, diameter, height, running time, etc. Through an example, the subsidence of the salt cavern gas storage located at Jiangsu of china obtained by the new model was compared with those by Peter A F formula, Schober & Sroka formula and FLAC3D through simulation. The results showed the proposed model is precise and correct, and can meet the actual engineering demands. The surface subsidence is equidirectional with the increase of salt ejection rate, depth, diameter, height, and running time, but reverse to the increase of internal pressure. The depth, diameter, running time and internal pressure have great effects on the subsidence, whereas the salt ejection rate and height have little influences on it.

Based on regional compensation and experimental isostasy, the calculating formula of theoretical isostatic response function is deduced when the loads on top correlate with loads from below. A variety of curves of theoretical isostatic response function were calculated with different effective elastic thickness of the plate and by using different proportions between loads on top and loads from below. And preliminary discussion is made on shapes of the curves.

Based on the simulation of a marine ecosystem dynamical model in the Bohai Sea, the Yellow Sea and the East china Sea, chlorophyll data are assimilated to study the spatially varying control parameters （CPs） by using the adjoint method. In this study, the CPs at some grid points are selected as the independent CPs, while the CPs at other grid points can be obtained through linear interpolation with the independent CPs. The independent CPs are uniformly selected from each 30′ × 30′area, and we confirm that the optimal influence radius is 1.2. by a twin experiment. In the following experiments, when only the maximum growth rate of phytoplankton （Vm） is estimated by two given types of spatially varying CPs, the mean relative errors of Vm are 1.2.% and 0.94% while the decrease rates of the mean error of chlorophyll in the surface are 94.6% and 95.8%, respectively. When the other four CPs are estimated respectively, the results are also satisfactory, which indicates that the adjoint method has a strong ability of optimizing the prescribed CP with spatial variations. However, when all these five most important CPs are estimated simultaneously, the collocation of the changing trend of each parameter influences the estimation results remarkably. Only when the collocation of the changing trend of each parameter is consistent with the ecological mechanisms which influence the growth of the phytoplankton in marine ecosystem, could the five most important CPs be estimated more accurately.

The carbon cycle of lower trophic level in the Bohai Sea is studied with a three-dimensional biological and physical coupled model. The influences of the processes (including horizontal advection, river nutrient load, active transport etc. ) on the phytoplankton biomass and its evolution are estimated. The Bohai Sea is a weak sink of the CO2.in the atmosphere. During the cycle, 13.7% of the gross production of the phytoplankton enter the higher trophic level and 76.8 % of it are consumed by the respiration itself. The nutrient reproduction comes mainly from the internal biogeochemical loop and the rem-ineralization is an important mechanism of the nutrient transfer from organic form to inorganic. Horizontal advection decreases the total biomass and the eutrophication in some sea areas. Change in the nutrient load of a river can only adjust the local system near its estuary. Controlling the input of the nutrient, which limits the alga growth, can be very useful in lessening the phytoplankton biomass.
