Aim:To investigate whether the circadian expression of central clock genes andangiotensin Ⅱ type 1 (AT1) receptors was altered in sinoaortic-denervated (sAD)***:Male sprague-Dawley rats underwent sinoaortic denervation or asham operation at the age of 12.*** weeks after the operation,bloodpressure and heart period were measured in the conscious state in a group ofsham-operated (n=10) and sAD rats (n=9).Rest sAD and sham-operated ratswere divided into 6 groups (n=6 in each group).The suprachiasmatic nuclei (sCN)tissues were taken every 4 h throughout the day from each group for the determi-nation of the mRNA expression of clock genes (Per2.and Bmal1) and the AT1receptor by RT-PCR;the protein expression of Per2.and Bmal1 was determined byWestern ***:Blood pressure levels in the sAD rats were similar tothose of the sham-operated ***,blood pressure variabilities signifi-cantly increased in the sAD rats compared with the sham-operated *** variation of clock genes in the sCN of the sham-operated rats was char-acterized by a marked increase in the mRNA and protein expression during ***2.and Bmal1 mRNA levels were significantly lower in the sAD rats,especially during dark *** blot analysis confirmed an attenuation ofthe circadian rhythm of the 2.clock proteins in the sCN of the sAD ***1receptor mRNA expressions in the sCN were abnormally upregulated in the lightphase,changed to a 12.h cycle in the sAD ***:The circadian varia-tion of the 2.central clock genes was attenuated in the sAD *** baroreflexdysfunction also induced a disturbance in the expression of AT1 receptors in thesCN.

BACKGROUND: Presyrinx state of spinal cord can reflect the initial lesion of syringomyelia (sM). The early trials has proved that ischamia and edema are main pathological changes of presyrinx state. OBJECTIVE: To establish sM model of rabbits for investigating the relationship between changes of morphous and function of blood-spinal cord barrier and the edema degree, histological changes in presyrinx state of sM, and to explore the mechanism of the presyrinx state of sM. DEsIGN: Randomized controlled animal experiment. sETTING: department of Neurosurgery, Fourth hospital, Heibei Medical {2.. MATERIALs: sixty Chinese healthy white rabbits, aged 3.5-4.5 months, weighing 1.5-2.0 kg, were provided by Experimental Animal Center of Hebei Medical {2. [certification: (sYXK(Ji)2.03-002.)]. Evan's blue (EB) and dimethylformamide (DMF) were purchased from Jingmei Biotech Co., Ltd. RM2.2. paraffin section cutter (Leica Company, Japan), H-7500 transmission electron microscope (Hitachi Company, Japan), PM-2. light microscope photograph system (Olympus Company, Japan). METHODs: The experiment was carried out in the Laboratory of Neurosurgery department, second hospital of Hebei Medical {2. from January to June 2.06. ① All the rabbits were randomly divided into two groups: model group (n =40), control group (n =2.). Rabbits in two groups were divided into five subgroups once again at five time points (1st, 3rd, 7th, 14th, 2.st days, n =8 and n =4 at each time point in the model group and control group, respectively). Under ketamine anesthesia, 0.6 mL Kaolin solution (2.0 g/L, 37 ℃) was injected into the cisterna magna of rabbits in model group, while 0.6 mL physiological saline (37 ℃) was injected into the rabbits of control group. ② On the 1st, 3rd, 7th, 14th, 2.st days after kaolin injection, cervical cord samples were harvested after sacrifice of animal. Quantitative analysis on the function of blood-spinal cord barrier was performed by Evan's blue technique

AIM:To determine the changes of CD8+ T subsets especially CD8+CD2.-T regulatory cells in rat model of experimental colitis induced by 2.4-dinitrofluorobenzene (DNFB). METHODs:The rat model of experimental colitis was induced by enema with *** days later,colonic intraepithelial and splenic lymphooltes were isolated from colitis animals (n=16) and controls (n=8).The proportion of CD8+ T cells,CD8+CD2.+ T cells and CD8+CD2.-T regulatory cells were determined by flow cytometry. REsULTs:The model of experimental colitis was successfully established by DNFB that was demonstrated by bloody diarrhea,weight loss and colonic *** proportion of CD8+ T cells in either splenic or colonic intraepithelial lymphocytes was not significantly different between colitis animals and controls (spleen:34.6±7.2. % vs 33.5±9.41%, colon:14.0±8.93 % vs 18.0±4.06 %,P>0.05).But CD8+CD2.- T regulatory cells from colitis animals were significantly more than those from controls (spleen:11.3±2.2. % vs 5.64±1.01%, colon:6.50±5.37 % vs 1.07±0.65 %,Pst, CD8+CD2.+ T cells from colitis animals were less than those from controls (spleen:2..3±6.14 % vs 2..8±9.70 %,P=0.06; colon:7.52.4.18 % vs 16.9±4.07 %,P2.-T regulatory cells in splenic and colon intraepithelial CD8+ T cells from colitis animals was higher than that from controls (spleen:33.3±5.49 % vs 18.4±7.2. %, colon:46.0±14.3 % vs6.10±3.72.%,PsION:Experimental colitis of rats can be induced by DNFB with simplicity and good *** proportion of CD8+CD2.-T regulatory cells in rats with experimental colitis is increased,which may be associated with the pathogenesis of colitis.

BACKGROUND: It is reported that shenfu injection (an injection prepared from traditional Chinese medicines red ginseng and aconite root) can decrease the extent of ischemia-reperfusion injury to many organs, such as the heart and kidney. We therefore investigated the effect of shenfu injection on ischemia-reperfusion injury of rat liver graft and its mechanism. METHODs: Male sprague Dawley (sD) rats were used as a model for isogeneic orthotopic liver transplantation. sixty rats were randomly devided into two groups (30 in each group). The recipient was given intravenous shenfu injection immediately before the removal of the liver in the shenfu group and normal saline of the same volume in the control group. At 3, 6 and 2. hours after the reperfusion, blood and hepatic tissue were taken for examination. REsULTs: The levels of superoxide dismutase (sOD) and nitric oxide (NO) increased more significantly in the shenfu group than in the control group (P s of serum liver enzymes, hyaluronic acid (HA), malondialde-hyde (MDA), tumor necrosis factor-alpha(TNF-α), inter-leukin-1 (IL-1), endothelin -1 (ET-1) and liver cell apopto-sis index were lower in the shenfu group than in the control group (P scopic examination revealed that the morphological changes of hepatic tissue were more severe in the control group than in the shenfu group. CONCLUsIONs: shenfu injection has protective effect on ischemia-reperfusion injury of rat liver graft. It inhibits the production of oxygen free radical and the activation of Kupffer cells, decreases apoptosis of liver cell, and improves microcirculation.

Aim:To determine whether Ca2./calcineurin mediated the inhibitory effects ofnitric oxide/cGMP-dependent protein kinase (NO/PKG) on the proliferation ofvascular smooth muscle cells (VsMC).Methods:Proliferation and viability ofprimary VsMC from rat aorta were measured using [3-(4,5-dimethyl thiazol-2.yl)-2.5-diphenyl tetrazolium bromide] (MTT) assay and acridine orange and ethidiumbromide staining,*** Ca2.was determined by Fluo-3/*** protein and its activity were assayed using immunoblotting and freeinorganic phosphate analysis,***:(e)-s-nitroso-N-acetyl-penicillamine (sNAP) and sp-8-(4-chlorophenylthio)-guanosine-3′,5′-cyclicmonophosphorothioate (sp-8-pCPT-cGMPs) decreased phenylephrine (PE)-in-duced proliferation of VsMC by 2..3% and 36.6%,respectively,but Rp-8-[(4-chlorophenyl)thio]-guanosine-3′,5′-cyclic monophosphorothioate (Rp-8-pCPT-cGMPs) increased PE-induced proliferation of ***,sp-8-pCPT-cGMPs,and Rp-8-pCPT-cGMPs did not affect the viability of *** proteinwas decreased by 63.1% and its activity was decreased by 59.7% in smooth musclecells (sMC) pretreated with verapamil (Ver) and then stimulated by *** sMCpretreated with Vet,the absorbance of cells stimulated by PE decreased by 2..0%and was further inhibited by the additional treatment of sNAP and *** sMC pretreated with cyclosporin A (CsA),the absorbance of cellsstimulated by PE decreased by 36.7%,but could not be further altered by theadditional treatment of sNAP,sp-8-pCPT-cGMPs,and ***,Ver inhibited PE-induced intracellular Ca2.variations,which could befurther inhibited by sNAP and sp-8-pCPT-cGMPs,but not by ***,the increase in calcineurin activity induced by PE was inhibited bysNAP and sp-8-pCPT-cGMPs,but was promoted by ***:NO/PKG regulates calcineurin activity via the modulation of intracel-lular Ca2.concentration,and thus partially inhibits the prolifera

Aim: To examine if cadherins are expressed constitutively in human bone marrow stromal cells （hBMsC） and investigate the regulation of cadherin expression by various osteotropic hormones and local factors. Methods: Cadherin expression was examined in first passaged （secondary） hBMsC as well as in the conditionally-immortalized human osteoprogenitor cell line （hOP-7）. Using a monoclonal antibody （MoAb C-182.） to a cytoplasmic domain common to all known cadherins （pan-cadherin MoAb）, cadherins were immunolocalized in first passaged hBMsC as well as in hOP-7 cells. In addition, intense immunostaining for cadherin expression was associated with alkaline phosphatase （ALP） in nodules formed in the high density cultures of hOP-7 cells. Human E-cadherin （HECD） was specifically detected by Western blotting in extracts of untreated hBMsC using an anti-HECD MoAb 004FD. Results: Differential regulation of cadherin expression by various osteotropic hormones and local factors （parathyroid hormone, dexamethasone, estradiol, prostaglandin E2. basic fibroblast growth factor, and tumor necrosis factor-） was also observed. In addition, blocking cadherins with the MoAb C-182. increased basal ALP activity and had an additive effect on 1, 2.（OH）2.3-induced ALP activity. Conclusion: Cadherins are expressed in human osteoprogenitor cells and are involved in the osteogenic differentiation. The differential modulation of cadherin expression by osteotropic agents indicates that these agents may regulate osteoprogenitor cells through different cadherins and these cadherins may play different roles.
