Aim:To develop high-throughput screening(HTS)assays for monoamine oxi-dase(MAO)-A and MAO-B ***:A fluorescence probe basedmethod measuring MAO-A and MAO-B activity was established and optimized,with its sensitivity,stability and specificity *** conditions in-cluding enzyme sources,substrate concentrations,incubation volume and reac-tion time in 384-well format were optimized to achieve sensitive and low consump-tive ***:In optimized conditions,dynamic parameters of MAO-A andMAO-B were *** Kmvalue of serotonin to MAO-A was 1.66μmol/L,while that of benzylamine to MAO-B was 0.80 μmol/*** IC50value of clorgylineto MAO-A was 2.99 nmol/L,and that of deprenyl to MAO-B was 7.04 nmol/L,matching those obtained from traditional spectrometric *** testedsamples,one compound exerted an inhibitory effect on MAO-A activity with IC50as 0.36 μmol/L,and three compounds had an inhibitory effect on MAO-B activitywith IC50as 0.13,0.19,and 0.13 μmol/*** Z′ factor was 0.71±0.03 and 0.75±0.03in MAO-A-inhibitor and MAO-B-inhibitor HTS system,***:The established assays can be well applied to MAO-A and MAO-B inhibitorscreening with high quality,precision and reproducibility.

Aim:To study the in vivo and in vitro metabolism and the effect of para-toluene-sulfonamide （PTS） on cytochrome P450 enzymes （CYP450）.Methods:Total CYP450and microsome protein content were determined after iv pretreatment of rats ***-specific substrates were incubated with rat liver *** isoform activities were determined by using *** chemical inhibitorsadded to the incubation mixture were used to investigate the principal CYP isoformsinvolved in PTS *** effect of PTS on CYP isoforms was investigatedby incubating PTS with specific ***:The groups treated with 33and 99 mg/kg per d PTS,respectively,had a total CYP content of 0.66±0.17 and0.60±0.12.mol/*** Kmand Vmaxwere 92.2.mol/L and 0.0137nmol/min per ***2.7,CYP2.1 and CYP3A2.might contribute to PTS metabolism in therat *** inhibitory effects of sulfaphenazole and ketoconazole on PTS me-tabolism were shown to have a mixed mechanism,whereas PTS metabolism wasinhibited noncompetitively by *** had little effect on the activities ofthe selected CYP ***:Generally speaking,it is relatively safefor PTS to be co-administered with other ***,care should be takenwhen administering PTS with CYP inhibitors and the substrates of CYP2.,CYP2.and CYP3A.

The objective of this study is to investigate the hemodynamics in patient-specific thoracic aortic aneurysm and discuss the reason for formation of aortic plaque.A 3-Dimensional pulsatile blood flow in thoracic aorta with a fusiform aneurysm and 3 main branched vessels was studied numerically with the average Reynolds number of 1399 and the Womersley number of *** on the clinical 2.Dimensional CT slice data,the patient-specific geometry model was constructed using medical image process ***,incompressible,3-Dimensional Navier-Stokes equations were employed to solve the flow *** temporal distributions of hemodynamic variables during the cardiac cycle such as streamlines,wall shear stresses in the arteries and aneurysm were analyzed. Growth and rupture mechanisms of thoracic aortic aneurysm in the patient can be analyzed based on patient-specific model and hemodynamics simulation.

Direct simulation of 3-D MHD(magnetohydrodynamics) flows in liquid metal fusion blanket with flow channel insert(FCI) has been *** kinds of pressure equilibrium slot (PES) in FCI,which are used to balance the pressure difference between the inside and outside of FCI,are considered with a slot in Hartmann wall or a slot in side wall,*** velocity and pressure distribution of FCI made of SiC/SiC_f are numerically studied to illustrate the 3-D MHD flow effects,which clearly show that the flows in fusion blanket with FCI are typical three-dimensional issues and the assumption of 2.D fully developed flows is not the real physical problem of the MHD flows in dual-coolant liquid metal fusion *** optimum opening location of PES has been analyzed based on the 3-D pressure and velocity distributions.

We consider some hyperbolic systems whose solutions are measures, not theusual functions under certain initial data. By introducing new methods which are generatedfrom a new notion on generalized solution, some results of existence and uniqueness ofentropy solution for these systems are obtained. In particular, our results include the caseof gas dynamical equations with zero ***, we also investigate two-dimensional problems of conservation laws. For a specialhyperbolic system, we prove existence and uniqueness of solution, and for simplified Eulerequations, we construct solutions of Riemann problems where initial data have two differentconstant states which are separated by initial discontinuity. Some new phenomena of 2.Dsolutions are discovered.

Applying Hopf bifurcation theory and qualitative theory, we show that the general cubic Lienard equations with quadratic damping have at most three limit cycles. This implies that the guess in which the system has at most two limit cycles is false. We give the sufficient conditions for the system has at most three limit cycles or two limit cycles. We present two examples with three limit cycles or two limit cycles by using numerical simulation.

Objective To search for the microorganisms which have the high selectivity of hydrolyzing glycyrrhizic acid(GL) into 18β-glycyrrhetinic acid-3-O-β-D-glucuronide(GAMG) without glycyrrhetinic acid(GA) *** GL was biotransformed by Aspergillus sp.,the products were separated by chromatography on reverse phase C18 column and semi-preparative HPLC,and their structures were elucidated on the basis of HR-ESI-MS,1D NMR(1H-NMR,13C-NMR,and NOESY) and 2. NMR(1H-1H COSY,HSQC,and HMBC) spectral *** Aspergillus *** partially hydrolyze GL into GAMG(3),along with two minor byproducts,3-O-β-D-glucuronopyranosyl-18β-liquiritic acid(1) and 3-O-β-D-glucuronopyranosyl-2.-hydroxy-18β-glycyrrhetinic acid(2..Conclusion Aspergillus *** the high selectivity of hydrolyzing GL into GAMG without GA byproduct and the yield of GAMG is about 60%.The complete assignments of 1H-NMR and 13C-NMR data for compounds 1 and 2.are reported for the first time.
