原钙黏附蛋白18b(Protocadherin18b,Pcdh18b)属于钙黏附蛋白家族成员.为了研究pcdh18b基因抑制对斑马鱼神经系统发育的影响,针对pcdh18b的翻译起始位点设计一个吗啡啉修饰的反义寡核苷酸抑制其表达,在斑马鱼受精卵一到二细胞期注射并且验证其有效性.注射后用原位杂交和吖啶橙染色检测神经系统的表型和标志基因的表达.pcdh18b下调使神经前体细胞的标志基因neurog1、神经元标志基因elavl3和神经胶质细胞标志基因gfap的表达均出现下调,中后脑边界的标志基因pax2.和wnt1表达减弱并出现神经管分叉现象,同时与后脑分节相关的基因krox2.表达减少.吖啶橙染色显示pcdh18b下调后斑马鱼中脑、后脑及中后脑边界细胞凋亡增多.这些结果表明pcdh18b抑制导致了斑马鱼神经系统发育的异常。

＜正＞Objective Perinatal malnutrition has adverse effects on brain physiology as well as learning and memory activity. However,the mechanism is still incompletely *** oxide（NO） synthesized by neuronal nitric oxide synthase （nNOS） plays an important role in neuronal survival and synaptic plasticity as well as the learning and memory *** present study was to investigate whether 50%perinatal food restriction（FR50） produced deleterious effects on the population of nNOS neurons in C A1 and C A3 and the dentate gyrus（DG） region of the *** Adult virgin female Wistar rats were paired with male rats for a period of 4-5 d until mating was confirmed by observation of a copulatory plug or the presence of sperm in a vaginal rinse under a *** pregnant rats were divided into the control and 50%food-restricted（FR50） *** FR50 groups,dams received 50%of the daily food intake of control mothers from embryonic day 7 until the end of lactation and were returned to normal diet after *** body weight of pup rats were measured on postnatal day（PD）1 to *** PD70,male rat offsprings from control and FR50 groups were subjected to spatial learning and memory test with a Morris water maze（MWM）.After the end of spatial learning and memory ability test,the animals were anesthetized and brain sections including hippocampus were chosen for detection of nNOS-positive neurons by ABC immunohistochemical *** Perinatal FR50 reduced body weight of offspring on postnatal day 1 to 2.,and this type of food restriction impaired learning and memory of adult male offspring rats（PD70） and decreased the number of nNOS-positive cells in the C A1,CA3 and DG regions of the *** Perinatal malnutrition decreases the activity of nNOS in ***,these results provide clues to the knowledge of malnutrition effects upon the brain.

＜正＞Objective Memories are dynamic in *** retrieval,ample evidence has suggested that memories can enter into a labile,protein-synthesis dependent process,referred to as reconsolidation,from which memories can be modified or *** the molecular mechanisms underlying memory reconsolidation could provide crucial insight into the dynamic aspects of memory formation and psychological *** investigate the molecular basis underlying this labile state,we have employed an inducible protein activity-manipulation technique and examined the molecular dynamics of aCaMKIl during long-term memory reactivation in the mouse *** show that an upward-shift of CaMKII activity within the initial 10 minute time-window of synaptic activation is capable of shifting the established long-term potentiation to the higher *** critical time window parallels the initial labile phase of post-reactive long-term memory since post-reactive upward-shift of CaMKII activity during this period also disrupts post-reactive long-term memory ***,post-reactive short-term memory is still intact upon the same ***,we conclude that CaMKII plays a crucial role in numerical and dynamical control and maintenance of synaptic representation of long-term memory traces during reconsolidation process. ＜正＞Fine dispersion or full exfoliation of nanoparticles in matrix is one of the most important factors to achieve high performance polymer *** this talk,mechanically robust,magnetic nylon-6 nanocomposites reinforced by 1D carbon nanotube（CNT）-2. clay nanoplatelet hybrids have been prepared using simple melt-compounding *** direct iron-catalyzed chemical vapor deposition growth of multi-walled carbon nanotubes utilizes iron oxide-immobilized clay nanoplatelets as substrates,carrying out in-situ intercalation and exfoliation of clay nanoplatelets[1].By using such hybridization and co-exfoliation method,the as-obtained heterostructured hybrids used without any purification are demonstrated to be ideal and excellent nanofillers for mechanical reinforcement for fabricating nylon-6 nanocomposites,due to their homogeneous dispersion and strong interfacial interaction with the polymer *** nucleation sites provided by the nanohybrids seem to be favorable to the formation of thermodynamically stable a-phase crystals of nylon-6 with much higher stiffness and hardness thanγ-form of nylon-6,namely,a silicate-induced crystal transformation from theα-form to theγ-form of nylon-6 was greatly inhibited or "shielded" by the CNTs wrapped clay ***,the nanostructured CNT-clay hybrid heterostructures containing residual iron oxide nanoparticles show novel magnetic properties either in bulk solids or in polymer nanocomposites[2..Therefore,this can be probably developed into a facile and practical method to fabricate polymer nanocomposites with high performance and multi-functionality.

Preparation of a gene-engineering mutant of bacteriorhodopsin BR-D96V and corresponding poly(vinyl alcohol)-based functional composite films

Bacteriorhodopsin(BR)exhibits,as a membrane protein in Halobacterium salinarum,unique photoresponsive behaviors,and shows promise as a functional information material.A new mutant of BR with the 96th aspartic acid replaced by valine(BR-D96V)was obtained and then a composite film of BR-D96V in a synthetic polymer matrix was prepared in this *** mutant BR-D96V was expressed in a bacterio-opsin deficient halobacterial strain(L33)by gene *** valine is very hydrophobic,this point mutant keeps the basic biological activities,namely,photoelectric and photochromic ***,the lifetime of M intermediate in the BR mutant is nearly two orders of magnitude longer than that of wild-type BR in neutral aqueous solution,which benefits its potential application as an information *** M lifetime is further significantly prolonged after embedding BR-D96V into poly(vinyl alcohol)(PVA).It was also found that BR-D96V is very sensitive to water content in comparison with wild-type BR and another BR mutant.

Objective As a neuromodulator,somatostatin(SOM) and its receptors are involved in the antinociception in the spinal cord and the supraspinal ***,sex differences are observed in some pain-related *** studies have indicated that the ventrolateral orbital cortex(VLO) is involved in modulation of nociception as part of an endogenous analgesic system consisting of the spinal cord-nucleus submedius-VLO-periaqueductal gray-spinal cord *** aim of the current study was to examine whether the SOM and its receptors are involved in the VLO-induced antinociception in the rat formalin test,and determine whether there was a sex differentiation in the formalin *** After 5%formalin (50μl) was injected subcutaneously into the plantar surface of the hindpaw,the number of times of flinching the hindpaw per 5 min was measured as an index of nociceptive response to observe the effect of microinjection of SOM into the VLO on the nociceptive behavior in different sex *** SOM(10μg/0.5μl)microinjected into the VLO significantly decreased the number of the flinches in either early or later phases in the female rat,and this inhibitory effect was blocked by pre-treatment with its antagonist cyclo-somatostatin(c-SOM,2.μg/0.5μl),whereas the SOM had no effect on the formalin-induced flinching behavior in the male ***,c-SOM microinjected alone into the VLO did not influence the flinching behavior in both male and female *** These results suggest that SOM and its receptors are involved in the antinociception in the VLO of the female,but not the male rats in the formalin test.
