Aim:The aim of the present study was to determine the effect of 5-aminoimidazole-4-carboxamide-ribonucleoside （AICAR） on proliferation,cell cycle,and apoptosisin the human epithelial cervical cancer cell line CaSki ***:Cell countand 3-（4,5-dimethylthiazol-2.yl）-2.5-diphenyltetrazolium bromide assay were usedto determine cell proliferation and *** 332.8 staining was con-ducted to distinguish the apoptotic *** cycle and Annexin-V/propidiumiodide staining were analyzed by fluorescence-activated cell sorting （FACS）.AWestern blot assay was used to evaluate the expression of AKT （also known asprotein kinase B）,mammalian target of rapamycin （mTOR）,p53,and extracellularsignal-regulated kinase （ERK）.Results:AICAR （500 μmol/L） significantly inhibi-ted the proliferation of CaSki cells treated for 2.,48,and 72.h as determined by *** cells at the G1and G2.hases were dramatically decreased while cells atthe S phase were increased in response to AICAR treatment for 2.,48,and 72.*** MTT assay showed less viable cells and Hoechst fluorescent staining showedmore apoptotic cells upon AICAR *** results of the Annexin-Vstaining demonstrated a time-dependent increase of apoptosis in cells treatedwith AICAR for 2.,36,and 48 ***,AICAR activated caspase-3 in atime-dependent *** was also found that AICAR inhibited the phosphory-lation of AKT and mTOR,which are important kinases regulating cell growth *** stimulation obviously increased the expression of the tumorsuppressor p53 and the phosphorylation of ***:AICAR inhibitedproliferation and induced S phase arrest and promoted apoptosis in CaSki cells,which might be mediated by the downregulation of the AKT/mTOR pathway andthe upregulation of the p53/ERK pathway.

荧光各向异性度(fluorescence anisotropy) 测量可以获得荧光分子的转动速度信息,进而了解分子质量、结构、以及与周边环境的相互作用情况. 围绕一台双光子激发扫描荧光成像系统,通过改变外光路和图像记录与处理程序,从而实现了双光子激发荧光各向异性度成像,并针对一些典型样品和体系,展示了该方法的应用. 实验中观察了FITC荧光分子、FITC结合的CD44抗体分子及与肿瘤细胞表面受体结合的FITC-CD44抗体分子. 测量结果表明,不同分子质量、不同微观环境状态下的荧光分子,其各向异性度大小不同,在各向异性度图中能够被明显区分. 荧光各向异性度成像能够定量测量样品微区的各向异性度值,并以二维图像的形式直观表达,是各向异性度测量与成像技术的良好结合.

To determine which subtype of α1-adrenergic receptors plays a role in the regulation of blood pressure, with α1A-adrenergic receptor-mediated vasoconstriction in perfused hindlimb as a control, we compared the inhibitory effects of various ai-adrenergic receptor selective antagonists on the vasopressure responses to phenylephrine between the mean arterial pressure and hindlimb perfusion pressure in anesthetized rats. In Normotensive Wistar rats, the results showed that the inhibitory effects (dose ratios of ED50, Dr) of α1-adrenoceptor selective antagonist (prazosin, Dr 13.5±3.6 vs.15.1±4.3, n = 11), α1A-adrenoceptor selective antagonist (5-methyl-urapidil, Dr 2.4±0.9 vs. 3.7±2.3, n = 12. RS-17053, Dr 3.2.1.6 vs. 4.4±3.3, n =12. and α1D-adrenoceptor selective antagonist (BMY7378, Dr 1.9±0.9 vs. 2.2.0.8, n = 8) on phenylephrine- induced increases of perfusion pressure in the autoperfused femoral beds were the same as that in the mean arterial blood pressure in normotensive Wistar rats. Aim:To investigate the movement of α1A-adrenergic receptors(α1A-AR) stimu-lated by agonist,phenylephrine （PE）,and the dynamics of receptor movement inreal time in single living cells with millisecond ***:We labeledα1A-AR using the monoclonal,anti-FLAG （a kind of tag） antibody and Cy3-conju-gated goat anti-mouse IgG and recorded the trajectory of their transport processin living HEK2.3A cells stimulated by agonist,PE,and then analyzed their dy-namic ***:The specific detection of α1A-AR on the surface ofliving HEK2.3A-α1Acells was achieved.α1A-AR internalize under the stimulationof *** the cells were stimulated with PE for 2. min,apparent colocalizationwas found between α1A-AR and *** 40 min stimulation of PE,trajecto-ries of approximate linear motion in HEK2.3A-α1Acells were recorded,and theirvelocity was ***:The specific labeling method on the livingcell surface provides a convenient means of real-time detection of the behavior ofsurface *** this method we were able to specifically detect α1A-AR andrecord the behavior of individual particles of receptors with 50 ms exposure time inreal time in single living cells.

Many GPCR agonists can activate receptor-tyrosine-kinases(RTKs) through transactivation,the mechanisms of which are poorly ***,we have investigated IGF-1 receptor(IGF-IR) transactivation by GABA receptor using a chemical biology approach in rat primary *** show that GABA receptor activation induces a dynamic assembly and disassembly of a protein complex,including both receptors and their downstream ***,a non-receptor tyrosine kinase,plays a key role in coordinating this dynamic ***,such a dynamic process is critical for transactivation and transactivation-dependent neuronal *** results reveal an important mechanism underlying GPCR transactivation of RTKs.

为了观察尿激酶受体(uPAR)反义核酸对肿瘤细胞体外侵袭的抑制作用,用PCR方法扩增uPARcDNA 5′端 -4 6bp～ +45 4bp一段长 5 0 0bp的序列,利用DNA重组技术将其克隆到病毒载体pAdeno X中,构建出重组腺病毒载体pAdeno X uPAR( - )和pAdeno X uPAR( +).线性化后的重组腺病毒载体转染HEK2.93细胞 7天后,可以获得滴度分别为 1 5× 10 8pfu/ml和 0 5× 10 8pfu/ml的uPAR反义和正义核酸表达重组腺病毒,分别命名为Ad uPAR( -)和Ad uPAR( +).以不同的病毒感染指数(MOI)感染人肺巨细胞癌高转移株 95D肿瘤细胞,3天后用RNA印迹法可以检测肿瘤细胞uPAR正义和反义核酸表达水平,随着MOI的升高,Ad uPAR( - )感染的肿瘤细胞uPAR蛋白水平逐渐下降,肿瘤细胞的体外侵袭能力也明显下降,而感染Ad uPAR( +)的肿瘤细胞无明显变化.结果表明,重组腺病毒可以表达uPAR正义和反义核酸,uPAR反义核酸可以明显抑制人肺巨细胞癌高转移株 95D肿瘤细胞在体外的侵袭能力.
