The development of functional magnetic resonance imaging (fMRI) technology has made it possible to carry out functional brain imaging experiments in small animals. Usually, group data is required to form the assessment of population, which can not only increase the sensitivity of the overall experiment, but also allow the generalization of the conclusion to the whole population. In order to average the signals of functional brain images from different subjects, it is necessary to put all the mapping images into the same standard space (template image). However, up to now, most animal brain templates remain unavailable and it must be done by ourselves. In this study, a template image based on the brains of eight male Wistar rats is obtained, and it is successfully used in our present Alzheimer disease (AD)-like rat model studies as template for spatially normalizing images to the same stereotaxical space. The fMRI results processed with statistical parametric mapping (SPM99) software are in agreement with the results from immunohistochemical experiment, which proves that this method is universally applicable to the pathologic models of other small animals and to human brain lesion studies.

EFFECTS OF PARENT ARTERY SEGMENTATION AND ANEURISMALWALL ELASTICITY ON PATIENT-SPECIFIC HEMODYNAMIC SIMULATIONS

It is well known that hemodynamics and wall tension play an important role in the formation, growth and rupture of aneurysms. In the present study, the authors investigated the influence of parent artery segmentation and aneurismal-wall elasticity on patient-specific hemodynamic simulations with two patient-specific cases of cerebral aneurysms. Realistic models of the aneurysms were constructed from 3-D angiography images and blood flow dynamics was studied under physiologically representative waveform of inflow. For each aneurysm three computational models were constructed： Model 1 with more extensive upstream parent artery with the rigid arterial and aneurismal wall, Model 2 with the partial upstream parent artery with the elastic arterial and aneurismal wall, Model 3 with more extensive upstream parent artery with the rigid wall for arterial wall far from the aneurysm and the elastic wall for arterial wall near the aneurysm. The results show that Model 1 could predict complex intra-aneurismal flow patterns and wall shear stress distribution in the aneurysm, but is unable to give aneurismal wall deformation and tension, Model 2 demonstrates aneurismal wall deformation and tension, but fails to properly model inflow pattern contributed by the upstream parent artery, resulting in local misunderstanding Wall Shear Stress （WSS） distribution, Model 3 can overcome limitations of the former two models, and give an overall and accurate analysis on intra-aneufismal flow patterns, wall shear stress distribution, aneurismal-wall deformation and tension. Therefore we suggest that the proper length of extensive upstream parent artery and aneurismal-wall elasticity should be considered carefully in establishing computational model to predict the intra-aneurismal hemodynamic and wall tension.

Mesenchymal stem cells derived from human placenta suppress allogeneic umbilical cord blood lymphocyte proliferation

ABSTRACTHuman placenta-derived mononuclear cells (MNC) were isolated by a Percoll density gradient and cultured in mesen-chymal stem cell (MSC) maintenance medium. The homogenous layer of adherent cells exhibited a typical fibroblast-like morphology, a large expansive potential, and cell cycle characteristics including a subset of quiescent cells. In vitrodifferentiation assays showed the tripotential differentiation capacity of these cells toward adipogenic, osteogenic andchondrogenic lineages. Flow cytometry analyses and immunocytochemistry stain showed that placental MSC was ahomogeneous cell population devoid of hematopoietic cells, which uniformly expressed CD29, CD44, CD73, CD105,CD166, laminin, fibronectin and vimentin while being negative for expression of CD31, CD34, CD45 and α-smoothmuscle actin. Most importantly, immuno-phenotypic analyses demonstrated that these cells expressed class I majorhistocompatibility complex (MHC-I), but they did not express MHC-II molecules. Additionally these cells could sup-press umbilical cord blood (UCB) lymphocytes proliferation induced by cellular or nonspecific mitogenic stimuli. Thisstrongly implies that they may have potential application in allograft transplantation. Since placenta and UCB arehomogeneous, the MSC derived from human placenta can be transplanted combined with hematopoietic stem cells(HSC) from UCB to reduce the potential graft -versus-host disease (GVHD) in recipients.

AIM:To further investigate the important role of store-operated calcium channels (SOCs) in rat hepatocytes and to explore the effects of SOC blockers on hepatic ischemia-reperfusion injury (HIRI).METHODS:Using freshly isolated hepatocytes from a rat model of HIRI (and controls),we measured cyto-solic free Ca 2+ concentration (by calcium imaging),net Ca 2+ fluxes (by a non-invasive micro-test technique),the SOC current (I SOC ;by whole-cell patch-clamp record-ing),and taurocholate secretion [by high-performance liquid chromatography and 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assays].RESULTS:Ca 2+ oscillations and net Ca 2+ fluxes medi-ated by Ca 2+ entry via SOCs were observed in rat he-patocytes.I SOC was significantly higher in HIRI groups than in controls (57.0 ± 7.5 pA vs 31.6 ± 2.7 pA,P <0.05) and was inhibited by La 3+.Taurocholate secretion by hepatocytes into culture supernatant was distinctly lower in HIRI hepatocytes than in controls,an effect reversed by SOC ***:SOCs are pivotal in *** blockers protected against HIRI and assisted the recovery of se-cretory function in ***,they are likely to become a novel class of effective drugs for prevention or therapy of HIRI patients in the future.

The α subunit of the stimulatory G protein, Gsα, is involved in the stimulation of adenylate cyclase pathway of signal transduction. The various Gsα mRNAs are generated either by alternative splicing or by using alternative promoter. A novel aberrant splicing of Gsα transcript in human leukemia cell lines is reported. The entire coding region of Gsα gene obtained, referred to as GsαLeu, was amplified by PCR with cDNA from Jurkat human leukemia cDNA library or the reverse-transcribed first strand cDNA from human leukemia cell line HL-60 as a template. The result of DNA sequencing indicated that,compared with originally reported Gsα, GsαLeu has two in-frame deletions, one in amino acid residues 23—249 and the other in amino acid residues 254—260, suggesting that G（sαLeu） may play an important role in leukemia. Objective To test a modified otologic drill under different drilling conditions for its ability to identify drilling faults and stop *** Based on force analysis and previous works,an otologic drill was modified and equipped with three *** various conditions,the drill was used to simulate three drilling faults and normal drilling,and signals from the drill were analyzed to extract the characteristic signal.A multi-sensor information fusion system and a stop program were designed to recognize drilling faults and stop *** Signals from each sensor changed consistently in response to drilling condition changes,with high repeatability and *** average identification rate was 72.625%,68.575%,70.5% and 81.3% respectively for the three simulated drilling faults and normal *** stop program stopped drilling in 0.2～ 0.3 seconds when a drilling faults was *** This study shows that the forces acting on the drill bit change predictably in the three simulated drilling conditions;that using suitable BP neural networks,the drilling faults can be reliably identified,and that a stop program based upon characteristic signal recognition can stop drilling quickly upon detecting drilling *** lays a foundation for development of a system capable of predicting drilling faults and automatic drill *** studies are being undertaken for practical application of such a system.

Recruitment of stem cells by hepatocyte growth factor via intracoronary gene transfection in the postinfarction heart failure

We aim to study the amelioration effect of adenovirus5-mediated human hepatocyte growth factor gene transfer on postinfarction heart failure in swine model. Twelve Suzhong young swine were randomly divided into 2 groups of 6 pigs each: Ad5-HGF group and mock-vector Ad5 group. Four weeks after ligation of the left anterior descending coronary artery, Ad5-HGF was intracoronarily transferred into the myocardium. Simultaneously, gate cardiac perfusion imaging was performed to evaluate the heart function. Three weeks later, gate cardiac perfusion imaging was performed again, then the hearts were removed and sectioned for immunohistochemical examination to illustrate the effects of Ad5-HGF on infarcted myocardium. The expression of HGF was examined by ELISA. The results were: (1) compared with the mock-vector Ad5 group, high expression of human HGF was observed in the myocardium of Ad5-HGF group; (2) in the Ad5-HGF group, the number of CD117+ cells co-expressing c-Met per mm2 was significantly larger; (3) the improvement in LVEF was greater in the Ad5-HGF group than in the mock-vector Ad5 group. We concluded that: (1) high expression of human HGF was observed in the myocardium through intracoronary gene transfection; (2) HGF can improve the mobilization of CD117+/c-Met+ stem cells into ischemic myocardium. The amelioration effect of HGF on postinfarction heart failure could not be limited to stimulating angiogenesis, anti-apoptosis, anti-fibrosis, but was also involved in the recruitment of stem cells into myocardium.
