Simulation of 1991-2005 Meiyu seasons in the Yangtze-Huaihe region using BCC_RegCM 1.0

By using BCC_RegCM 1.0 (RegCM for short) from Beijing climate center (BCC), china Meteorological Administration (CMA), the Meiyu season characters over the Yangtze-Huaihe region during 1991-2005 are simulated. The major conclusions of this study may be summarized as the following: (1) RegCM can reproduce the interannual variation and the spatial distribution of the summertime precipitation and temperature in the Yangtze-Huaihe region. (2) By use of a generalized Meiyu criterion and in accordance with model-calculated precipitation and temperature, the Meiyu onset and ending date have been determined. Compared with the observation, RegCM can simulate the interannual variation of the Yangtze-Huaihe Meiyu with preferable capability for most of the normal Meiyu years (such as 1995, 1997, 2000, 2001, 2002 and 2004), especially for the rich Meiyu years of 1996, 1998 and 1999. (3) In terms of the average simulation for the recent 15 years, the timings of onset and ending of Meiyu occur on June 1 and July 13, respectively, which are earlier than the climatological observation. For duration, Meiyu persists for 32 d, 3 d shorter than the observation. The index of Meiyu intensity is 2.45, while the climatological one is 3.00. Therefore, RegCM is capable of simulating the climatological Meiyu duration and intensity, while the capability of simulating the onset and ending date of Meiyu still needs to be improved.

Field measurement and analysis of turbulence coherence for Typhoon Nuri at Macao Friendship Bridge

For the purpose of investigating the turbulent and spatial coherent characteristics of strong wind during typhoon landing period,two 3-dimensional ultrasonic anemometer stations were set up 30 m horizontally apart on the Macao Friendship Bridge to caputure the turbulent wind velocities of Typhoon Nuri. Based on the reliable and representative field measured data,the mean wind speed and direction,turbulence intensity,turbulence integral scale,turbulence power spectra,spatial correlation coefficient and coherence function were statistically evaluated. The field measurement analysis have presented the following results: 1) Two anemometer stations provided consistent results. The mean wind speed variation in time domain presented typical M-shape curves. The strong wind (10-minute mean wind speed higher than 8th grade in Beaufort wind scale) direction changed in a big range up to 122-degrees-angle,indicating the field measurements scoped over the typhoon landing period. 2) The ratio of the longitudinal,lateral and vertical turbulence intensities of the strong wind in the typhoon eye wall region was 1:0.96:0.36. Compared with the code defined ratio 1:0.88:0.5,the lateral component was larger and the vertical component was smaller. 3) The value of integral scale increased when the eye wall of Typhoon Nuri passed over the field measurement site. Before the center of Typhoon Nuri arrived,the integral scale of the strong typhoon wind was about twice compared with that for the non-typhoon wind. 4) The spatial correlation of the turbulent wind,coherence function curve and the decay factor had significant differences at different times during the typhoon process. In the eye wall of the typhoon,the horizontal spatial correlation was relatively strong and horizontally spatial correlation spectrum decayed slower with frequency increase. Impact of EnKF assimilating Himawari-8 all-sky infrared radiance on the forecasting of a warm-sector rainstorm event

Warm-sector rainstorms are highly localized events, with weather systems and triggering mechanisms are not obvious,leading to limited forecasting capabilities in numerical models. Based on the ensemble Kalman filter(PSU-En KF) assimilation system and the regional mesoscale model WRF, this study conducted a simulation experiment assimilating all-sky infrared(IR)radiance for a warm-sector rainstorm in East china and investigated the positive impact of assimilating the Himawari-8 moisture channel all-sky IR radiance on the forecast of the rainstorm. Results indicate that hourly cycling assimilation of all-sky IR radiance can significantly improve the forecast accuracy of this warm-sector rainstorm. There is a notable increase in the Threat Score(TS), with the simulated location and intensity of the 3-hour precipitation aligning more closely with observations. These improvements result from the assimilation of cloud-affected radiance, which introduces more mesoscale convective information into the model's initial fields. The adjustments include enhancements to the moisture field, such as increased humidity and moisture transport, and modifications to the wind field, including the intrusion of mid-level cold air and the strengthening of lowlevel convergent shear. These factors are critical in improving the forecast of this warm-sector rainstorm event.

Temporal and Tight Hepatitis C Virus Gene Activation in Cultured Human Hepatoma Cells Mediated by a Cell-Permeable Cre Recombinase

Conditional gene expression has greatly facilitated the examination of the functions of particular gene products. Using the Cre/lox P switching expression system, we plan to develop efficient conditional transgeneactivation of hepatitis C virus core protein (HCV-C) cDNA (nucleotide 342–914) in the transgenic mice to overcome “immune tolerance” formed during the embryonic period and “immune escape” against hepatitis virus antigen in our project. To use this system in vivo, the dormant transgenic construct, i.e., pApoE-SCS-EGFP-HCV-C, was generated using techniques of standard molecular biology. The liver- specific human apoE promoter was here used to target expression of genes of interest (EGFP and HCV-C) to murine liver. Prior to generating the transgenic mice, the availability of Cre/lox P system and construct functionality were successfully verified by a cell-free recombination system and via checking the expression of EGFP and HCV-C in the human hepatoma cells at the mRNA and protein levels. These results suggest that the Cre/lox P system could tightly control expression of EGFP and HCV-C in vitro, which laid a solid foundation to conditionally activate expression of target gene(s) in transgenic mice by Cre-mediated site-specific recombination.

Tetracycline-controlled transcriptional regulation systems: countermeasures to eliminate basal transgene leaks in Tet-based systems

To analyze the function of any given transgene(s) accurately in transgenic mice, and to produce credible transgenic animal models of various human diseases (precisely and realistically mimicking disease states), it is critical to be able to control gene expression in the animals conditionally. The ability to switch gene expression "on" or "off" in the restricted cells or tissue(s) at specific time(s) allows unprecedented flexibility for exploring gene function(s) in both the health and the disease. Pioneering work on inducible transgene expression has led to the development of a wide variety of controlled gene expression systems that meet this criterion. Among them, the tetracycline-inducible systems (e.g. Tet-off and Tet-on) have been widely, frequently and successfully employed in vitro and in vivo. These systems, however, are not always tight but leaky; sometimes the leakage is significant. In some circumstances, the resulting leak is acceptable, but in others, it is more problematic. Though these systems face this disadvantage, i. e. basal transgene leakage in vitro and in vivo, several approaches, including using improved versions (e.g. rtTA2S-M2 and rtTA2S-S2) of rtTA, tetracycline-controlled transcriptional silencer (tTS), an "ideal" minimal promoter in responsive components or combinations thereof, have been developed to avoid this limitation effectively. In this review we discuss the countermeasures available to eliminate basal transgene leakage from Tet-based systems.

Entropy correction of BTZ black holes in a tunneling framework

In this paper, using the Parikh-Wilczek tunneling framework, we first calculate the emission rates of non-rotating BTZ black holes and rotating BTZ black holes to second order accuracy. Then, by assuming that the emission process satisfies an underlying unitary theory, we obtain the corrected entropy of the BTZ black holes. A log term emerges naturally in the expression of the corrected entropy. A discussion about the inverse area term is also presented.

Antiangiogenic therapy:a novel approach to overcome tumor hypoxia

Hypoxia is a common phenomenon in solid tumors. Resistance of hypoxic tumor cells to radiation is a significant reason of failure in the local control of tumors. The growth and metastasis of solid tumors rely on blood vessels. Antiangiogenic agents mainly target tumor blood vessels, and radiation therapy mainly targets tumor cells. Combination of antiangiogenic treatment and radiation exhibits synergistic effect, which improves the response of tumors to radiation therapy. The mechanisms of interaction between antiangiogenic agents and ionizing radiation are complex and involve interactions between tumor cells and tumor microenvironment, including tumor oxygenation, stroma, and vasculature. The original mechanism of antiangiogenesis is to induce ischemia and hypoxia in tumors, thereby, "starve" the tumors. However, recently, emerging data suggest that antiangiogenic agents could reduce the proportion of hypoxic cells through normalizing tumor vasculature, decreasing oxygen consumption, and other mechanisms. The use of antiangiogenic agents provides a new approach to overcome the hypoxia problem, and ultimately improves the efficacy of radiation therapy. In this review, we discuss tumor hypoxia, tumor angiogenesis and its regulation, mechanisms of antiangiogenic therapy combined with radiation therapy, and how antiangiogenic therapy overcomes tumor hypoxia.

Either cetuximab or bevacizumab can improve the survival of patients with metastastic colorectal cancer (mCRC) if administered combided with cytotoxic agents. However, the effect of two or more target agents in combination is uncertain in these patients. Here, we reported a patient with mCRC successfully treated by a combination of target agents after the failure of chemotherapy. The patient received palliative resection of primary tumor followed by 9 cycles of postoperative XELOX regimen, cytokine-induced killer cell (CIK)-based biotherapy, traditional Chinese medicine, particle implantation in the lung metastatic lesions. The tumor progressed 20 months after the standard treatments. Then, the regimen cetuximab, bevacizumab and cefitinib was applied. During the treatment with targeted agents, grade IV acne-like rash and relatively severe parionychia of the toes occurred. Both of them recovered smoothly. The PET-CT reexamination at 40 days after the target treatment showed that the metabolism of mediastinal lymph nodes basically recovered to a normal level. The combination of multiple targeted agents obtained a progression-free survival(PFS) of 11 months and the patient with a good quality of life during this period.

Chandra observations of the H_2O megamaser galaxy Mrk1210

We present the first Chandra X-ray observations of the H2O megamaser galaxy Mrk1210 (UGC4203), a Seyfert 2 galaxy at an approximate distance of D～57.6 Mpc. The Chandra X-ray image, with by far the highest angular resolution (～1"), displays an unresolved compact core toward the nuclear region of Mrk1210. Comparisons with the previous X-ray observations in the nuclear emission and the spectral shape indicate a fairly stable phase between 2001 (BeppoSAX and XMM-Newton) and 2004 (Chandra) after a dramatic variation since 1995 (ASCA). The best-fit model of Chandra X-ray spectrum consists of two components. The soft scattered component can be best fitted by a moderately absorbed power-law model adding a spectral line at ～0.9 keV (possibly a Ne-Kα fluorescent line), while the hard nuclear component can be well reproduced by a heavily absorbed power-law model (NH～2×1023 cm-2) with an additional line at ～6.19 keV (close to the Fe-Kα fluorescent line). The derived absorption-corrected X-ray luminosity implies that the dramatic variation of spectral properties is caused by significant changes of the absorbing column density along the line-of-sight, while the intrinsic nuclear X-ray lu-minosity remains stable. In this case, the absorbers should be anisotropic and its size can be constrained to be less than 0.0013 pc. In addition, we also estimate the mass of central engine, the disk radius and the accretion rate of the accretion disk to be 107.12±0.31M⊙, ～1 pc and 0.006, respectively.
