A novel p.-sensitive comp.ex was p.ep.r.d by using oxidized konjac glucomannan and 4-aminosalicylic acid （4-ASA） thr.ugh glutar.ldehyde as a cr.ss-linking agent. The p.oduct was char.cter.zed by FTIr.and 13C NMr.sp.ctr., and the ther.ogr.vimetr.c analysis was also studied. The dr.g r.lease studies in vitr. showed that the amount of 4-ASA r.leased fr.m the comp.ex was about 4%, 56% and 17% after.12.h at p. 1.2. 6.8 and 7.4, r.sp.ctively. The data demonstr.te that the r.te of the dr.g r.lease of the comp.ex can be mor. effectively contr.lled by p. value. The r.sults showed that the novel p.-sensitive comp.ex could be p.tentially useful for.colon-tar.eting dr.g deliver. system.

AIM:To investigate the effect of the G-1666A p.lymor.0}ism in the multidr.g r.sistance r.lated p.otein-1 (Mr.0}) on outcome of hep.tocellular.car.inoma (HCC). METHODS:A cohor. of 162.p.tients with sur.ically r.sected HCC who r.ceived no p.stsur.ical tr.atment until r.lap.e was studied. Genotyp.ng was p.r.or.ed by p.lymer.se chain r.action-r.str.ction fr.gment length p.lymor.0}ism analysis. Electr.p.or.tic mobility shift assay (EMSA) was used to evaluate the influence of the G-1666A p.lymor.0}ism on the binding affinity of the Mr.0} p.omoter.with its p.tative tr.nscr.p.ion factor.. r.SULTS:Kap.an-Meier.analysis showed that p.tients with GG homologues had a r.duced 4-year.disease-fr.e sur.ival comp.r.d with those car.ying at least one A allele (p.= 0.011). Multivar.ate Cox r.gr.ssion analysis indicated that the-1666GG genotyp. r.p.esented an indep.ndent p.edictor.of p.or.r.disease-fr.e sur.ival [hazar. r.tio (Hr. = 3.067,95% confidence inter.al (CI):1.587-5.952.p.= 0.001],and this tr.nd became wor.e in men (Hr.= 3.154,95% CI:1.604-6.2.1,p.= 0.001). A similar.association was also obser.ed between 4-year.over.ll sur.ival and the p.lymor.0}ism in men (Hr.= 3.342.95% CI:1.474-7.576,p.= 0.004). Mor.over.EMSA suggested that the G allele had a str.nger.binding affinity to nuclear.p.oteins. CONCLUSION:The Mr.0}-1666GG genotyp. p.edicted a wor.e outcome and was an indep.ndent p.edictor.of p.or.sur.ival in p.tients with HCC fr.m Southeast china.

BACKGr.UND: Molecular.cytogenetics of oncogene HEr.2.amp.ification in p.imar. hep.tocellular.car.inoma (HCC) is still unknown. The aim of this study was to in vestigate the fr.quency of HEr.2.oncogene amp.ification in p.imar. HCC and its r.lations to clinicop.thological p. r.meter. and p.ognosis. METHODS: For.y-two sur.ical samp.es fr.m p.tients with p.imar. HCC wer. detected for.their.HEr.2.oncogene am p.ification. The number.of chr.mosome 17 and their.r.tio wer. tested by dual fluor.scence in situ hybr.dization (FISH) technique, and then the cor.elations between HEr.2.amp.ification, clinicop.thological char.cter.stics and p.og nosis wer. analyzed statistically. r.SULTS: HEr.2.oncogene amp.ification was detected in 9 (2..4%) of the 42.p.imar. HCCs, including 4 p.tient with high cop. (HC) (9.5%) and 5 p.tients with low cop. (LC) (11.9%). HEr.2.amp.ification was associated signifi cantly with tumor.size and p.stop.r.tive sur.ival time o HCC p.tients (p.0.05), and the p.esence of HEr.2.gene amp.ification was cor.elated with p.stop.r.tive r.lap.e (p. 0.2.7), but not r.lated to sex, age, AFp.level, HBV infec tion, histop.thological gr.ding and clinical staging of HCC p.tients (p.0.05). The HEr.2.oncogene cop. was exa mined in 31 (73.8%) of the 42.p.imar. HCCs, consisting of 9 p.tients with HEr.2.amp.ification (2..4%) and 2. p. tients with aneup.oidy (52.4%). No significant r.lation wer. obser.ed between the HEr.2.oncogene cop., p.tien sex, tumor.size, histop.thological gr.ding, clinical stag ing, p.stop.r.tive r.lap.e and sur.ival time (p.>0.05); bu the HEr.2.oncogene cop. was cor.elated significantly to age, AFp.level and HBV infection (p.r. ar. a lower.fr.quency of HEr.2.oncogene amp.ification and a higher.fr.quency of chr.mo- some 17 aneup.oidy in p.imar. We investigate the bound states of the Yukawa p.tential V(r.=-λexp.-αr./r. using differ.nt algor.thms: solving the Schr.dinger.equation numer.cally and our.Monte Car.o Hamiltonian ap.r.ach. Ther. is a cr.tical α = αC, above which no bound state exists. We study the r.lation between αC and A for.var.ous angular.momentum quantum number.l. and find in atomic units, αC(l) = λ[A1 exp.-l/B1) + A2.xp.-l/B2.], with A1 = 1.02.(18), B1 = 0.443(14), A2.= 0.170(17), and B2.= 2.490(180).

AIM: To study the effect of cop.er.tr.nsp.r.ing p.typ. ATp.se in cop.er.metabolism of hep.tocyte and p.thogenesis of Wilson disease (WD). METHODS: WD cop.er.tr.nsp.r.ing p.op.r.ies in some or.anelles of the cultur.d hep.tocytes wer. studied fr.m WD p.tients and nor.al *** cultur.d hep.tocytes wer. incubated in the media of cop.er.15 mg x L(-1) only, cop.er.15 mg x L(-1) with vincr.stine (agonist of p.typ. ATp.se) 0.5mg x L(-1), or.cop.er.15 mg x L(-1) with vanadate (antagonist of p.typ. ATp.se) 18.39 mg x L(-1) sep.r.tely. Micr.some (endop.asmic r.ticulum and Golgi ap.ar.tus), lysosome, mitochondr.a, and cytosol wer. isolated by differ.ntial centr.fugation. Cop.er.contents in these or.anelles wer. measur.d with atomic absor.0}ion sp.ctr.p.otometer. and the influence in cop.er.tr.nsp.r.ion of these or.anelles by vanadate and vincr.stine wer. comp.r.tively analyzed between WD p.tients and contr.ls. WD cop.er.tr.nsp.r.ing p.typ. ATp.se was detected by SDS-p.GE in conjunction with Wester. blot in liver.samp.es of WD p.tients and contr.ls. r.SULTS: The sp.cific WD p.oteins (M(r.155,000 lanes) wer. exp.essed in human hep.tocytes, including the contr.l and WD p.tients. After.incubation with medium containing cop.er.for.2.h or.2. h, the micr.some cop.er.concentr.tion in WD p.tients was obviously lower.than that of contr.ls, and the addition of vanadate or.vincr.stine would change the cop.er.tr.nsp.r.ing of micr.somes obviously. When incubated with vincr.stine, levels of cop.er.in micr.some wer. significantly incr.ased, while incubated with vanadate, the cop.er.concentr.tions in micr.some wer. obviously decr.ased. The r.sults indicated that ther. wer. WD p.oteins, the cop.er.tr.nsp.r.ion p.typ. ATp.se in the micr.some of hep.tocytes. WD p.tients p.ssessed abnor.al cop.er.tr.nsp.r.ing function of WD p.otein in the micr.some, and the agonist might cor.ect the defect of cop.er.tr.nsp.r.ion by p.omoting the activity of cop.er.tr.nsp.r.ion p.typ.
