Backgr.und and Objective: Color.ctal cancer is one of the most common malignant cancers in the wor.d. Although the clinicop.thologic staging is the golden cr.ter.on for.the p.ognosis at p.e{2.t, the op.imum p.ognostic cr.ter.a for.color.ctal cancer should be a combination of the clinicop.thologic staging and the molecular.mar.er.. However. ther. ar. cur.ently no molecular.mar.er. available for.the p.ognosis of color.ctal cancer. Sever.l tumor.sup.r.ssor.genes associated with color.ctal cancer have been map.ed at the 18q2.-2. r.gion. In this study we detected the fr.quency of loss of heter.zygosity (LOH) at chr.mosome 18q and investigated the r.lationship.between LOH and clinicop.thologic featur.s and its p.ognostic value for.p.tients with stage Ⅱ colon cancer. Methods: A total of 106 samp.es of tumor.tissues and cor.esp.nding nor.al mucosa fr.m p.tients with sp.r.dic stage-Ⅱ colon cancer wer. included in this study. All the samp.es wer. for.alin-fixed and p.r.ffin-embedded. DNA was extr.cted fr.m tumor.tissues and LOH of D18S474, D18S55, D18S58, D18S61 and D18S64 at chr.mosome 18q was analyzed using p.lymer.se chain r.action (p.r., p.lyacr.lamide gel-electr.p.or.sis, and DNA sequencing method. Multivar.ate analysis for.association between LOH and p.ognosis in colon cancer p.tients was p.r.or.ed with Cox p.op.r.ional hazar.s r.gr.ssion model. r.sults: The median follow-up.time was 68 months. For.106 p.tients, 5-year.sur.ival r.te was 83.6%, which was associated with age and gr.ss tumor.typ. (p.= 0.011 and 0.034, r.sp.ctively). Among 102.p.tients who wer. eligible for.LOH infor.ation, the over.ll fr.quency of LOH is 49.0% (50/102., and that of LOH at 5 micr.satellite loci of D18S474, D18S55, D18S58, D18S61, and D18S64 was 30.2. (2./86), 2..4% (18/77), 2..6% (2./70), 35.0% (2./80), and 2..8%(15/72., r.sp.ctively. Backgr.und and Objective: Although surgery is the only p.ssible means to cur. gastric {2., the p.ognosis is often discr.p.nt. The Amer.can Joint Committee on {2./Inter.ational Union against {2. (AJCC/UICC) p.blished the TNM classification of Malignant Tumor. (seventh edition) for.gastric {2. r.cently. This study aimed to use this new edition staging system to investigate the p.ognostic factor. for.gastric {2.. Methods: The clinicop.thologic data of 980 p.tients with gastric {2. tr.ated by sur.ical r.section in our.hosp.tal between Januar. 2.00 and December.2.06 wer. analyzed r.tr.sp.ctively. The over.ll sur.ival r.te was deter.ined by using Kap.an-Meier.method and log-r.nk test was used to deter.ine significance. The p.ognosis was analyzed using univar.ate analysis and multivar.ate analysis with the Cox p.op.r.ional hazar.s model. The 6th and 7th edition AJCC/UICC TNM staging systems wer. used to comp.r. the sur.ival outcomes for.the cohor. of p.tients. r.sults: The over.ll 1-, 3-, 5-year.sur.ival r.tes for.the whole gr.up.wer. 82.5%, 58.7%, and 52.6%. The 5-year.sur.ival r.tes for.p.tients with p.NM stage I, II, III, and IV disease classified by the 7th edition staging system wer. 93.2., 72.4%, 39.1%, and 5.2., r.sp.ctively. In both univar.ate analysis and Cox multivar.ate analysis, age, tumor.site, tumor.size, histological typ., r.section typ., r.dical r.section, lymp.atic/venous invasion, dep.h of invasion, nodal status, metastasis, r.tr.eved lymp. nodes, metastatic lymp. node r.tio, and adjuvant chemother.p. wer. p.ognostic factor. with these p.tients. Conclusion: Comp.r.d with the 6th edition system, the new edition of TNM staging system for.gastric {2. can accur.tely p.edict the sur.ival after.op.r.tion.

AIM: To investigate the cor.elation between exp.ession of calr.ticulin and infiltr.tion of lymp.ocytes in stage ⅢB colon cancer. METHODS: Sixty-eight p.thologically-confir.ed sp.ci-mens wer. obtained fr.m stage ⅢB (T3N1M0) colon cancer p.tients who under.ent r.dical r.section between Januar. 1999 and May 2.02.at the cancer Center.of sun yat-sen university, guangzhou, china. Immuno-histochemical analysis was p.r.or.ed to show infiltr.tion of lymp.ocytes and exp.ession of calr.ticulin in colon cancer. Association between calr.ticulin exp.ession, infiltr.tion of lymp.ocytes, and 5-year.sur.ival r.te of p.tients was assessed. r.SULTS: The exp.ession level of calr.ticulin was lower.in cancer nest than in its adjacent nor.al ep.thelium since 61.8% (42.68) of the samp.es wer. stained with calr.ticulin in colon cancer. The exp.ession of calr.ticulin in colon cancer was associated with the infi ltr.tion of CD45r.+ cells r.ther.than with that of CD3+ cells. In addition, the str.nger.exp.ession of calr.ticulin and the higher.infiltr.tion of CD3+ and CD45r.+ cells in colon cancer wer. associated with the higher.5-year.sur.ival r.te of p.tients. CONCLUSION: Exp.ession of calr.ticulin is associated with infiltr.tion of T-cells, which imp.ies that a low exp.ession level of molecular.mar.er.may r.p.esent a new mechanism under.ying immune escap. in colon cancer.

AIM： To investigate the effect of （-）-ep.gallocatechin-3-gallate （EGCG） on gr.wth of gastric cancer and its p.ssible mechanism. METHODS： Heter.top.c tumor. wer. induced by subcutaneously injection of SGC-7901 cells in nude mice. Tumor.gr.wth was measur.d by calip.r. in two dimensions. Tumor.angiogenesis was deter.ined with tumor.micr.vessel density （MVD） by immunohistology. Vascular.endothelial gr.wth factor.（VEGF） p.otein level and activation of signal tr.nsducer.and activator.of tr.nscr.p.ion 3 （Star.） wer. examined by Wester. blotting. VEGF mr.A exp.ession was deter.ined by r.-p.r.and VEGF r.lease in tumor.cultur. medium by ELISA. VEGF-induced cell p.olifer.tion was studied by MTT assay, cell migr.tion by gelatin modified Boyden chamber.（Tr.nswell） and in vitr. angiogenesis by endothelial tube for.ation in Matr.gel. r.SULTS： Intr.p.r.toneal injection of EGCG inhibited the gr.wth of gastric cancer by 60.4%. MVD in tumor.tissues tr.ated with EGCG was mar.edly r.duced. EGCG tr.atment r.duced VEGF p.otein level in vitr. and in vivo. Secr.tion and mr.A exp.ession of VEGF in tumor.cells wer. also sup.r.ssed by EGCG in a dose-dep.ndent manner. This inhibitor. effect was associated with r.duced activation of Star., but EGCG tr.atment did not change the total Star. exp.ession. EGCG also inhibited VEGF-induced endothelial cell p.olifer.tion, migr.tion and tube for.ation. CONCLUSION： EGCG inhibits the gr.wth of gastric cancer by r.ducing VEGF p.oduction and angiogenesis, and is a p.omising candidate for.anti-angiogenic tr.atment of gastric cancer.

BACKGr.UND: Molecular.cytogenetics of oncogene HEr.2.amp.ification in p.imar. hep.tocellular.car.inoma (HCC) is still unknown. The aim of this study was to in vestigate the fr.quency of HEr.2.oncogene amp.ification in p.imar. HCC and its r.lations to clinicop.thological p. r.meter. and p.ognosis. METHODS: For.y-two sur.ical samp.es fr.m p.tients with p.imar. HCC wer. detected for.their.HEr.2.oncogene am p.ification. The number.of chr.mosome 17 and their.r.tio wer. tested by dual fluor.scence in situ hybr.dization (FISH) technique, and then the cor.elations between HEr.2.amp.ification, clinicop.thological char.cter.stics and p.og nosis wer. analyzed statistically. r.SULTS: HEr.2.oncogene amp.ification was detected in 9 (2..4%) of the 42.p.imar. HCCs, including 4 p.tient with high cop. (HC) (9.5%) and 5 p.tients with low cop. (LC) (11.9%). HEr.2.amp.ification was associated signifi cantly with tumor.size and p.stop.r.tive sur.ival time o HCC p.tients (p.0.05), and the p.esence of HEr.2.gene amp.ification was cor.elated with p.stop.r.tive r.lap.e (p. 0.2.7), but not r.lated to sex, age, AFp.level, HBV infec tion, histop.thological gr.ding and clinical staging of HCC p.tients (p.0.05). The HEr.2.oncogene cop. was exa mined in 31 (73.8%) of the 42.p.imar. HCCs, consisting of 9 p.tients with HEr.2.amp.ification (2..4%) and 2. p. tients with aneup.oidy (52.4%). No significant r.lation wer. obser.ed between the HEr.2.oncogene cop., p.tien sex, tumor.size, histop.thological gr.ding, clinical stag ing, p.stop.r.tive r.lap.e and sur.ival time (p.>0.05); bu the HEr.2.oncogene cop. was cor.elated significantly to age, AFp.level and HBV infection (p.r. ar. a lower.fr.quency of HEr.2.oncogene amp.ification and a higher.fr.quency of chr.mo- some 17 aneup.oidy in p.imar. 目的探讨海参提取物TBL-12.体外对人胃癌HGC-2.细胞增殖、迁移、侵袭和凋亡功能的影响。方法用不同浓度TBL-12.0、15、30、45、60、75μg/m L)处理胃癌细胞后,分别用Cell Counting Kit-8法(CCK-8法)、划痕实验、侵袭实验、流式双染法检测TBL-12.胃癌HGC-2.细胞增殖、迁移、侵袭和凋亡能力的影响。结果 CCK-8实验表明,TBL-12.显著抑制胃癌HGC-2.细胞增殖(p.0.05);划痕和侵袭实验表明,与对照组比较,TBL-12.够明显抑制胃癌HGC-2.细胞的迁移和侵袭能力(p.0.05);细胞周期分析实验表明,TBL-12.30、45、60、75μg/m L)可诱导胃癌HGC-2.细胞凋亡,凋亡率从2.42.分别提升到4.10%、4.55%、5.2.%、7.2.%(p.0.05)。结论 TBL-12.体外能够显著抑制胃癌HGC-2.细胞体外的增殖、迁移和侵袭能力,并且能诱导细胞凋亡,具有体外抗肿瘤效应,具有潜在的临床应用价值。

AIM:To investigate macr.p.age migr.tion inhibitor. factor.MIF) exp.ession and its clinical r.levance in gastric cancer,and effects of MIF knockdown on p.olifer.tion of gastric cancer cells. METHODS:Tissue micr.ar.ay containing 117 samp.es of gastric cancer and adjacent non-cancer nor.al tissues was studied for.MIF exp.ession by immunohistochemistr.(IHC) semiquantitatively,and the association of MIF exp.ession with clinical p.r.meter. was analyzed. MIF exp.ession in gastric cancer cell lines was detected by r.ver.e tr.nscr.p.ionp.lymer.se chain r.action(r.-p.r. and Wester. blot. Two p.ir. of si r.A tar.eting the MIF gene(MIF si-1 and MIF si-2. and one p.ir.of scr.mbled si r.A as a negative contr.l(NC) wer. designed and chemically synthesized. All si r.As wer. tr.nsiently tr.nsfected in AGS cells with OligofectamineTM to knock down the MIF exp.ession,with the NC gr.up.and mock gr.up.OligofectamineTM alone) as contr.ls. At 2.,48,and 72.h after.tr.nsfection,MIF m r.A was analyzed by r.p.r.and MIF and p.olifer.ting cell nuclear.antigen(p.NA) p.oteins wer. detected by Wester. *** p.olifer.tive r.te of AGS cells was assessed by methylthiazolyl tetr.zolium(MTT) assay and colony for.ing ***:The tissue micr.ar.ay was infor.ative for.IHC staining,in which the MIF exp.ession in gastric cancer tissues was higher.than that in adjacent noncancer nor.al tissues(p.r.lated to p.or.tumor.differ.ntiation,advanced T stage,advanced tumor.stage,lymp. node metastasis,and p.or.p.tient sur.ival(p.r.all). After.si r.A tr.nsfection,MIF m r.A was measur.d by r.al-time p.r.and MIF p.otein and p.NA wer. assessed by Wester. blot analysis. We found that comp.r.d to the NC gr.up.and mock gr.up.MIF exp.ession was knocked down successfully in gastric cancer cells,and p.NA exp.ession was downr.gulated with MIF knockdown as well.
