Altered expression of nuclear matrix proteins in etoposide induced apoptosis in HL-60 cells

The events of cell death and the expression of nuclear matrix protein (NMP) have been investigated in a promyelocytic leukemic cell line HL-60 induced with etoposide. By means of TUNEL assay, the nuclei displayed a characteristic morphology change, and the amount of apoptotic cells increased early and reached maximun about 39% after treatment with etoposide for 2 h. Nucleosomal DNA fragmentation was observed after treatment for 4 h. The morphological change of HL-60 cells, thus, occurred earlier than the appearance of DNA ladder. Total nuclear matrix proteins were analyzed by 2-dimensional gel electrophoresis. Differential expression of 59 nuclear matrix proteins was found in 4 h etoposide treated cells. Western blotting was then performed on three nuclear matrix acssociated proteins, PML, HSC70 and NuMA. The expression of the suppressor PML protein and heat shock protein HSC70 were significantly upregulated after etoposide treatment, while NuMA, a nuclear mitotic apparatus protein, was down regulated. These results demonstrate that significant biochemical alterations in nuclear matrix proteins take place during the apoptotic process.

The control of molecular assembly in the solid state is an important theme of modern chemistry. The activity in the area of supramolecular structures is investigated. The self-assembly of molecules can form well-defined supramolecular structures under the influence of drive forces. The self-assembly properties of Cu(pp)2[pp = 2-(2-hydroxyphenol)-pyridine] are analyzed under different conditions.

Current serum neutralization assays based on the inhibition of the cytopathic effect（Nt-CPE） need to ma nipulate live viruses, which are time-consuming, labor-intensive, and have the potential exposure to infectious agents, so a safe and objective assay via pseudovirus for the fast and efficient detection of enterovirus 71（EV71） neutralizing antibodies was developed. First, we generated EV71 pseudovirus containing firefly luciferase gene in place of the capsid gene P1 in EV71 genome. Vero cells infected with 200 CCID50（50% cell culture infective dose） of EV71 pseudovirus for 24 h were found to have the best performance. Seval sera were measured by EV71 pseudoparticle neutralization assay（Nt-PPN） and the conventional serological method Nt-CPE. Neutralizing antibody titers measured by Nt-PPN and those obtained by Nt-CPE demonstrate a high correlation between the two methods. Overall, the PPN assay represents a valid alternative to conventional serological methods for the evaluation of EV71 neutralizing anti bodies. This method can be used for detecting neutralizing antibodies of other picornaviruses, such as hepatitis A vi rus（HAV） and coxsackievirus 16（CVA16）, and make it possible to determine whether there is cross-reactivity be tween EV71 and CVA16.

Dose-dependent Cytotoxicity and Oxidative Stress Induced by "Naked" Fe_3O_4 Nanoparticles in Human Hepatocyte

The dose-dependent cytotoxicity effect on human hepatocyte（HL-7702 cells） induced by ＂naked＂ Fe3O4 nanoparticles was assessed through cell viabilities and lactate dehydrogenase（LDH） activities. Three important oxi dative indexes of the cells by glutathione peroxidase（GSH-Px）, superoxide dismutase（SOD） and malondialde hyde（MDA） were determined. The good correlation of the cell viabilities with their GSH-Px, SOD and MDA levels indicated that the cytotoxicity is related to activation of oxidative stress induced by Fe3O4 nanoparticles. The oxida tive stress also leads to corresponding DNA damage in a similar dose-dependent manner, followed by the changes of cell cycle and cell apoptosis. Such work provides important experimental data for the safety evaluation of superpa ramagnetic Fe3O4 nanoparticles.

By introducing the distribution probability of structural units in austenite contain- ing alloying elements and considering its effects on phase transformation, this paper establishes a calculation model of distribution probability of structural units. A new valence electron structure (VES) parameter-transformation effect coefficient of alloying elements (HL), is defined and then studied both theoretically and ex- perimentally. The relationship between the parameter HL and the multiplying factor (the quenching capability factor) of alloying elements is studied. The results indi- cate that the two parameters (HL and the quenching capability factor) have the same variation characteristic and substance feature. Therefore, the multiplying factor virtually expresses the relative quantity of structural units in the alloying elements-containing austenite.

Artificial Induction of Twinning by an Active Immunization of Beef Cows Against Inhibin Partially Purified from Porcine Seminal Plasma

Two hundred and seventy multiparous Chinese Yellow cattle (beef) were selected at 1 to 3 months postpartum and divided into three groups (90 cows for each). animals were given both a primary and booster immunizations with a total dose of 3 mg (Group Th) or 1.5 mg (Group Tl) of seminal preparation containing inhibin activity, emulsified with Freund's complete adjuvant and incomplete adjuvant (for booster) , at 3 or 4-week intervals. Other cows were treated with the same volume of seminal preparation without inhibin activity as procedures mentioned above to serve as a control (Group C). Artificial inseminations were given twice at 8 - 12 h intervals when the cow was in heat. Jugular venous blood samples were collected from each cow and used to assay the presence of antibody against seminal preparation by double-diffusion in agar precipitation test and to detect the titer of inhibin antibody by an ELISA method. Data from 247 cows showed that 83.9% (73/87) of cows were in estrus and ovulated 89 ova altogether, of which 19 cows ovulated twin ova and 15 cows produced twins in Group Th (n = 87). However, only 61.1% (44/72) of cows in Group TI (n = 72) and 62.5% (55/88) of cows in Group C were in estrus and ovulated 46 and 52 ova altogether respectively. The ovulation rate (1.27 ± 0.03), calving rate (126.3%) and twinning rate (26.3%) in Group Th were greater than those in Groups Tl or C (Panimals (r = 0.7507, P<0.01). These results indicate that active immunization of postpartum cows against inhibin purified from porcine seminal plasma may increase the ovulation rate and induce twinning, suggesting the potential to develop a method to improve fertility in cows.

AIM:To study the effects of florfenicol on early cytokine response and survival in treatment of infected murine in- duced by septic shock. METHODS:The effect of florfenicol on cytokine produc- tion from lipopolysaccharide(LPS)-stimulated macrophag- es was investigated by using a mouse macrophage cell line. The influence of florfenicol on the survival rate of mice in- jected with lethal dose of endotoxin and early cytokine re- sponses in endotoxemia mice were *** serum cytokine productions of mice were determined at different time points. RESULTS:Florfenicol significantly reduced the produc- tions of tumor necrosis factor(TNF)and interleukin-6 (IL-6)in the supernatant of cells as well as slightly atten- uated the secretions of interleukin-1β(IL-1β)and interleu- kin IL-10(IL-10).No significant difference in IL-1β con- centration was observed between florfenicol-treated group and control ***,florfenicol significantly in- creased serum IL-10 concentration of ***,it was found that florfenicol(at the dose of 100 mg/kg)signifi- cantly increased the survival rate of mice compared to the control group after mice were administrated LPS. CONCLUSION:These data indicate that florfenicol can improve survival rate of endotoxemia mice via modulating early cytokine responses and antibiotics mighty be of po- tential usefulness in treatment of bacterial shock.

The 1074-bp phyCs gene （optimized phyC gene） encoding neutral phytase was designed andsynthesized according to the methylotrophic yeast Pichia pastoris codon usage bias without altering theprotein *** expression vector,pP9K-phyCs,was linearized and transformed in *** of total extracellular phytase activity was 17.6 U/ml induced in Buffered Methanol-complex Medium（BMMY） and 18.5 U/ml in Wheat Bran Extract Induction （WBEI） medium at the flask scale,respectively,improving over 90 folds compared with the wild-type *** enzyme showed temperature optimumof 70℃ and pH optimum of *** enzyme activity retained 97% of the relative activity afterincubation at 80℃ for 5 *** of the heavy glycosylation the expressed phytase had a molecularsize of approximately 51 *** deglycosylation by endoglycosylase H （EndoH_f）,the enzyme had anapparent molecular size of 42 *** property and thermostability was affected by the glycosylation.

The influence of external noise on intracellular Ca2+ oscillations dynamics in a single cell of rat hepatocytes is *** coherence of noise-induced intracellular Ca2+ oscillations reached two maxima for multiple external noise intensity,indicating the occurrence of external noise stochastic bi-resonance (SBR).This phenomenon is compared with our previous result that was induced by internal ***,the deterministic bifurcation character of the system was also discussed.

Study on the Pathogenicity of Chinese Strains of Subgroup J Avian Leukosis Viruses

The pathogenicity of 4 Chinese strains of subgroup J avian leukosis viruses (ALV-J), SD9901, SD9902, YZ9901 and YZ9902,was studied. The results showed that only SD9902 among the 4 strains induced mortality from myeloid leukosis (ML). In the 12 meat-type chickens inoculated with SD9902 at 1-day-old, 9 died between 22 days and 38 days after inoculation. No death or ML was found in chickens inoculated with the other 3 strains during the period of 6 months. These results suggested the SD9902 strain of ALV-J was an acute transforming virus, but SD9901, YZ9901 and YZ9902 were non-transforming viruses. All 4 Chinese strains did not induce any tumors in egg-type SPF chickens during 7 months after hatching when viruses were injected into 11-day-old embryos.
