Rational redesign of inhibitors of furin/kexin processing proteases by electrostatic mutations

AIM: To model the three-dimensional structure and investigatethe interaction mechanism ofthe proprotein convertase furin/kexin and their inhibitors (eglin c mutants). METHODS: The three-dimensional complex structures of furin/ kexin with its inhibitors, eglin c mutants, were generated by modeller program using the newly published X-ray crystallographical structures of mouse furin and yeast kexin as templates. The electrostatic interaction energy of each complex was calculated and the results were compared with the experimentally determined inhibition con- stants to find the correlation between them. RESULTS: High quality models of furin/kexin-eglin c mutants were obtained and used for calculation of the electrostatic interaction energies between the proteases and their inhibitors. The calculated electrostaticenergies of interaction showeda linear correlation to the experimental inhibition constants. CONCLUSION: The modeled structures give good explanations of the specificity of eglin c mutants to furin/ kexin. The electrostatic interactions play important roles in inhibitory activity of eglin c mutants to furin/kexin. The results presented here provided quantitative structural and functional information concerning the role of the charge- charge interactions in the binding of furin/kexin and their inhibitors.

Global convergence property of s-dependent GFR conjugate gradient method

An \%S\%_dependent GFR method and a generalized conjugate gradient method are designed. Two estimated values of upper bounds for the ratio of β k to β FR k are given. The two convergence theorems are proved. The global convergent properties of \%s\%_dependent GFR method are obtained adopting several choice strategies of steplength and some results in recent literature are extended.

Simulations for optimization of SSRF BPM

The wake field, impedance and output signal for SSRF BPM have been calculated and analyzed by us-ing the numerical simulation code of MAFIA. The narrow-band impedance of BPM arising from formation of reso-nance in its cavity like structure either is harmful to beam or limits the performance of BPM itself, and should be re-duced to tolerable levels. The calculated results show that there are three main peaks in the impedance spectrum ofthe SSRF BPM prototype, and two of which are above the limit. After lots of simulations for different shapes of theBPM button, a new structure of SSRF BPM have been found, and there is only one main peak, which is under thelimit, in the impedance spectrum. Its output signal also meets the requirement.

Steady-state responses of axially accelerating viscoelastic beams: Approximate analysis and numerical confirmation

Nonlinear parametric vibration of axially accelerating viscoelastic beams is inves-tigated via an approximate analytical method with numerical confirmations. Based on nonlinear models of a finite-small-stretching slender beam moving at a speed with a periodic fluctuation, a solvability condition is established via the method of multiple scales for subharmonic resonance. Therefore, the amplitudes of steady-state periodic responses and their existence conditions are derived. The amplitudes of stable steady-state responses increase with the amplitude of the axial speed fluctuation, and decrease with the viscosity coefficient and the nonlinear coefficient. The minimum of the detuning parameter which causes the existence of a stable steady-state periodic response decreases with the amplitude of the axial speed fluctuation, and increases with the viscosity coefficient. Nu-merical solutions are sought via the finite difference scheme for a nonlinear par-tial-differential equation and a nonlinear integro-partial-differential equation. The calculation results qualitatively confirm the effects of the related parameters pre-dicted by the approximate analysis on the amplitude and the existence condition of the stable steady-state periodic responses. Quantitative comparisons demonstrate that the approximate analysis results have rather high precision.

Undoped LaAlO3 and 1 at.%Ce:LaAlO3 single crystals were grown by the Czochralski *** and fluorescence spectra were measured at room *** energy levels structure of Ce:LaAlO3 was *** this paper,two viewpoints were *** first one is:the energy levels structure of Ce:LaAlO3 is very similar to that of Ce:Lu2.SiO4)O which is a well-known *** the energy levels structure of Ce:LaAlO3 and Ce:Lu2.SiO4)O,the lowest 5d energy level of Ce 3+ is located below the bottom of the conduction band of host crystal and the other higher 5d energy levels of Ce 3+ are located above the bottom of the conduction band of host *** second one is:Ce:LaAlO3 single crystal may not be suitable for scintillation application;by comparing the energy levels structures of Ce:LaAlO3 and Ce:Lu2.SiO4)O,the large energy difference(1.13 eV)between the two lowest 5d energy levels of Ce 3+ in LaAlO3 is a crucial factor that causes the luminescence quenching.

Jingdezhen blue and white imperial porcelain specimens from the Yuan, Ming and Qing dynasties have been systematically analyzed using a nondestructive test method—— energy dispersive X-ray fluorescence analysis (EDXRF). The derived data of major and trace element compositions have been treated by correspondence analysis. The variation laws of the composition patterns for Jingdezhen blue and white imperial porcelain in different historical periods owing to the change in raw materials, recipe and technology have been discussed, and a time model related to variation of element composition has been preliminarily established, It would be helpful for scientific dating of Jingdezhen blue and white imperial porcelain, and even for the studies on the whole field of identification of ancient ceramics.

Experimental research on the residual magnetization of a rare-earth permanent magnet for a cryogenic undulator

The magnetic properties of commercial polycrystalline Nd2.e14B （N50M,N45H,N40SH,N35EH） and Sm2.o17（XG30/2.,XG2./2.,XG2./2.） magnets at cryogenic temperatures were tested by using a comprehensive physical properties measurement system （PPMS）. The results show that the spin tilt temperature Tst of Nd2.e14B magnets is closely related to intrinsic coercivity Hci , N50M and N45H with smaller Hci show a residual magnetization jump at 2.5K and 2.5K, respectively. For Sm2.o17 magnets, in 50-300 K, with temperature decreasing, residual magnetization Mrc shows a nearly linear increase, while in 10-50 K, Mrc has little change. The research results provide a reference for cryogenic undulators and other high-precision cryogenic devices.

This paper reports the development of solar-blind aluminum gallium nitride (AlGaN) 12.×12. UV Focal Plane Arrays (FPAs). The back-illuminated hybrid FPA archi- tecture consists of an 12.x12. back-illuminated AlGaN PIN detector array that is bump-mounted to a matching 12.x12. silicon CMOS readout integrated circuit (ROIC) chip. The 12.×12. p-i-n photodiode arrays with cuton and cutoff wave-lengths of 2.3 and 2.8 nm, with a sharp reduction in response to UVB (2.0―32. nm) light. Several examples of solar-blind images are provided. This solar-blind band FPA has much better application prospect.

Synthesis of O-(2.[^(18)F]fluoroethyl)-L-tyrosine and its biological evaluation in B16 melanoma-bearing mice as PET tracer for tumor imaging

O-(2.[18F]fluoroethyl) -L-tyrosine([18F]FET) ,a fluorine-18 labeled analogue of tyrosine,has been syn-thesized and biologically evaluated in tumor-bearing mice. The whole synthesis procedure is com-pleted within 50 min. The radiochemical yield is about 40%(no decay corrected) and radiochemical purity more than 97% after simplified solid phase extraction. [18F]FET shows rapid,high uptake and long retention in the tumor as well as low uptake in the brain. The ratios of tumor-to-muscle(T/M) and tumor-to-blood(T/B) of [18F]FET are similar to those of [18F]FDG,but the ratios of tumor-to-brain(T/Br) are 2.3 times higher than that of [18F]FDG. Autoradiography of [18F]FET demonstrates a remarkable accumulation in melanoma with high contrast. It appears to be a probable competitive candidate for melanoma imaging with PET.
