Objective Secreted proteins of Helicobacter pylori （H. pylori） interact with gastric epithelium cells and may contribute to cell damage. Considering the fact that HPO17S and hypothetical conserved protein HP12.6 are included in the group and that HP0175 is a well-known apoptosis-induced factor, the present study aims to clarify whether HP12.6 plays a role in bacterial pathogenicity or even functions as an apoptosis-induced factor in human stomach. Methods Two genes encoding HP12.6 and HPO175 were cloned into pET32. vector and expressed as recombinant proteins in Escherichia coil （E. coil） BL2.. Signal peptide sequences were not included. The recombinant proteins were purified with His SpinTrap and desalted by using HiTrap Desalting. Immunoreactivity of the proteins was determined by Western blot. Human gastric epithelial cell AGS was challenged with these endotoxin-free proteins; and apoptosis of cells was assayed with the Cell Death ELISA kit. Results Recombinant proteins and their respective products whose N-terminal his-tag were removed with thrombin were recognized by serum from the patient infected with H. pylori. Apoptotic AGS cells challenged by HP12.6 of H. pylori 2.695 were four times more than untreated cells. In addition, apoptosis-induced ability of HP12.6 of SS1 was not as strong as that of H. pylori 2.695 strain. Conclusion HP12.6 of H. pylori 2.695 induces apoptosis of AGS cells in a time-dependent manner, however the apoptosis-induced ability of HP12.6 may differ due to variations between different strains.

DNA polymerase delta is encoded by the POLD1 gene,the transcription of which is strictly cell ***,the means by which POLD1 transcription is regulated by the cell cycle mechanism is currently *** discovered a novel element in the POLD1 promoter known as a CDE(cell cycle-dependent element)/CHR(cell cycle gene homology region) element.A series of luciferase reporter constructs containing various POLD1 promoter mutations were used to investigate the role of the CDE/CHR element in POLD1 *** the CDE/CHR element was mutated,the promoter activity was up-regulated,and the cell-cycle related factors E2.1 and p2. stopped regulating the ***,cell cycle-dependent changes in the promoter activity required the integrative CDE/CHR *** mobility shift assay(EMSA) revealed the presence of at least three types of DNA/protein complexes binding to the CDE/CHR *** findings provide strong evidence that the CDE/CHR-like sequence is an active functional element in the POLD1 promoter,which is important for the cell cycle regulation of the POLD1 gene.

Highly pathogenic influenza A (H5N1) virus causes a widespread poultry deaths worldwide. The first human H5N1 infected case was reported in Hong Kong Special Administrative Region of china in 1997. Since then, the virus re-emerged in 2.03 and continues to infect people worldwide. Currently, over 400 human infections have been reported in more than 15 countries and mortality rate is greater than 60%. H5N1 viruses still pose a potential pandemic threat in the future because of the continuing global spread and evolution. Here, we summarize the epidemiological, clinical and virological characteristics of human H5N1 infection in china monitored and identified by our national surveillance systems.

One of the physiological functions of cellular prion protein(PrP C )is believed to work as a cellular resistance to oxidative stress,in which the octarepeats region within PrP plays an important ***,the detailed mechanism is less *** this study,the expressing plasmids of wild-type PrP (PrP-PG5)and various PrP mutants containing 0(PrP-PG0),9(PrP-PG9)and 12.PrP-PG12.octarepeats were generated and PrP proteins were expressed both in *** and in mammalian *** aggregation and formation of carbonyl groups were clearly seen in the recombinant PrPs expressed from *** after treatment of *** and trypan blue staining assays revealed that the cells expressing the mutated PrPs within octarepeats are less viable than the cells expressing wild-type *** significant high levels of intracellular free radicals and low levels of glutathione peroxidase were observed in the cells transfected with plasmids containing deleted or inserted *** more productions of carbonyl groups were detected in the cells expressing PrPs with deleted and inserted octarepeats after exposing to ***,cells expressing wild-type PrP showed stronger resistant activity to the challenge of H2.2.at certain extent than the mutated PrPs and mock. Avian influenza virus（AIV） nonstructural 1（NS1） gene was amplified by real-time polymerse chain reac tion（RT-PCR） and inserted into pET2.a, then transformed into E. coli BL2.（DE3） competent cell. With the induction of isopropyl-β-D-thiogalactoside（IPTG） and the purification of Ni-NTA column, we finally obtained purified NS1 protein. T7-phage display system was used to screen the proteins that interacted with NS1 from lung cell cDNA li brary. The selected positive clones were identified by DNA sequencing and analyzed by BLAST program in Gene Bank. Two proteins were obtained as NS1 binding proteins, Homo sapiens nucleolar and coiled-body phosphoprotein 1（NOLC1） and Homo sapiens similar to colon cancer-associated antigen. By co-immunoprecipitation and other me thods, Homo sapiens NOLC1 was found to interact with the NS1 protein, the results would provide the basis for fur ther studying biological function of NS1 protein.

Meta-analysis of the relationship between Epstein-Barr virus infection and clinicopathological features of patients with gastric carcinoma

Epstein-Barr virus (EBV) infection has been causally associated with occurrence of many malignant neoplasms. EBV-encoded small RNAs (EBERs) have been detected from about 10% of gastric carcinoma tissue cells, suggesting that EBV infection is associated with the development of gastric carcinoma. The present study pooled the data from the papers concerning EBV-related gastric cancers and performed a meta-analysis of 2. research papers. Among these papers, a total of 5475 cases with gastric cancer were enrolled, of whom 411 cases were found EBV-positive, with the EBV-positive rate being 7.5%. Among the EBV-positive gastric cancer cases, the detection rate was 11.1% in males and 3.0% in females. Compared with EBV-negative gastric cancer, EBV-positive gastric cancer had less lymph node metastasis. Based on the histological typing, of the EBV-positive gastric cancers, the diffuse type was 8.1%, and intestinal type was 8.0%. The examined specimen types included stored paraffin blocks and fresh surgically removed specimens, their EBV positive rates were 7.9% and 6.5% respectively. In terms of geographical distribution, the detection rate of EBV-positive gastric cancer was 9.4% in America, 6.1% in Asia and 9.1% in Europe. Meta-analysis showed that EBV infection occurred only in gastric cancer tissue cells and was significantly associated with the patients’ gender, lymph node metastases, and the location where tumor tissue generated and geographical distribution (P0.05). These results suggested that EBV-positive gastric cancer has distinct clinicopathological features.
