Genes present in virulent bacterial strains but absent in avirulent close relatives can be of great biologic and clinical interest. This project aimed to identify strain specific DNA sequences of Leptospira interrogans serovar lai, which is absent in the saprophytic L. biflexa serovar monvalerio, via suppression subtractive hybridization with the former as the tester while the latter as the driver. The mixture of PCR amplified DNA fragments from two subtractive hybridization experiments were cloned into pMD18-T vector and the positive clones were identified by dot blotting against the chromosome DNA of the two strains individually. After DNA sequencing and analysis, the distribution of these genomic fragment sequences in a panel of pathogenic and nonpathogenic leptospires was investigated employing dot blot analysis. Among the 188 positive clones randomly chosen, 2. contained the tester strain specific genomic regions, of which, 5 were non-coding fragments while the others contained 2. distinct protein coding sequences. Besides 9 genes encoding functional proteins, 12.genes encode unknown proteins and the rest two genes encode proteins with recognizable domain structures, one for a putative leucine-rich repeats (LRR) family protein while the other as an outer-membrane protein. Our experiment results indicated that suppression subtractive hybridiza- tion is effective for screening specific DNA sequences between two leptospiral strains, and some of these sequences might be responsible for virulence determination. Further analysis of these DNA sequences will provide important information on the pathogenesis of Leptospira.

This paper reports the study of ancient potteries unearthed from the Guangfulin site in shanghai. From the external features of these potteries, archeologists can classify them into two different cultural types: Liangzhu culture (local culture) and a new culture which might be derived elsewhere, but they can not recognize whether they were produced in the same place. Proton induced X-ray emission (PIXE) and instrumental neutron activation analysis (INAA) were used to measure the chemical compositions of the samples and the results were analyzed using multivariate statistics. We found that the two methods can complement each other and their database can be mutually referenced in the study of ancient potteries. Experimental results show that the compositions of the potteries from the two cultural types are different, which means that the materials for ancient potteries of new culture originate from elsewhere. This supports the archaeologists’ speculation that a group of ancient people migrated to shanghai from some other places 4000 years ago.

The geocentric coordinates of three mobile sites including Fangshan in Beijing, Urumqi in the Xinjiang Uy-gur Autonomous Region and Lhasa in Tibet are determinedin the ITRF2.00 frame by means of both the global SLRdata and the mobile (SLR) data obtained by a mobile satel-lite laser ranging (SLR) apparatus from Aug. 2.00 to Dec.2.01. The results showed that the geometrical performance of chinese SLR network has been significantly improved.

Based on the similarity theory,a scale effect model of the spraying water cooling system of shanghai expo Theme Pavilion's roof was set *** to the typical weather conditions in shanghai city,different models were analyzed on solar radiation,and different heat transfer amount was *** the following conclusions could be made:At the sunny day of summer in shanghai,the temperature of inner roof with sprinkling system descended about 3 ℃,and the air conditioning load of the whole Theme Pavilion descend more than 32. kW.

Effects of A1 Additions on Microstructure and Mechanical Properties of MoSi_2

The effects of Al additions on the microstructure and mechanical properties of MoSi2.have been studied. With the A1 additions from 2.5 wt pct to 5 wt pct, the siliceous grain boundary phase in hot pressed samples was eliminated because of the formation of Al2.3 particles. It was shown that Al and SiO2.reacted at 860℃. During the reaction, A1 atoms were mainly transferred to Al2.3 particles, and to some extent, diffused into MoSi2.grains. Both the toughness and strength of Al containing composites exceeded those of pure MoSi2.material. Bending strength and fracture toughness reach the highest value of 350 MPa, 4.05 MPa.m1/2. respectively, at ambient temperature when Al addition was of 3.5 wt pct.

Expression of Bcl-2.inhibited Fas-mediated apoptosis in human hepatocellular carcinoma BEL-7404 cells

Apoptosis plays an important role in embryonic development, tissue remodeling, immune regulation and tumor regression. Two groups of molecules (Bcl-2.family and 'Death factor' family) are involved in regulating apoptosis. In order to know about the effect of Bcl-2.on apoptosis induced by Fas, a typical member of 'Death factor' family, the transfection experiments with expression vectors pcDNA3-fl and pcDNA3-bcl-2.were performed in BEL-7404 cells, a human hepatocellular carcinoma cell line which expresses endogenous Fas, but not FasL and Bcl-2. The data showed that the expression of FasL in pcDNA3-fl transfected hepatoma cells obviously induced the apoptosis of the cells. However, the overexpression of Bcl-2.in pcDNA3-bcl-2.transfected 7404/b-16 cells counteracted pcDNA3-fl transient transfection mediated apoptosis. Further study by cotransfection experiments indicated that Bid but not Bax (both were pro-apoptotic proteins of Bcl-2.family) blocked the inhibitory effect of Bcl-2.on Fas-mediated apoptosis. These results suggested that Fas-mediated apoptosis in human hepatoma cells is possibly regulated by Bcl-2.family proteins via mitochondria pathway.

VMIB (virtual multi-dimensional internal bonds) is a multiscale mechanical model developed from the VIB (virtual internal bond) theory. In VIB theory,the solid mate-rial is considered to consist of random-distributed material particles in microscale. These particles are connected with normal bonds. The macro constitutive relation is derived from the cohesive law between particles. However,in VMIB,the micro particles are connected with both normal and shear bonds. The macro constitutive relation is derived in terms of bond stiffness coefficients. It has been theoretically certified that there exists a corresponding relationship between the two bond stiffness coefficients and the two macro material constants,i.e. the Young’s modulus and Poisson ratio. This corresponding relationship suggests that it should be necessary and sufficient to simultaneously account for the normal and shear interactions between particles. Due to the fact that the fracture criterion is directly incorporated into the constitutive relation,both VIB and VMIB present many advantages in simulating fractures of materials. In the damage model of rock mass,a damage tensor is usually defined to describe the distribution of cracks. The damage value in one direction determines the relative stiffness of rock mass in this direction. In VMIB solid,the relative distribution density of micro bonds in one direction determines the relative macro stiffness of the material in this direction. The effects of the damage value and the relative distribution density of bonds are consistent. To simulate the failure behavior of rock mass with VMIB,the presented paper sets up a quantitative relationship between the damage tensor and the rela-tive distribution density of bonds. Comparison of the theoretical and the experi-mental results shows that VMIB model can represent the effect of distributed cracks on rock mass with this relationship.
