塞来考昔对女性卵巢功能的影响

目的 :观察塞来考昔 (celecoxib)对女性卵巢功能的影响。方法 :5 6例痛经病人分为 2. ,A组给予塞来考昔治疗 1wk ,B组给予山莨菪碱治疗1wk ,采用对照的方法进行临床试验。结果 :塞来考昔对治疗前后病人性激素 (PRL ,E2.,LH ,FSH)水平无明显影响 (P >0 .0 5 ) ,与山莨菪碱治疗组比较无显著差异 (P >0 .0 5 ) ,其对滤泡直径的影响与山莨菪碱治疗组比较亦无显著差异 [(2.0 .1±s 1.3)mmvs(19.4± 1.1)mm ,P >0 .0 5 ]。结论 :短期使用塞来考昔对女性卵巢功能可能无明显影响。

BACKGROUND: Tea polyphenols have been shown to protect against carbon tetrachloride ( CCl4) -induced liver injury, liver fibrosis, hepatic ischemia-reperfusion injury. In this study, we examined the effect of tea polyphenols on lipopolysaccharide ( LPS ) -induced liver injury, and explored its mechanisms. METHODS: Sprague-Dawley rats received tea polyphenols (100 mg · kg-1·d-1) or vehicle (water) intragastrically by gavage for 14 days, followed by LPS (5 mg/kg) or saline injection intraperitoneally. Liver injury was assessed by biochemical assay and pathological analysis. Serum tumor necrosis factor-α (TNF-α) levels and liver malondialdehyde (MOA) contents were determined. Inducible nitric oxide synthase (iNOS) protein and TNF-α, iNOS and en-dothelial nitric oxide synthase (eNOS) mRNA expressions in the liver were detected by immunohistochemistry and reverse transcriptase-polymerase chain reaction (RT-PCR), respectively. RESULTS: Administration of LPS resulted in liver injury in rats, evidenced by elevated activities of serum alanine aminotransferase (ALT) and aspartate aminotransferase (AST), hepatocellular necrosis, and neutrophil infiltration in the liver. These responses were associated with increased serum TNF-α levels, induced iNOS protein, expressions of TNF-α, iNOS mRNA in the liver and elevated lipid peroxidation at 90 minutes or 6 hours after LPS injection. Pretreatment with tea polyphenols attenuated LPS-induced liver injury, and blunted the rises of serum TNF-α levels and lipid peroxidation and the induction of expressions of TNF-α, iNOS in the liver. CONCLUSION: Tea polyphenols prevent LPS-induced liver injury, and the mechanisms may involve the reduction of serum TNF-α levels and lipid peroxidation and the suppression of TNF-α, iNOS expressions in the liver.

Objective： The aim of this study was to evaluate the recent efficacy and adverse reactions of paclitaxel liposome plus nedaplatin in the treatment of advanced non-small cell lung cancer （NSCLC）. Methods： Seventeen cases of NSCLC treated with paclitaxel liposome and nedaplatin for 2.to 6 cycles, by infusing paclitaxel liposome 135 mg/m^2.for 3 h on d 1 and nedaplatin 80 mg/m^2.as infusion on d2. Results： Among 17 patients being evaluated for response to treatment, 1 achieved complete response （CR）, 4 achieved partial response （PR）, 3 achieved stable disease （SD）, 9 achieved progress disease （PD）. The main adverse reaction was haematological toxicities, especially leukopenia and thrombocytopenia. The non-hae- matological toxicities included nausea, vomiting, mild hepatic dysfunction, alopecia and so on. Conclusion： Paclitaxel lipo- some plus nedaplatin was effective and well tolerated for treating patients with advanced NSCLCo

Preventive effect of hydrotalcite on gastric mucosal injury in rats induced by taurocholate

Objective:To investigate the effect of ginsenoside Rb1 preconditioning on endothelial nitric oxide synthase (eNOS) and nitric oxide (NO) expression levels during myocardial ischemia and reperfusion in diabetic rats and establish a possible molecular mechanism. Methods:Male adult SD rats weighing 2.0～2.0g were used for this study. Type I diabetes mellitus was induced by intraperitoneal injection of streptozotocin (65mg·kg-1) and this was confirmed by fasting blood glucose ≥16.7mmol·L-1 (300mg·L-1). The animals were randomly divided into 5 groups:sham-operated (sham),ischemia/reperfusion (I/R),ginsenoside Rb1+I/R,L-NAME (a specific eNOS inhibitor) +I/R,and L-NAME+ginsenoside Rb1 group (L-NAME+Rb1). Ischemia/reperfusion was produced by occlusion of the anterior descending branch of left coronary artery (LAD) for 30 minutes and the occlusion of LAD was released to allow reperfusion for 12. minutes. In sham-operated group,LAD was exposed but not occluded. In groups L-NAME,L-NAME+Rb1 and L-NAME,10mg·kg-1 of L-NAME was injected into the femoral vein 2. minutes before initiation of occlusion. In groups Rb1 and L-NAME+Rb1,ginsenoside Rb1 40mg·kg-1 was administered intravenously vein 10 minutes before occlusion of LAD. In group sham-operated and group I/R,a mixture of balanced solution and polygeline (to the ratio of 3:1) was infused. Heart rate (HR) and mean arterial pressure (MAP) were recorded 10 minutes before ischemia (T1,baseline),after 15 minutes of ischemia(T2.,and on the 30th minute,60th minute,and 12.th minute (T3-T5) of reperfusion. The rate pressure product (RPP) was calculated as the product of the heart rate and the peak mean arterial pressure. The myocardial ischemia reperfusion injury was assessed by infarct size,plasma creatine kinase (CK),lactate dehydrogenase (LDH) and tissue histological changes. AIM: To study the preventive effect of hydrotalcite on gastric mucosal injury in rat induced by taurocholate, and to investigate the relationship between the protective mechanism of hydrotalcite and the expression of trefoil factor family 2.(TFF2. mRNA and c-fos ***: Forty five male Wistar rats were randomly divided into hydrotalcite group, ranitidine group and control group. Gastric mucosal injury was induced by introgastric acidified taurocholate. OD value of TFF2.mRNA expression in gastric mucous cells was determined by hybridization and computer image analysis system. OD value of c-fos protein expression in gastric mucous cells was measured by immunohistochemistry and computer image analysis ***: The gross mucosal injury index in hydrotalcite group was significantly lower than that in ranitidine group and control group (8.60±2.2. vs 16.32.4.2., 2..53±5.39;P＜0.05, P＜0.01). The expression level of TFF2.mRNA in hydrotalcite group was markedly higher than that in ranitidine group and control group (0.56±0.09 vs 0.30±0.05, 0.2.±0.03,P＜0.05). The OD value of c-fos protein in hydrotalcite group was higher than that in ranitidine group and control group (0.52.0.07 vs 0.31±0.04, 0.32.0.05, P＜0.05).CONCLUSION: Hydrotalcite can protect gastric mucosal injury in rats induced by taurocholate, which may be related to the increased expression of TFF2.and c-fos protein.

Aim:To investigate the antioxidant properties of berberine (Ber) on corpuscavenosum smooth muscle cells (CCSMC) in penile erectile ***:We examined the effects of Ber on cultured rabbit CCSMC damaged by hydrogenperoxide (H2.2. through examining cell viability by methyl thiazolyl tetrazoliumassay and assessing the level of malondialdehyde (MDA),superoxide dismutase,(SOD) activity,nitric oxide (NO) products,and lactate dehydrogenase (LDH) re-lease in cells after stimulation with ***:Treatment with 1 mmol/L H2.2.ignificantly decreased the cell viability,NO products,and SOD activity of CCSMCfrom 100% to 48.57%±4.1% (P2.1 μmol/L (P2.6 U/mL (P2.9nmol/mg protein (P2.2.withincreased cell viability (P<0.05 or P<0.01),NO production (P<0.01),and SODactivity (P<0.01) and decreased LDH release and MDA content (both P<0.01).Conclusion:Bet could produce its antioxidant action on oxidative stress-inducedcultured *** effects may be of benefit in the prevention of penileerectile dysfunction.
