In this study,freeze-fractured specimens of Allium ceparoot tip meristems were examined under the scanning elec-tron microscope(SEM).This technique permitted the visu-alization of the outer membrane of the nuclear envelope withnuclear pore complexes and *** of the cellnuclei prepared witn this procedure had fissures of variouswidths no their nuclear envelopes through which the nuclearlamina-like filaments(LLF)underneath the nucleoplasmicside of the envelopes were clearly *** diameters ofthese filaments varied between 2. and 12. *** of theLLFs showed granular thickenings at places,and were at-tached to the inner surface of nuclear envelope in some *** LLFs were also seen at the peripheries of thefreeze-fractured faces of ***,the spatial re-lation between the nuclear matrix filaments(NMF)and oth-er nuclear structures(nucleoli,chromatin and peripherallamina-like filaments)was revealed in these *** addition,the methods and techniques instudying the nuclear lamina morphology and the roles playedby NMFs in activities of various nuclear structures were dis-cussed in brief.

In this paper, the structural, electrical and optical properties of Al-doped zinc oxide (ZnO: Al) thin films onto quartz substrates prepared by electron beam evaporation technique were studied. All samples are polycrystal with a hexagonal structure andc-axis preferred orientation. The intense UV emissions and weak deep-level green emissions were observed. The films show high transparency, conductivity and high carrier concentration. Van der Pauw measurements show that the films are n-type degenerate semiconductor. The lowest resistivity is 6.7×10?4 Ω⊙cm at room temperature. These films exhibit a carrier density above 102. cm?3. The conduction mechanism was discussed. The behavior of metallic conduction can be observed in the high temperature range.

The alien gene flow between genetically modified glyphosate-resistant rapeseed variety Q3.(Brassica napus L.) and four cruciferous weeds was studied under mentor pollen inducement. The results showed that when Thlaspi arvense L., Capsella bursa-pastoris (L.) Medic, Cardamine hirsuta L. and Rorippa palustris (L.) Besser were pollinated with mentor pollen, the mixed Q3.and the weed, pollen grains aggregated largely and germinated quickly, and the numbers of pollen tubes penetrating into the style and the ovary were greatly increased as compared with corresponding self-pollination groups. Twenty four to forty eight hours after pollination, several pollen tubes were observed to penetrate into the ovule via micropyle in each mentor combination. However, when the mentor progenies were analyzed by PCR, all of them showed negative for the Q3.herbicide-resistant gene. Collectively, these results indicated that crossing between T. arvense, C. bursa-pastoris, C. hirsuta, R. palustris (as female) and Q3.(as male) was highly incompatible and the herbicide-resistant gene could not flow from Q3.to these four weeds.

To identify ecotypic variation of L. chinensis, the physiological characteristics and environmental factors were measured and assessed for grey-green and yellow-green ecotypes of L. chinensis grown in the salt-alkalized and aeolian sandy soils. Although both ecotypes presented a similar salt-alkaline tolerance under the experimental conditions, they differed in several aspects. The L. chinensis grown in the aeolian sandy soils had higher water potential and chlorophyll content and lower proline content in their leaves than those grown in the salt-alkalized habitat, and the grey-green ecotype of L. chinensis in both types of soils had higher water potential, chlorophyll and proline contents than the yellow-green ecotype. Besides, grey-green ecotypes had higher salt-alkaline tolerance than the yellow-green ecotype. The stepwise regression analysis showed that soil conductance and pH value were the main ecological factors affecting the physiology of L. chinensis. From the natural distribution of the two ecotypes, we considered that salt-alkaline in the soils is the leading factor in determining ecotypic variation of L. chinensis. The two ecotypes were first named grey-green and yellow-green ecotypes respectively.

hELP3.Subunit of the Elongator Complex Regulates the Transcription of HSP70 Gene in Human Cells

The human Elongator complex is remarkably similar to its yeast counterpart in several aspects. In a previous study, we analyzed the functions of the human elongation protein 3.(hELP3. subunit of the human Elongator by using an in vivo yeast complementation system. However, direct evidence for hELP3.functions in regulating gene expression in human cells was not obtained. In this study, we used hELP3.antisense oligonucleotide inhibitors to knock down hELP3.gene expression to investigate its function in human 2.3. cells. The results showed that specific reduction of hELP3.mRNA and protein caused a significant suppression of HSP70-2.gene expression, and this was accompanied by histone H3.hypoacetylation and decreased RNA polymerase Ⅱ density at the HSP70-2.gene. Moreover, the data also showed that hELP3.exerted the transcriptional regulatory function directly through its presence on the HSP70-2.gene. Data presented in this report provide further insight and direct evidence of the functions of hELP3.in HSP70-2.gene transcriptional elongation in human cells.
