N-glycosylation proteome of endoplasmic reticulum in mouse liver by ConA affinity chromatography coupled with LTQ-FT mass spectrometry

Glycosylation is the most versatile and one of the most significant protein post-translational modifications. It is generally classified into three categories according to the amino acid to which the glycan is attached: N-glycosylation, O-glycosylation and C-glycosylation. Synthesis of N-glycoproteins occurs in the rough endoplasmic reticulum (rER), and all N-glycoproteins synthesized in rER have uniform glycan endings with mannose (Man) and glucose (Glc). A systematic strategy was developed to comprehensively profile N-glycoproteins in complex biological samples. The lectin Concanvalin A (ConA), which has a high affinity for glycans ending with Man, was used to extract the N-glycoproteins synthesized or located in the rER, and identified the N-glycoproteins and their glycosylation sites by LTQ-FT MS. The analysis was repeated three times at both the biological sample and mass spectrometry levels. In total, 3.0} glycosylation sites on 2.2.N-glycoproteins were identified in the mouse liver. Of these, 13. were known N-glycoproteins in the Swissprot database. The identified N-glycoproteins were classified according to their cell location by the Swissprot database and GO software. The identified N-glycoproteins in this study would serve as a good complement to the N-glycoprotein database for the mouse liver.

SHRIMP zircon U-Pb age and Nd isotopic study on the Nyainqêntanglha Group in Tibet

The Nyainqêntanglha Group is traditionally viewed as the oldest metamorphic basement in the Lhasa block, but its formation age and tectonic setting remain debate. Zircons extracted from the metamorphic sequence of volcanics and intrusions of the Nyainqêntanglha Group, 10 km west of Nam Co in northern Lhasa block, have been investigated by cathodolu- minescence (CL), backscattered (BSE) and dated by ion microprobe (SHRIMP). We conclude that the U-Pb age of 787±9 Ma of zircons from the trondhjemite imposes a constraint on maxi- mum protolith age, and minimum formation age of the Nyainqêntanglha Group is constrained by U-Pb age of 748±8 Ma of zircons from the granite. The formation age of the Nyainqêntanglha Group is consistent with sedimentary age of Greater Himalayan rocks, showing that they devel- oped coevally in an arc-basin tectonic setting of Neoproterozoic active continental margin along the northern margin of the India shield. The inherited zircons from the tholetiite and granite give older 2.7Pb/2.6Pb ages from 947 to 1766 Ma. The positive εNd( t ) value indicates that the mafic rocks were derived from the depleted mantle, but contaminated by the older continental crustal material. Integrated Nd model age and U-Pb age data provide excellent evidence for the exis- tence of Mesoproterozoic basement in the Lhasa block during Neoproterozoic time.

Establishment of liver specific glucokinase gene knockout mice: a new animal model for screening anti-diabetic drugs

AIM: To characterize the liver-specific role of glucokinase in maintaining glucose homeostasis and to create an animal model for diabetes. METHODS: We performed hepatocyte-specific gene knockout of glucokinase in mice using Cre-loxP gene targeting strategy. First, two directly repeated loxP sequences were inserted to flank the exon 9 and exon 10 of glucokinase in genomic DNA. To achieve this, linearized targeting vector was electroporated into ES cells. Then G418- and Gancyclovir-double-resistant clones were picked and screened by PCR analysis and the positives identified by PCR were confirmed by Southern blot. A targeted clone was selected for microinjection into C57BL/6J blastocysts and implanted into pseudopregnant FVB recipient. Chimeric mice and their offspring were analyzed by Southern blot. Then by intercrossing the Alb-Cre transgenic mice with mice containing a conditional gk allele, we obtained mice with liver-specific glucokinase gene knockout. RESULTS: Among 161 double resistant clones 4 were positive to PCR and Southern blot and only one was used for further experiments. Eventually we generated the liver specific glucokinase knockout mice. These mice showed increased glucose level with age and at the age of 6 weeks fasting blood glucose level was significantly higher than control and they also displayed impaired glucose tolerance. CONCLUSION: Our studies indicate that hepatic glucokinase plays an important role in glucose homeostasis and its deficiencies contribute to the development of diabetes. The liver glucokinase knock- out mouse is an ideal animal model for MODY2. and it also can be applied for screening anti-diabetic drugs.

Quantitative estimation of vegetation water content(VWC) using optical remote sensing techniques is helpful in forest fire as-sessment,agricultural drought monitoring and crop yield *** paper reviews the research advances of VWC retrieval using spectral reflectance,spectral water index and radiative transfer model(RTM) *** also evaluates the reli-ability of VWC estimation using spectral water index from the observation data and the *** on two main definitions of VWC-the fuel moisture content(FMC) and the equivalent water thickness(EWT),the retrieval accuracies of FMC and EWT using vegetation water indices are ***,the measured information and the dataset are used to estimate VWC,the results show there are significant correlations among three kinds of vegetation water indices(i.e.,WSI,NDⅡ,NDWI1640,WI/NDVI) and canopy FMC of winter wheat(n=45).Finally,the future development directions of VWC detection based on optical remote sensing techniques are also summarized.

The purpose of this paper is to discuss the poly-phase salt tectonics and its relation to the hydrocarbon accumulation of the Tarim superimposed basin. Several salt sequences are developed in the Tarim basin, they are: (1) the Mid-Early Cambrian salt sequence, mainly distributed in the west part of the north Tarim uplift and Keping uplift; (2. the Early Carboniferous salt sequence, mainly distributed in the south slope of the north Tarim uplift; (3. the Paleogene salt sequence, mainly distributed in the mid-west part of the Kuqa foreland fold belt and north Tarim uplift; and (4) the Miocene salt sequence, mainly distributed in the east part of the Kuqa foreland fold belt. The salt sequences deposited in the tectonically calm scenario, while the salt layers deformed during the period of intense tectonism. Although the salt sequences are characteristic of plastic flow, the differences of salt deformation styles exist in the different salt sequences be-cause of the different deformation mechanism. It is attractive that the distribution of the large oil-gas fields or pools has a bearing upon the salt sequences and salt structures, such as the Tahe oilfield related to the Lower Carboniferous salt sequence and laterally facies changed mudstone, the Kela No.2.gas field to the Paleogene salt structures, and the Dina gas field to the Miocene salt structures. It is indicated that the large-scale hydrocarbon accumulation is con-trolled by the poly-phase salt sequences and structures. The deep analysis of the poly-phase salt tectonics is helpful to understanding the characteristics of the structural deformation and oil-gas accumulation of the Tarim basin.

The aim of this work is to study the effect of Sr substitution on the redox properties and catalytic ac- tivity of La2.xSrxNiO4(x=0.0-1.2.for NO *** suggest that the x=0.6 sample shows the highest *** characterization(TPD,TPR,etc.)of samples indicates that the x=0.6 sample possesses suitable abilities in both oxidation and reduction,which facilitates the proceeding of oxygen desorption and NO *** temperature below 700℃,the oxygen desorption is difficult, and is the rate-determining step of NO *** the increase of reaction temperature(T> 700℃),the oxygen desorption is favorable and,the active adsorption of NO on the active site(NO+Vo+ Ni2.→NO^--Ni3.)turns out to be the rate-determining *** existence of oxygen vacancy is the prerequisite condition for NO decomposition,but its quantity does not relate much to the activity.

To reduce data variance caused by individual differences of different samples,a new experimental method is proposed by loading and unloading the axial forces with different frequencies and different waves of a certain load amplitude to the same rock sample without damaging *** time segments are defined and fractionized into segments I and Ⅱ.Criterions for seg-mentation,definitions of relevant parameters and empirical analysis are also offered *** the course of sinusoidal loading,the serious peak value dislocation of the displacement variation rate and the loading rate is defined as peak ***,the definition of the apparent tangent modulus is put forward and the linear relation between it and the vertical force in the frequency of 0.1,0.2.0.5 Hz sinusoidal loading segment is confirmed to be ever-present on the basis of the test *** the calculating formula of the deformation rate in non-lag time is *** is thus suggested that the deformation rate should be codetermined by the loading rate df/dt and instant load f(t),which well explains the peak dislocation of the time-variable curve peak value of dl/dt and deformation rate of df/*** the lag time derivation model is established and by comparing the calculated values with the measured ones,it is demonstrated that the above formula offers a better simulation of the sandstone deformation rate in the sinusoidal loading segment,with the load amplitude being 96 kN and the frequency ranging from 0.1 Hz to 0.5 Hz.

By using a simple barrier penetration approach,we predict the α-decay branching ratios to members of ground-state rotational band of heavy even-even No isotopes. We also extend our approach to calculate the α-decay branching ratios to the rotational band of heavy odd-A nuclei. The theoretical branching ratios of α-decays are found in good agreement with the available experimental data.

Shape phase transitions in nuclei:Effective order parameters and trajectories

We analyze systematically the effective order parameters in nuclear shape phase transition both in experiments and in the interacting boson model. We find that energy ratios and B(E2. ratios can distinguish the first from the second-order phase transition in theory above a certain boson number N (about 50), but in experiments, only those quantities, such as E(L1+)/E(02.) and B(E2. (L+2.1 → L1)/B(E2. 2. → 01), etc., of which the monotonous transitional behavior in the second-order phase transition is broken in the first order phase transition independent of N, are qualified as the effective order parameters. By implementing the originally proposed effective order parameters and the new ones, we find that the isotones with neutron number Nn = 62.are a trajectory of the second order phase transition. In addition, we predict that the transitional behavior of isomer shifts of Xe, Ba isotopes and Nn = 62.isotones is approximately monotonous due to the finiteness of nuclear system.

Management Communication in University’s Digital Library

Digital library is a process of exploiting more scientific methods to construct a social knowledge network. It represents an “extension” from a traditional library. The core and essence is to build a favorable surroundings for knowledge accumulation, knowledge transfer and knowledge communication. Management communication covers information transfer and organization cooperation. Compared with traditional library, the management communication in digital library shoulders the commitment to support knowledge innovation. On the basis of analysis of the connotation of digital library, this paper analysed the aspects of management communication, including communicators, communication styles, contents, channels and outer communication, all of which tries to construct an integrated model of management communication in a digital library and turns a library into an actual “knowledge fountain”.
