Urea transporters are a group of proteins that facilitate urea movement across biological membranes. Kidd blood group (JK antigen) and urea transporter of human erythrocytes are carried by the same protein UT-B. To investigate the molecular basis of the Jknull phenotype in the Chinese population, blood samples from Chinese individuals were screened using the 2.mol/L urea solution hemolysis test. Urea and water permeability of erythrocytes membrane was measured by stopped-flow light scattering. Genomic DNA was extracted from lymphocytes. UT-B gene of Jknull's family was analyzed using genomic PCR by primers designed to cover sequences of all exons and exon-intron boundaries in human UT-B gene. One Jknull subject was found from twenty thousa.d screened Chinese individuals, and it was confirmed that this individual did not express the erythrocyte urea transporter. Genomic sequence analysis of the Jknull individual showed that there were two point mutations, G→C, which is novel, and G→A, at the 3(-acceptor splice site (AG) of intron 5 of UT-B gene. Exon 6 is spliced out in the UT-B transcript due to either of these mutations. Water permeability in Jknull erythrocytes (Pf, ~0.00037 cm/s) was significantly lower than that in normal erythrocytes (Pf, ~0.00062.cm/s) after HgCl2.incubation, providing evidence for UT-B facilitated water transport in human erythrocytes.

Integrated geochemical and Sr-Nd-Pb isotopic studies of the Early Jurassic Nandaling flood basalts(NFB) in the Yanshan belt, northern margin of the North China Craton(NCC),are presented in this *** sub-alkaline basalts evolved from a more magnesium-rich parental magma through fractional crystallization of olivine and *** primitive magma of the NFB originated from 2.5%partial melting of spinel to garnet transitional peridotite at about 70-80 km depth in the Mesozoic lithosphere *** NFB contain a distinctive lithospheric component,characterized by Nb(Ta),Th,U and Ti depletions,LREE enrichments, moderate Sr,and low Nd and Pb initial isotopic ratios,as a result of an interaction between lower crust(15-2.%) and primitive magma evoked by magmatic underplating at crust-mantle *** Early Jurassic NFB extruded in an intraplate extensional setting related to post-orogenic collapse in the northern margin of the NCC,indicating an event of lithospheric modification earlier than that in the southern margin(Early Cretaceous).The temporal similarity of the Jurassic-Cretaceous mantle-derived mafic rocks to lower crust replacement,and the decoupling of surface shortening with lithospheric thinning during the Late Jurassic-Early Cretaceous, suggest the important role of magmatic underplating and subsequent crust-mantle interaction accompanied by asthenosphere upwelling on the evolution of the Mesozoic lithosphere of the *** correlation between lithospheric thinning and magmatic underplating may be an important process in continental rifting.

Tests for paired count outcomes

Paired t-tests are widely used to compare differences in paired continuous outcomes. This test, however, does not apply to non-continuous outcomes. For binary outcomes,McNemar's test is the de facto method for comparing differences in paired binary variables. For count or frequency outcomes, there is not much discussion in the literature. The Wilcoxon signed-rank test is often used to assess differences in paired count outcomes. However, like the Mann-Whitney U test, or Wilcoxon rank-sum test for comparing two independent groups, the Wilcoxon signedrank test is generally not efficient(powerful). In this essay,we consider some alternative methods for comparing paired count outcomes and compare them with the Wilcoxon signed-rank test using Monte Carlo simulations.

The pool of microbes inhabiting our body is known as .1c;microbiota.1d; and their collective genomes as .1c;microbiome.1d;. The colon is the most densely populated organ in the human body, although other parts, such as the skin, vaginal mucosa, or respiratory tract, also harbour specific microbiota. This microbial community regulates some important metabolic and physiological functions of the host, and drives the maturation of the immune system in early life, contributing to its homeostasis during life. Alterations of the intestinal microbiota can occur by changes in composition (dysbiosis), function, or microbiota-host interactions and they can be directly correlated with several diseases. The only disease in which a clear causa. role of a dysbiotic microbiota has been demonstrated is the case of Clostridium difficile infections. Nonetheless, alterations in composition and function of the microbiota have been associated with several gastrointestinal diseases (inflammatory bowel disease, colorectal cancer, or irritable bowel syndrome), as well as extra-intestinal pathologies, such as those affecting the liver, or the respiratory tract (e.g., allergy, bronchial asthma, and cystic fibrosis), among others. Species of Bifidobacterium genus are the normal inhabitants of a healthy human gut and alterations in number and composition of their populations is one of the most frequent features present in these diseases. The use of probiotics, including bifidobacteria strains, in preventive medicine to maintain a healthy intestinal function is well documented. Probiotics are also proposed as therapeutic agents for gastrointestinal disorders and other pathologies. The World Gastroenterology Organization recently published potential clinical applications for several probiotic formulations, in which species of lactobacilli are predominant. In an effort to study the systematic affinities and species- level phylogenetic relationships of the enigmatic anurans vadably assigned to the genera Ingerana or Limnonectes （family Dicroglossidae）, we collected new molecular sequence data for five species including four Himalayan taxa, Limnonectes xizangensis, Lim. medogensis, Lim. alpine, Ingerana borealis and one southeast Asian species, I. tasanae, and analyzed these together with data from previous studies involving other ostensibly related taxa. Our surprising results demonstrate unequivocally that Lim. xizangensis, Lim. medogensis and Lira. alpine form a strongly supported clade, the sister-group of the family Australasian forest frog family Ceratobatrachidae. This discovery requires an expansion of the definition of Ceratobatrachidae and represents the first record of this family in China. These three species are distinguished from the species of Ingerana and Limnonectes by the：（1） absence of interdigital webbing of the foot, （2. absence of terminal discs on fingers and toes, （3） absence of circumarginal grooves on the fingers and toes, and （4） absence of tarsal folds. Given their phylogenetic and morphological distinctiveness, we assign them to the oldest available generic name for this clade, Liurana Dubois 1987, and transfer Liurana from Dicroglossidae to the family Ceratobatrachidae. In contrast, Ingerana tasanae was found to be clustered with strong support with the recently described genus Alcalus （Ceratobatrachidae）, a small clade of otherwise Sundaic species; this constitutes a new record of the family Ceratobatrachidae for Myanmar and Thailand. Finally, Ingerana borealis clustered with the ＂true＂ Ingerana （family Dicroglossidae）, for which the type species is L tenasserimensis.

In Arabidopsis,photoperiodic flowering is controlled by the regulatory hub gene CONSTANS(CO),whereas floral organ senescence is regulated by the jasmonates(JAs).Because these processes are chronologically ordered,it remains unknown whether there are common regulators of both *** this study,we discovered that CO protein accumulates in Arabidopsis flowers after floral induction,and it displays a diurnal pattern in floral organs different from that in the *** observed that altered CO expression could affect flower senescence and abscission by interfering with JA response,as shown by petal-specific transcriptomic analysis as well as CO overexpression in JA synthesis and signaling *** found that CO has a ZIM(ZINC-FINGER INFLORESCENCE MERISTEM)like domain that mediates its interaction with the JA response repressor JAZ3(jasmonate ZIM-domain 3).Their interaction inhibits the repressor activity of JAZ3,resulting in activation of downstream transcription factors involved in promoting flower ***,we showed that CO,JAZ3,and the E3 ubiquitin ligase COI1(Coronatine Insensitive 1)could form a protein complex in planta,which promotes the degradation of both CO and JAZ3 in the presence of *** together,our results indicate that CO,a key regulator of photoperiodic flowering,is also involved in promoting flower senescence and abscission by augmenting JA signaling and *** propose that coordinated recruitment of photoperiodic and JA signaling pathways could be an efficient way for plants to chronologically order floral processes and ensure the success of offspring production.

Temperature variable and the efficiency of sperm mediated transfection of HPV16 DNA into cells

Aim:To pretreat sperm at various temperatures before exposure to human papillomavirus(HPV)16 DNA fragments and to assess the efficiency of HPV carrier sperm to transfect cumulus ***:Cumulus cells from follicular aspirates were obtained,pooled and divided into culture dishes containing Sybr Gold-stained HPV DNA carrying sperm that were either pretreated at 4℃,37℃or 40℃(n=5).The cells were incubated in 5%CO_(2. in air mixture at 37℃for 2. *** efficiency of sperm to take up fluorescent HPV DNA was determined at hour *** incubation,cumulus cell viability was assessed using the e0sin method and the percentages of fluorescent cumulus cells ***:Over half of all the cumulus cells became fluorescent with the highest percentage in the 37℃*** pretreated at 4℃had the greatest amount of HPV DNA *** sperm motility was similar for the 3 pretreatment *** were no differences in cumulus viability among the ***:Sperm pretreated at 37℃transferred the greatest amount of fluorescent HPV DNA fragments to the cumulus *** HPV DNA was observed in the nuclear and cytoplasmic *** data suggested the possibility of sperm as a vector for the transmission of HPV DNA to the cumulus cells surrounding ovulated oocytes,which might lead to early implantation failures.

The capabilities of the EISCAT Svalbard Radar for inter-hemispheric coordinated studies

In this article we want to present the EISCAT Svalbard Radar (ESR) in some detail, as well as some of the instruments of interest for ionospheric and magnetospheric research that are located in the vicinity of it. We particularly describe how this instrument cluster, close to the geomagnetic conjugate point of the Chinese Antarctic Zhongshan Station, can contribute to inter hemispheric coordinated studies of the polar ionosphere.
