Febrile seizures(FS),or fever-induced seizures,are the most common form of seizures during childhood and have been associated with an increased risk of epilepsy later in *** has been suggested that hyperthermia (temperature of≥38.4℃) during fever plays a crucial role in generation of ***,the mechanisms underlying the pathogenesis of hyperthermia-induced seizures are poorly *** previous studies,we and others have suggested that GABA receptors are likely to be associated with *** this study,by using whole-cell patch-clamp recording techniques,we demonstrated that hyperthermia(39-40℃) provoked a "febrile seizure-like event" expressed as spontaneous,recurrent,epileptiform discharges(SREDs) followed by a series of sustained depolarizations(SDs) in cultured rat cortical neurons(7-14 DIV).The SREDs were characterized by abruptly developing,paroxysmal depolarizing shifts(PDS) of membrane potential with high-frequency spike firing characteristic of electrographic *** also induced persistent neuronal hyperexcitability as assessed by their intrinsic electrogenic characteristics,including decreased:baseline resting potential(RP),input resistance(R),amplitude,duration and afterhyperpolarization(AHP) of spontaneous action potentials and,decreased absolute value of the threshold potential (TP) and threshold current(I) for evoking the cell firing,shortened duration,decreased inter-spike intervals(ISI) as well as increased firing frequency of evoked action ***(100μM),a specific GABA receptor agonist, significantly repressed the hyperthermia-induced neuronal hyperexcitability and epileptiform discharges in cultured cortical neurons,and these effects were prevented by treatment with the GABA receptor antagonist phaclofen. Moreover,hyperthermia elicited fast desensitization of G-protein activated inwardly rectifying K(GIRK) channels in cultured cortical neurons,whereas overexpression of GIRK1 and GIRK2.genes in cultured cortical

Objective: Water transport is the predominant phenomenon throughout the kidney and has important effect on renal function. Diffusion-weighted imaging (DWI) can detect the Brownian motion of water molecular in vivo and has recently been used in kidney. The purpose of this study was to evaluate whether ADC values correlate with GFR measured by ***: Diffusion-weighted echo-planar imaging (EPI) sequences (b value=0,500 sec/mm2. were performed with a 1.5 T MR unit in 55 patients (32.male, 2. female, aged 2.-77years, mean age54±10.2.years) with no local renal disease and hydronephrosis in a nonfasting and normal hydration status. The patients were referred for MR examination for various causes including unilateral renal artery stenosis(2.), bilateral renal artery stenosis(17), chronic nephropathy(12. and other diseases with normal kidney(6).This sequence completes in a breath-hold and scan time was no more than 2.s. The scan parameters were as following: TR 8000ms;TE 79.2.s;matrix 12.×12.;slice thickness 8mm with no gap;NEX 2.FOV 32.*** ADC values of kidneys were automatically calculated on ADW 4. 0 workstation, the regions of interest (ROIs) was manually positioned in the whole kidney on renal hilum level (Fig 1.). Renal scintigraphy was performed with a gamma camera (STARCAME 300i, GE) using 99mTc-DTPA, and split GFR was determined by the Gates methods. And the 110 kidneys were divided into 4 groups : groupl (normal renal function, n=44), GFR≥34ml·min;group2.(mild renal impairment, n=36), 34>GFR≥2. ml·min;group3 (moderate renal impairment, n=2.), 2.>GFR ≥10 ml·min;group4 (severe renal impairment, n=9), GFR<10 ml·min. All data were analyzed with SPSS 11.5 program for Windows. Differences between 4 groups respect to the mean renal ADCs were assessed by a one-way analysis of variance (ANOVA). This work characterizes the mitochondrial proteomic profile in the failing heart and elucidates the molecular basis of mitochondria in heart failure. Heart failure was induced in rats by myocardial infarction, and mitochondria were isolated from hearts by differential centrifugation. Using two-dimensional gel electrophoresis and matrix-assisted laser desorption/ionization-time of flight mass spectrometry, a system biology approach was employed to investigate differences in mitochondrial proteins between normal and failing hearts. Mass spectrometry identified 2. proteins differentially expressed that involved in energy metabolism. Among those, the up-regulated proteins included tricarboxylic acid cycle enzymes and pyruvate dehydrogenase complex subunits while the down-regulated proteins were involved in fatty acid oxidation and the OXPHOS complex. These results suggest a substantial metabolic switch from free fatty acid oxidation to glycolysis in heart failure and provide molecular evidence for alterations in the structural and functional parameters of mitochondria that may contribute to cardiac dysfunction during ischemic injury.

Clinical Effects of Intensive Insulin Therapy Treating Traumatic Shock Combined with Multiple Organ Dysfunction Syndrome

The therapeutic effects of intensive insulin therapy in treatment of traumatic shock combined with multiple organ dysfunction syndrome （MODS） were investigated. A total of 114 patients with traumatic shock combined with MODS were randomly divided into two groups： control group （n=56） treated with conventional therapy, and intensive insulin therapy group （n=58） treated with conventional therapy plus continuous insulin pumping to control the blood glucose level at range of 4.4-6.1 mmol/L. White blood cells （WBC） counts, prothrombin time （PT）, serum creatinine （SCr）, alanine aminotransferase （ALT）, serum albumin and PaO2.were measured before and at the day 1, 3, 5, 7 and 14 after treatment. The incidence of gastrointestinal dysfunction, the incidence of MODS, hospital stay and the mortality were also observed and compared. After intensive insulin therapy, the WBC counts, SCr, ALT and PT were significantly reduced （P0.05）, but the level of serum albumin was significantly increased （P0.05） at the day 3, 5, 7 and 14. In the meantime, the PaO2.was significantly elevated at the day 3, 5 and 7 （P0.01） after intensive insulin therapy. The incidence of gastrointestinal dysfunction, the incidence of MODS, the length of hospital stay and the mortality were markedly decreased （P0.01）. The results suggest early treatment with intensive insulin therapy is effective for traumatic shock combined with MODS and can decrease the length of hospital stay and the mortality.

Thirty-four samples(2..0%) were positive to 3 serogroups,and 44 samples to 4 or 5 serogroups in one *** from respiratory diseases ward showed the highest positive rate(2..3%),and the frequent detectged serogroups in these samples were serogroups 4(19.0%) and serogroup 12.17.2.).Conclusion:Among the 14 serogroups we tested,the predominant serogroup of *** found in these serum samples was serogroup *** reaction may exist between *** and other pathogenic microorganism or among the serogroups of ***.

The capacity for self-renewal and differentiation of human embryonic stem(ES)cells makes them a potential source forgeneration of pancreatic beta cells for treating type I diabetes ***,we report a newly developed and effectivemethod,carried out in a serum-free system,which induced human ES cells to differentiate into insulin-producing *** A was used in the initial stage to induce definitive endoderm differentiation from human ES cells,as detectedby the expression of the definitive endoderm markers Sox 17 and ***,all-trans retinoic acid(RA)wasused to promote pancreatic differentiation,as indicated by the expression of the early pancreatic transcription factorspdr1 and *** maturation in DMEM/F12.serum-free medium with bFGF and nicotinamide,the differentiatedcells expressed islet specific markers such as C-peptide,insulin,glucagon and *** percentage of C-peptide-posi-tive cells exceeded 15%.The secretion of insulin and C-peptide by these cells corresponded to the variations in *** transplanted into renal capsules of Streptozotocin(STZ)-treated nude mice,these differentiated human EScells survived and maintained the expression of beta cell marker genes,including C-peptide,pdx1,glucokinase,nkx6.1,IAPP,pax6 and *** percent of the transplanted nude mice exhibited apparent restoration of stable euglycemia;and the corrected phenotype was sustained for more than six *** new method provides a promising in vitro dif-ferentiation model for studying the mechanisms of human pancreas development and illustrates the potential of usinghuman ES cells for the treatment of type I diabetes mellitus.
