Simulation study on the efficiencies of MOET nucleus breeding schemes applying marker assisted selection in dairy cattle

Advantages of breeding schemes using genetic marker information and/or multiple ovulation and embryo transfer(MOET) technology over the traditional approach were extensively evaluated through *** yield was the trait of interest and QTL was the genetic marker *** dairy cattle breeding scenarios were considered,i.e.,traditional progeny testing breeding scheme(denoted as STANPT),GASPT scheme including a pre-selection of young bulls entering progeny testing based on their own QTL information,MOETPT scheme using MOET technology to generate young bulls and a selection of young bulls limited within the full-sib family,GAMOPT scheme adopting both QTL pre-selection and MOET technology,COMBPT scheme using a mixed linear model which considered QTL genotype instead of the BLUP model in GAMOPT,and three non-progeny testing schemes,*** MOET,GAMO and COMB schemes,corresponding to MOETPT,GAMOPT and COMBPT with progeny testing being part of the *** were selected based on their breeding value which was estimated under an animal model *** selection over 17 years was performed in the simulations and 30 replicates were designed for each *** influences of using QTL information and MOET technology on favorable QTL allele frequency,true breeding values,polygenetic breeding values and the accumulated genetic superiority were extensively evaluated,for five different populations including active sires,lactating cows,bull dams,bull sires,and young *** results showed that the combined schemes significantly outperformed other approaches wherein accumulated true breeding value *** difference between schemes exclusively using QTL information or MOET technology was not *** STANPT scheme was the least efficient among the 8 *** schemes using MOET technology had a higher polygenetic response than others in the 17th *** increases of frequency of the favorable QTL allele varied more greatly across t

Seedlings of three species of Malus were used to study the expression of mitogen-activatedprotein kinase (MAPK) in response to water stress:Malus hupehensis,a drought-sensitive species;Malussieversii,a drought-tolerant species;and Malus micromalus,a middle *** showed that MalusMAPK (MaMAPK,GenBank accession ***435805) was expressed in both roots and leaves of seedlingsof the three Malus species treated with 2.% polyethylene glycol for different time *** levelspeaked at 1.5 h after treatment with polyethylene glycol,then decreased to their lowest *** kinaseassays indicated that the dynamic changes of MAPK activity were very similar to those of the relativeexpression of MaMAPK ***,the peak of the former occurred slightly behind the *** wasnoticed that,although the kinase activity decreased after the peak,it was still higher than that of the controlduring the whole time *** results suggested that MaMAPK was regulated not only by water stressat the transcription level,but also by phosphorylation and dephosphorylation at the protein *** addition,of these three apple species,the highest MAPK activity and MaMAPK expression level was found in ***,followed by *** and ***,suggesting that MAPK might be correlated withdrought tolerance in these three *** different expression levels might be one of the molecularmechanisms of the different drought tolerances in Malus.

The optimal output tracking control (OTC) problem for nonlinear systems with time-delay is *** a series-based approx- imate approach,the original OTC problem is transformed into iteration solving linear two-point boundary value problems without time- *** OTC law obtained consists of analytical linear feedback and feedforward terms and a nonlinear compensation term with an infinite series of the adjoint *** truncating a finite sum of the adjoint vector series,an approximate optimal tracking control law is obtained.A reduced-order reference input observer is constructed to make the feedforward term physically *** exam- ples are used to test the validity of the series-based approximate approach.

Metal Transfer of Twin-wire Indirect Arc Argon Welding

Metal transfer of twin-wire indirect arc argon welding was investigated with high speed camcorder system based on a xenon lamp source and digital oscillograph, forces on droplet were analyzed in this paper. Results show metal transfer mode that of anodal and cathodal is not completely the same. It was divided into seven types of transfer mode; spray transfer and spray-projected transfer are the important transfer mode. The base-plate was melted by energy that of the arc and drop carrying, the drop transfer direction is different from that of the current flow, anodal and cathodal drop transfer at the same time, only the frequence and size are different.

Study on the Pathogenicity of Chinese Strains of Subgroup J Avian Leukosis Viruses

The pathogenicity of 4 Chinese strains of subgroup J avian leukosis viruses (ALV-J), SD9901, SD9902. YZ9901 and YZ9902.was studied. The results showed that only SD9902.among the 4 strains induced mortality from myeloid leukosis (ML). In the 12.meat-type chickens inoculated with SD9902.at 1-day-old, 9 died between 2. days and 38 days after inoculation. No death or ML was found in chickens inoculated with the other 3 strains during the period of 6 months. These results suggested the SD9902.strain of ALV-J was an acute transforming virus, but SD9901, YZ9901 and YZ9902.were non-transforming viruses. All 4 Chinese strains did not induce any tumors in egg-type SPF chickens during 7 months after hatching when viruses were injected into 11-day-old embryos.

Effect of Nano Red Elemental Selenium on GPx Activity of Broiler Chick Kidney Cells in vitro

A new selenium source, Nano red elemental selenium (Nano-Se) was used to study the effect on the GPx activity of broiler chick kidney cells (BCKC) in vitro, Sodium selenite (Na_ 2.SeO_ 3 ) and seleno-1-methionine (Se-Met) were used as the controls. The results showed that the effects of three kinds of Se forms on the GPx activity of BCKC were accordant(p>0.05) compared with each other at 0.01,0.05 and 0.10 μmol/L Se concentrations treatments. In the range of 0.00-0.10 μmol/L Se concentrations, the GPx activity increased with elevation of Se concentrations in medium. For the three kinds of Se forms, the GPx activity reached the climax at 0.10 μmol/L Se concentration. At 0.2. and 0.30 μmol/L Se concentrations, the influnces of three kinds of Se forms were not accordant with one another. For Nano-Se, the GPx activity at 0.2. and 0.30 μmol/L Se concentrations remained the same as that at 0.10 μmol/L Se concentration treatment. For Se-Met, the GPx activity at 0.2. μmol/L Se concentration treatment remained the same with 0.10 μmol/L treatment; the GPx activity at 0.30 μmol/L Se concentration treatment was declined significantly(p2. μmol/L treatment. For Na_ 2.SeO_ 3 , the GPx activity falled gradually with Se concentration increasing from 0.10 μmol/L to 0.30 μmol/L, and at 0.30 μmol/L Se concentration treatment, the GPx activity was less than the original of BCKC. The results implicated, on the GPx activity of BCKC in vitro, the ranking of width range of the most suitable Se concentration for nutrition curve of the three Se formes is Nano-Se>Se-Met>Na_ 2.SeO_ 3 .

Monte Carlo calculation method on low energy electronscattering in polybasic medium

A novel Monte Carlo method of simulating low energy electron (LEE) scattering in polybasic medium is presented. The Mott cross section is adopted to describe the elastic scattering of LEE, and a mean cross section idea is proposed instead of usual determination of the scattering center. The Joy’’s modified Bethe equation is used to calculate energy loss of LEE. The scattering processes of LEE in bulk PMMA have been simulated. And the electron backscattering coefficients, the electron interaction range and energy dissipation distribution of LEE in bulk PMMA have been calculated and compared with those calculated by the method of determining scattering center.

Production of porcine cloned transgenic embryos expressing green fluorescent protein by somatic cell nuclear transfer

In the present study, nuclear transferred embryos (NTEs) were reconstructed by using pig fetal fibroblasts as donors; in vitro matured oocytes as recipients. The effects of G418 selection on donor cells, duration of IVM of prepubertal gilt oocytes; oxygen tension in IVM of oocytes were investigated. The results were as follows: (i) When G418 selected cells expressing GFP were used as donors, the cleavage rate of NTEs decreased drastically in comparison to NTEs derived from donors without antibiotic selection (47.5% vs. 71.6%, p>0.05). For the blastocyst rate, no significant difference was observed between two groups (10% vs. 10.4%, p2.h (83.6% vs. 96.7%, p>0.05). However, no statistical difference was observed between NTEs derived from oocytes of 36 h IVM group; NTEs from oocytes of 42.h IVM group in the rates of cleavage (59.3% vs. 73.6%, p2., p2. tension; NTEs derived from oocytes matured under higher oxygen tension (2.% O2. in cleavage rate (70.6% vs. 67.1%, p2.3%, p2.h, while no significant differences were observed for developmental potential of transgenic embryos. Thus IVM 42.h is the better choice in order to obtain maximum number of MII oocytes as recipients. (iii) Lower oxygen tension; higher oxygen tension in IVM have no significant effect on development of cloned embryos.

A pig BAC library was constructed with genomic DNA from a male Erhualian pig. After partial digestion with HindⅢor BamHⅠthe fragments obtained were cloned into the pBeloBAC11 vector. The library consists of 18432. clones which stored in 480 pieces 384-well plates (2. plates per superpool). A two-step 4-dimension PCR screening system was established to screen the positive clones. An average insert size of 12. kb was estimated from 105 randomly isolated clones, which indicates that the library is more than five times of genomic coverage. For the demonstration of the probability to pick out any unique genes or DNA markers from the library, 10 single-copy genes were screened out and the positive clones were yielded between 1 and 8 with an average of 3.6. Positive superpools were obtained for 32.microsatellite markers selected from different regions of pig genome.
