Holocene grassland vegetation, climate and human impact in central eastern Inner Mongolia

Phytolith and pollen preserved in the Taipusi Banner paleosol profile, central eastern Inner Mongolia, provide evidence of Holocene grassland vegetation, climate and human impact. The combined phytolith and pollen records reveal a major change in vegetation composition about 5000 a BP. Before 5000 a BP, the vegetation was dominated by Aneurolepidium Chinese-Stipa grandis grassland of C4 grasses. After 5000 a BP, C4 grasses rapidly retreated, indicating a shift to colder and more arid conditions. The gradual invasion of Stipa krylovii, Ag-ropyron desertorum, Ephedra, Chenopodiaceae and caragana reveal the onset of grassland degeneration. Between 10000 and 872. a BP, the Aneurolepidium Chinese-Stipa grandis grassland included a small proportion of Artemisia shrub and Echionopos type plants, implying a strong winter monsoon and very weak summer monsoon. From 872. to 7000 a BP, C4 grasses were common, indicating a strengthening of the summer monsoon. Between 7000 and 5000 a BP, the Holocene thermal maximum was evident, with a significant expansion of C4 grasses and the presence of some trees (such as Pinus and Betula, and so on) in or near the study site. From 42.0?000 a BP, a sandy grassland of Artemisia and Agropyron desertorum together with Aster-type taxa occurred. Precipitation amelioration took place between 3000 and 2.70 a BP, with a rapid development of Echinops type plants and a small expansion of C4 grasses. From 2.70 a BP to present, human activities accelerated the process of grassland degeneration.

High Power, Room Temperature Terahertz Emitters Based on Dopant Transitions in 6H-Silicon carbide

Electrically pumped high power terahertz （THz） emitters that operated above room temperature in a pulse mode were fabricated from nitrogen-doped n-type 6H-SiC. The emission spectra had peaks centered on 5 THz and 12.THz （2. meV and 50 meV） that were attributed to radiative transitions of excitons bound to nitrogen donor impurities. Due to the relatively deep binding energies of the nitrogen donors, above 100 meV, and the high thermal conductivity of the SiC substrates, the THz output power and operating temperature were significantly higher than previous dopant based emitters. With peak applied currents of a few amperes, and a top surface area of 1 mm2. the device emitted up to 0.5 mW at liquid nitrogen temperature （77 K）, and tens of microwatts up to 333 K. This result is the highest temperature of THz emission reported from impurity-based emitters.

DET1-mediated COP1 regulation avoids HY5 activity over second-site gene targets to tune plant photomorphogenesis

TTie Ultisols in the Rana de canamero area in Southwest Spain showed aluminum(Al)phytotoxicity,and the clearance of natural vegetation and decades of intensive conventional agriculture caused the deplation of soil organic matter(SOM).Therefore,we studied the long-term effects of no tillage and liming using sugar beet foam(SF)and red gypsum(RG),alone or in combination,on the restoration of Ultisols affected by acidification,Al phytotoxicity,and SOM *** measured the main soil chemical properties,soil microbial biomass,soil enzyme activities involved in carbon,nitrogen,phosphorus,and sulfur mineralization,and the emission of volatile organic compounds(VOCs).The results indicated that liming effectively neutralized the soil acidity in the long term and,in combination with no tillage,significantly increased soil microbial biomass and enzyme ***-three VOCs were detected using the proton transfer reaction-time of flight(PTR-ToF)technique,and both liming and tillage changed the VOC emission *** greatest difference in VOC emission pattern was observed between no-tilled un-amended soils and tilled lime-amended soils,suggesting the activation of different metabolic pathways within the microbial communities of soils under different *** in VOC emission patterns could be attributed to the decomposition of carbohydrates,which were also sustained by the higher enzyme activities in the lime-amended soils.

目的探讨白藜芦醇苷(PD)对内毒素(LPS)诱导的人支气管上皮细胞BEAS-2.中clara细胞分泌蛋白(CCSP)和ca2.的影响。方法 MTT法筛选PD和LPS最佳浓度;复制LPS诱导的人支气管上皮细胞炎症反应模型,实验分为正常对照组、LPS模型组、LPS+PD组、PD+LPS组及PD对照组。real-time PCR法检测CCSP mRNA的表达量,Western blotting检测CCSP蛋白含量,流式细胞仪检测细胞内ca2.浓度。结果经过MTT法筛选,PD的最适浓度为0.5 mmol·L-1,LPS的最适浓度为100 mg·L-1。各组CCSP mRNA和蛋白表达情况为:与正常对照组相比,LPS模型组的表达量显著减少(Pca2.浓度检测结果:LPS+PD组和PD+LPS组的荧光强度显著低于LPS模型组(P2.细胞中,PD可促进CCSP mRNA和蛋白的表达,降低胞内ca2.浓度,预防性应用的效果明显优于治疗组。

The Arabidopsis heterotrimeric G-protein controls defense responses to necrotrophic and vascular fungi. The agbl mutant impaired in the Gβ subunit displays enhanced susceptibility to these pathogens. Gβ/AGB1 forms an obligate dimer with either one of the Arabidopsis Gγsubunits （γ1/AGG1 and γ2.AGG2.. Accordingly, we now demonstrate that the aggl agg2.double mutant is as susceptible as agbl plants to the necrotrophic fungus Plectosphaerella cucumerina. To elucidate the molecular basis of heterotrimeric G-protein-mediated resistance, we performed a comparative transcriptomic analysis of agbl-1 mutant and wild-type plants upon inoculation with P cucumerina. This analysis, together with metab- olomic studies, demonstrated that G-protein-mediated resistance was independent of defensive pathways required for resistance to necrotrophic fungi, such as the salicylic acid, jasmonic acid, ethylene, abscisic acid, and tryptophan-derived metabolites signaling, as these pathways were not impaired in agbl and aggl agg2.mutants. Notably, many mis-reguiated genes in agbl plants were related with cell wall functions, which was also the case in aggl agg2.mutant. Biochemical analyses and Fourier Transform InfraRed （FTIR） spectroscopy of cell walls from G-protein mutants revealed that the xylose content was lower in agbl and aggl agg2.mutants than in wild-type plants, and that mutant walls had similar FTIR spec-tratypes, which differed from that of wild-type plants. The data presented here suggest a canonical functionality of the Gβ and Gγ1/γ2.subunits in the control of Arabidopsis immune responses and the regulation of cell wall composition.

Objective The large conductance ca-activated K(BK) channels,which are widely distributed in the central nervous system(CNS),have been considered as intracellular calcium sensors in neuron and play an important feedback role in controlling ca flux and subsequent ca-dependent processes including neurotransmitter release and cellular *** The effects of neuropeptide galanin on BK channels in human embryonic kidney(HEK2.3) cells co-expressing both GalR2.and BK alpha subunit were examined by determining the whole-cell currents and single-channel ***(1) The application of galanin enhanced the currents of BK channels,in a PTX-independent and concentration-dependent *** ED50 value of dose-dependent curve was 71.8+16.9 nM.(2. Galanin-induced activation of BK was mediated by GalR2.but not GalR1 nor GalR3 as galanin did not facilitate BK current in the cells co-expressed cDNAs of BK with GalR1 or GalR3.(3) Chelating intracellular ca by either the slow ca buffer EGTA or the fast ca buffer BAPTA in the pipette solution abolished galanin-mediated activation of BK channels,indicating the important role of ca in galanin-induced BK current.(4) The galanin-induce BK current persisted when the bath solution was replaced by ca-free solution,suggesting the extracellular ca in not required to lead activation of BK channels.(5) The galanin-induced BK channels activation mediated by the ca efflux from the sarcoplasmic reticulum /endoplasmic reticulum(SR/ER) was confirmed by application of thapsigargin,an irreversible inhibitor that emptied ca pool from SR/ER.(6) Inositol-1,4,5-triphosphate receptor(IP3R) on ER was identified as the mediator responsible for increased intracellular ca that activated BK *** Activation of GalR2.by its ligand galanin,led to elevation of intracellular ca level due to ca efflux through IP3R from ER and open BK channels.

To gain insight into the coordination of gene expression profiles under abscisic acid (ABA) and H2.2.applications, global changes in gene expression in response to ABA and H2.2.in Arabidopsis seedlings were investigated using GeneChip (santa clara, ca, {2.) arrays. Among over 2. 000 genes present in the arrays, 459 transcripts were found to be significantly increased, whereas another 2.1 decreased following H2.2.treatment compared with control. Similar to treatment with H2.2. ABA treatment elevated the transcription of 391 genes and repressed that of 32. genes. One hundred and forty-three upregulated genes and 75 downregulated genes were shared between the two treatments and these genes were mainly involved in metabolism, signal transduction, transcription, defense, and resistance. Only two genes, which encode an APETALA2.dehydra-tion-responsive element binding protein (AP2.DREBP) family transcriptional factor and a late embryogenesis-abundant protein, were downregulated by H2.2. but upregulated by ABA. These results suggest that, similar to ABA, H2.2.plays a global role in gene transcription of Arabidopsis seedlings. The transcriptional responses induced by the application of exogenous ABA and H2.2.overlapped substantially. These two treatments regulated most of the downstream genes in a coordinated manner.
