H-Ras is well known as one of the essential components of Ras/Raf/MEK/ERK cascade, which is a critical prosurvival signaling mechanism in most eukaryotic cells. Ras targets Raf/MEK/ERK cascade by integrating and transmitting extracellular signals from growth factor receptors to Raf, leading to the propagation of signals to modulate a serious of cellular survival events. Apoptosis signal-regulating kinasel (ASK1) serves as a general mediator of cell death because it is responsive to a variety of death signals. In this study, we found that H-Ras interacted with ASK1 to cause the inhibition of both ASK1 activity and ASK1-induced apoptosis in vivo, which was reversed only partially by addition of RafS621A, an antagonist of Raf, whereas MEK inhibitor, PD98059, and PI3K inhibitor, LY294002, did not disturb the inhibitory effect of H-Ras on ASK-1-induced apoptosis. Aim:To purify and characterize the coagulant protein FIa from Daboia russellisiamensis (Myanmar) ***:FIa was purified from Daboia russellisiamensis (Myanmar) venom by ion-exchange chromatography on CM-SephadexC-50,and gel filtration on Sephadex G-75 and a Superdex 75 *** hemo-static activity of FIa was determined by the method of Williams and *** chromogenic substrates were used respectively to determine the activa-tion of factor X and *** fibrinogen-clotting activity of FIa wasdetermined by the method of Gao et *** saline was used as a negativecontrol while factor Xa and thrombin were used as positive controls,***:FIa,a coagulant protein,was achieved by ion-exchange chromatogra-phy and gel filtration with a molecular weight of 34 479 and an isoelectric point *** was shown to have strong hemostatic *** hemostatic activity of0.5 mg FIa was equal to that of 1.5625 u *** primarily activated factor X,however,had no influence on prothrombin,nor did it cleave or clot ***:FIa is a factor X-activating enzyme,which could activate factor X tofactor Xa,but has no effect on prothrombin and fibrinogen.

BACKGROUND:Combined hepatocellular carcinoma and cholangiocarcinoma (cHCC-CC) is an uncommon subtype of primary hepatic carcinoma,and its prognosis is *** study was undertaken to investigate the prognosis and the clinicopathological characteristics of cHCC-CC,including their possible cellular ***:Among 852 patients with a primary hepatic carcinoma who underwent hepatectomy from January 1998 to April 2008 at our hospital,cHCC-CC was identified in 14 patients The clinicopathological characteristics of the 14 patients were analyzed *** expression of the liver stem cell markers (c-kit,CD90,CD133 and CK19) in the tumor tissue was detected by immunohistochemistry,and the Kaplan-Meier method was used to evaluate ***:Among the 14 patients,9 presented with abdominal pain,3 with anorexia and debilitation,and the remaining two patients were *** mean age was 53.6±3.0 (range 38-74) *** the included patients,11 had an elevated serum alpha-fetoprotein level,13 were infected with hepatitis B virus,9 had vascular invasion and 1 had lymph node metastasis The average diameter of the tumors was 9.9±1.1 (range 5.0-16.0) *** median overall survival time was 7.9±1.0 months In addition,the presence of the liver stem cell markers,c-kit CD90,CD133 and CK19 was 71.4%,85.7%,92.9% and 78.6% *** four markers were simultaneously expressed in eight ***:cHCC-CC has aggressive characteristics and the prognosis is extremely *** high expression of liver stem cell markers in the tumor tissue suggests that these tumors may derive from liver stem cells.

BACKGROUND:Liver enriched natural killer (NK) cells are of high immune ***,the function of donor liver NK cells in allogeneic liver transplantation (LTx) remains ***:Ten Gy of whole body gamma-irradiation (WBI) from a 60 Co source at 0.6 Gy/min was used for depleting donorderived leukocytes,and transfusion of purified liver NK cells isolated from the same type rat as donor (donor type liver NK cells,dtl NKs) through portal vein was performed immediately after grafting the irradiated ***-transplant survival observation on recipients and histopathological detection of liver grafts were adoptive to evaluate the biological impact of donor liver NK cells on recipients’ survival in rat ***:Transfusion of dtl NKs did not shorten the survival time among the recipients of spontaneous tolerance model (BN to LEW rat) after rat LTx,but prolonged the liver graft survival among the recipients depleted of donor-derived leukocytes in the acute rejection model (LEW to BN rat).Compared to the recipients in the groups which received the graft depleted of donor-derived leukocytes,better survival and less damage in the allografts were also found among the recipients in the two different strain combinations of liver allograft due to transfusion of dtl ***:Donor liver NK cells alone do not exacerbate liver allograft acute ***,they can alleviate it,and improve the recipients’ survival.

This paper considers a distributed interference avoidance problem employing frequency assignment in the Gaussian interference channel(IC).We divide the common channel into several sub-channels and each user chooses one sub-channel for transmission in such a way that the total interference in the IC is *** mechanism named interference avoidance in this paper can be modeled as a competitive game *** a completely autonomous distributed iterative algorithm called distributed interference avoidance algorithm(DIA)is adopted to achieve the Nash equilibrium(NE)of the *** to the self-optimum,the DIA is a sub-optimal ***,through introducing an optimal compensation(or price)into the competitive game model,we successfully develop a compensation-based game model to approximate the optimal interference avoidance ***,an optimal algorithm called iterative optimal interference avoidance algorithm(IOIA)is proposed to reach the optimality of the interference avoidance *** analyze the implementation complexity of the proposed algorithm which is only O(N),with N standing for the number of users in the *** also give the proof on the convergence of the proposed *** performance upper bound and lower bound are derived for the IOIA *** simulation results show that the IOIA does reach the optimality under the condition of interference avoidance mechanism.

This study aims to discuss the effect of preventing pathological changes and cognitive degeneration of Tg2576 mice by inoculating the subunit fragment of Aβ ***-two Tg2576 mice were randomly divided into four groups, each having eight mice:Group I, the control group, inoculated with adjuvants;Group II, the Aβ42 group, inoculated with Aβ42 vaccine;Group Ⅲ, the Aβ1―15 group, inoculated with Aβ1―15 vaccine;and Group IV, the Aβ36―42 group, inoculated with Aβ36―42 *** titer of the serum anti-body against Aβ42(Group II) was significantly higher than that of the control group(Group I), and a low level of antibodies could be detected in the brain homogenate in the three vaccine-inoculated *** water maze test showed that the Aβ42 group, Aβ1―15 group and Aβ36―42 group were obviously im-proved compared with the control *** cultured splenocytes sampled from each group were induced by Con A or their respective antigens, and the cell proliferation of the three vaccine-inoculated groups was significantly higher than that of the control *** the Aβ42 group, IL2 and IFN-γ were relatively low and IL4 and IL10 were relatively *** contrast, IL4 and IL10 were much higher in the Aβ1―15 group and IL2 and IFN-γ were much higher in the Aβ36―42 *** immunohistochemical test showed a large number of senile plaques in the brain cortex and hippocampus of the mice in the con-trol group, no senile plaque in the brain of the Aβ1―15 group and Aβ42 group mice, and a small number of senile plaques in the brain of the Aβ36―42 group *** results suggest that the subunit fragment of Aβ1―15 vaccine could prevent not only cognitive and behavioral degeneration but also Aβ deposition and formation of senile plaques in Tg2576 mice.

A comparative molecular field analysis of cytotoxic beta-carboline analogs

AIM: To derive a model that could be used in drug design. METHODS: Beta-carbolines are reported to have antitumor activities on cultured cancer cell lines. A comparative molecular field analysis (CoMFA) was undertaken to elucidate the correlation of cytotoxities and structural parameters of 16 beta-carboline analogs (1-16). The compound 12 was finally used as a template for the other compounds in the dataset because of its highest biological activity. RESULTS: The CoMFA applied to the final alignment resulted in a qcv of 0.656 and it showed that the 2 steric fields contributed 43.3 % of the model information while the electrostatic fields represented the other 56.7 %. CONCLUSION: Three designed compounds, which were predicted to have high, moderate and low activities respectively, were synthesized. The IC50 values of these compounds indicated the significance of the analysis in this study. The model derived from the current study could be further used in design for more active compounds.
