Aim:To investigate the possible synergism of hydrochlorothiazide andnitrendipine on reducing both blood pressure(BP)and blood pressure variabil-ity(BPV)in spontaneously hypertensive rats(SHR).Methods:Seventy ani-mals were randomly divided into seven *** doses were 5 and 10 mg/kgfor nitrendipine,10 and 20 mg/kg for hydrochlorothiazide and 10+5,20+10mg/kg,respectively,for the combination of these two drugs and 0.8% carboxym-ethylcellulose as *** drugs were given via a catheter of gastric *** was then continuously recorded for 5h from 1h before drug administrationto the end of 4th hour after drug administration,in conscious and freely ***:The effects on both BP and BPV reduction of the combination ofhydrochlorothiazide and nitrendipine were greater than the single drug in *** two drugs possessed an obvious synergism on both systolic blood pressure(q=1.79 with small dose and q=1.23 with large dose)and systolic blood pres-sure variability reduction(q=1.79 with small dose and q=1.39 with large dose)in ***:The present work clearly demonstrated that there was asynergistic effect between hydrochlorothiazide and nitrendipine in lowering andstabilizing BP in SHR.

Objective Colorectal cancer is one of the most common cancers in the world, accounting for nearly 10%of all incident cases of *** growth factor-β1 (TGF-β1) is a pleiotropic cytokine that has been implicated in the pathogenesis of colorectal *** most studied -509C>T polymorphism of TGF-β1 gene has been associated with various kinds of *** aim was to investigate the association between this genetic variant and the risk and/or progression of colorectal cancer. Method We conducted a case-control study of 150 colorectal cancer cases and 503 healthy *** was extracted from blood specimens,and -509C>T polymorphism in the TGF-β1 gene promoter was genotyped by polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP).In addition,colorectal cancer tissues(n=70) were obtained from the studied cases for measurement of TGF-β1 mRNA expression *** also assessed the plasma TGF-β1 levels of cases(n=88) and healthy subjects (n=120).Results In our study group,the high TGF-β1 producer genotypes,-509TT,was not associated with an increased risk of colorectal cancer compared with other genotypes, nor with the progression of colorectal ***,we observed that colorectal cancer patients especially those with a more aggressive disease behavior were more frequent with C *** Our results suggest that TGF-β1 -509C>T polymorphism was not associated with both an increased risk and progression of colorectal cancer.

Aim: To investigate the effects of CpG-oligodeoxynucleotide (CpG-ODN) on the formation of macrophage foam cells and related gene expression. Methods: A gene expression profile was examined by microarray techniques, and mRNA expression was detected by reverse transcriptase polymerase chain reaction (RT-PCR). The cholesterol and cholesteryl ester contents of cells were determined by high performance liquid chromatography. Results: CD36, LPL, and Fcγ2b, which were related to lipid metabolism and the formation of macrophage foam cells, were upregulated after CpG-ODN stimulation. The mRNA expression related to the formation of foam cells was confirmed by semiquantitative RT-PCR. Moreover, histochemical analysis confirmed that lipid deposits inside cells increased after CpG-ODN treatment. However, using flow cytometry, we found that CpG-ODN had no effect on the expression of membrane receptors. Conclusion: CpG-ODN up-regulated the expression of genes in macrophage foam cell formation.

Human fibrinogen-related protein-1/liver fibrinogen-related protein-1 (HFREP-l/LFIRE-1), a liver-specific protein, is a member of fibrinogen superfamily that exerts various biological activities. However, the function of HFREP-l/LFIRE-1 in liver remains unknown. Here we isolated its mouse ortholog gene-mouse fibrinogen-related protein-1 (mfrep-1), which encoded 314 amino acids, exhibiting 80.4% similarity to HFREP-l/LFIRE-1. Northern blot analysis revealed that 1.2-kb mfrep-1 mRNA was detected selectively in mouse liver. To explore the function of MFREP-1, we examined the levels of mfrep-1 mRNA during regeneration after 70% partial hepatectomy (PHx) in mice, mfrep-1 mRNA increased in the regenerating liver and reached the first shoulder peak at 2-4 h after PHx. Cycloheximide pretreatment could suppress the induction of mfrep-1, indicating the up-regulation of this gene need de novo protein synthesis. Aim:The present work was designed to investigate the effects of nine commonlyused antibypertensive drugs on blood pressure(BP)and blood pressure variabil-ity(BPV)in conscious sinoaortic-denervated(SAD)***:Seventy-twoSAD rats were randomly divided into nine *** were respectively givennifedipine 3 mg/kg,nitrendipine 5 mg/kg,amlodipine 1mg/kg,clonidine 10 μg/kg,prazosin 0.5 mg/kg,atenolol 20 mg/kg,telmisartan 20 mg/kg,hydrochlorothiazide40 mg/kg or captopril 50 mg/*** drugs were given via a catheter previouslyimplanted into the *** was recorded for 5 h from 1 h before drug admin-istration to 4 h after drug administration in conscious,freely moving ***:It was found that all these nine drugs significantly decreased BP in SAD *** these drugs(nifedipine,nitrendipine,amlodipine,clonidine,prazosin andatenolol)significantly decreased BPV in SAD rats,but the remaining three drugsdid *** and atenolol increased the heart period and the others did *** drugs affected the heart period ***:Among nine antihy-pertensive drugs from different classes,calcium antagonists and sympatheticinhibitors decreased BPV in SAD rats.
