Phage display selection on whole cells yields a small peptide specific for HCV receptor human CD81

The human CD81 (hCD81), the most recently proposed receptor of hepatitis C virus (HCV), can especifically bind to HCV envelope glycoprotein 2 (E2). In this study, hCD81-expressing murine NIH/3T3 cells were used to select hCD81-binding peptides from a phage displayed nonapeptide library (PVIII9aaCys). Eighteen of the 75 clones selected from the library showed specific binding to the hCD/81-expressing NIH/3T3 cells by enzyme linked immunosorbent assay (ELISA) and competitive inhibition test. Twelve out of the 18 clones shared the amino acid motif SPQYWTGPA. Sequence comparison of the motif showed no amino acid homology with the native HCV E2. The motif-containing phages could competitively inhibit the ability of HCV E2 binding to native hCD81-expressing MOLT-4 cells, and induce HCV E2 specific immune response in vivo. These results suggest that the selected motif SPQYWTGPA should be a mimotope of HCV E2 to bind to hCD81 molecules. Our findings cast new light on developing HCV receptor antagonists.

CYP2E1 Pst Ⅰ/Rsa Ⅰ polymorphism and colorectal cancer risk:A meta-analysis

AIM:To clarify the association between CYP2E1 PstⅠ/RsaⅠ polymorphism and susceptibility to colorectal ***:A meta-analysis based on 10 eligible casecontrol studies involving 4979 cases and 6012 controls was carried out to summarize the data on the association between CYP2E1 RsaⅠ/PstⅠ polymorphism and colorectal cancer ***:In comparison of the homozygote c2c2 and c2 carriers(c1c2 + c2c2) and the homozygous wild-type genotype(c1c1),no association was found between CYP2E1 RsaⅠ/PstⅠ polymorphism and colorectal cancer risk [odds ratio(OR)=1.24(95% CI:0.93-1.66) for c2c2;OR = 1.02(95% CI:0.88-1.19) for c2 carriers].In stratif ied analysis,Caucasians with c2c2 homozygote appeared to have an increased risk of colorectal cancer(OR=2.67,95% CI:1.03-6.89,P=0.043),no signif icant associations were found in other ***:c2c2 homozygote of CYP2E1 PstⅠ/RsaⅠ polymorphism may be associated with the increased risk of colorectal cancer in Caucasians,which needs further investigations.

The second envelope glycoprotein （E2） of hepatitis C virus has been shown to bind human target cells and has become a major target for the development of anti-HCV vaccines. Anti-E2 antibodies have been suggested to be of clinical significance in diagnosis, treatment and prognosis of hepatitis C. However, large-scale expression and purification of E2 proteins in mammalian cells is difficult. As an alternative, E2 fragment （aa 385-730） with a four-amino-acid mutation （aa 568-571 PCNI to RVTS） was expressed as hexa-histidine-tagged full length protein [E2N730（m）] in E. coli and purified to over 85% purity. Purified E2N730（m） was specifically recognized by homologous hepatitis C patient serum in Western blot, suggesting that it displayed E2specific antigenicity. Rabbit antiserum raised against E2N730（m） recognized E2 glyco-proteins expressed in mammalian cells in Western blot. Changes in Autoantibodies against β1-Adrenoceptor and M2-Muscarinic Receptor during Development of Renovascular Hypertension in Rats

In an experimental rat’s renovascular hypertension model, we studied the genesis of anti- cardiac β1-adrenoceptor and M2-muscarinic receptor autoantibodies in relation to the changes in immunological function during the development of renal hypertension. The biological activities of these autoantibodies were also examined. It was shown that after two weeks of operation both the frequency of occurrence and the titre of autoantibodies to cardiac β1-adrenoceptor and M2-muscarinic receptor were significantly increased as compared with the control of pre-treatment. The increased autoantibodies lasted for several weeks and then automatically decreased gradually to the pre-clipping level at 10 weeks. Meanwhile the ratio of CD4 /CD8 + + was also undergone an initial increase followed by gradual recovery and correlated well with the changes in antibody titre. The biological effects of these autoantibodies displayed an “agonistic-like” activities on the beating frequency of cultured neonatal cardiomyocyte. It is suggested that autoimmune mechanisms are involved in the pathogenesis of renal hypertension and the cardiac receptor autoantibodies might be one of the mechanisms leading to cardiac dysfunction.

Isolated metastatic melanoma of the pancreas is very ***,there is very limited experience with surgical resection of pancreatic *** potential benefit of metastasectomy can improve the quality of life and survival time of *** present a case of a 39-year-old Chinese male with a solitary pancreatic tumor which was considered a cystic benign lesion for *** and immunohistochemistry showed that the tumor in pancreatic tail was a metastasis from a malignant melanoma of the *** other metastastic foci were found in *** tumor was completely resected with combined distal pancreatectomy and *** patient has survived 25 mo without any signs of local recurrence or other metastatic lesions after operation,indicating that complete surgical resection of a solitary metastatic melanoma of the pancreas can prolong the survival time of patients.

DNA Vaccine of SARS-Cov S Gene Induces Antibody Response in Mice

The spike (S) protein, a main surface antigen of SARS-coronavirus (SARS-CoV), is one of themost important antigen candidates for vaccine design. In the present study, three fragments of the truncated S protein were expressed in E. coli, and analyzed with pooled sera of convalescence phase of SARS patients. The full length S gene DNA vaccine was constructed and used to immunize BALB/c mice. The mouse serum IgG antibody against SARS-CoV was measured by ELISA with E. coli expressed truncated S protein or SARS-CoV lysate as diagnostic antigen. The results showed that all the three fragments of S protein expressed by E. coli was able to react with sera of SARS patients and the S gene DNA candidate vaccine could induce the production of specific IgG antibody against SARS-CoV efficiently in mice with seroconversion ratio of 75% after 3 times of immunization. These findings lay some foundations for further understanding the immunology of SARS-CoV and developing SARS vaccines.

Frequent ventricular premature beats increase blood pressure variability in rats

AIM: The present study was designed to test a hypothesis that nonfatal ventricular arrhythmia such as ventricularpremature beats (VPB) is a contributing factor in the elevation of blood pressure variability (BPV). METHODS:Blood pressure (BP) and electrocardiogram were continuously recorded. The relation between VPB and BPV wasobserved under conscious state in chronic myocardial infarction (MI) rats one month after ligation of the leftcoronary artery, and further verified under anesthetized state in rat model of ventricular arrhythmia produced byacute intravenous infusion of aconitine. RESULTS: MI rats exhibited a big difference in the count and pattern ofVPB, and were divided into no VPB, occasional VPB, and frequent VPB groups. Among the three groups, therewere no differences in BP, heart period (HP), and MI size. However, BPV was markedly higher in frequent notoccasional VPB rats, and HP variability (HPV) was larger in both frequent and occasional VPB rats, when comparedwith no VPB rats. In the whole population of MI rats, BPV was positively correlated with VPB and HPV, not withBP, HP and MI size. Infusion of aconitine had no effect on BP, HP, BPV, and HPV during the period without *** VPB after several minutes of aconitine infusion induced significant increase in BPV and HPV with nochange in BP and HP. BPV was also positively correlated with VPB and HPV, not with BP and HP. Hemodynamicsin aconitine-evoked ventricular tachycardia was characterized as lower BP, higher BPV, and higher ***: High BPV can be caused by frequent not occasional VPB in rats.

In the field of stem cell research, SP (side population) phenotype is used to define the property that cells maintain a high efflux capability for some fluorescent dye, such as Hoechst 33342. Recently, many researches proposed that SP phenotype is a phenotype shared by some stem cells; some progenitor cells,; that SP phenotype is regarded as a candidate purification marker for stem cells. In this research, murine fertilized ova (including conjugate; single nucleus fertilized ova), 2-cell stage; 8-cell stage blastomeres, morulas; blastocysts were isolated; directly stained by Hoechst 33342 dye. The results show that fertilized ovum, blastomere; morula cells do not demonstrate any ability to efflux the dye. However, the inner cell mass (ICM) cells of blastocyst exhibit SP phenotype, which is consistent with the result of embryonic stem cells (ESCs) in vitro. These results indicate that the SP phenotype of ICM-derived ESCs is an intrinsic property; independent of the culture condition in vitro,; that SP phenotype is one of the characteristics of at least some pluripotent stem cells, but is not shared by totipotent stem cells. In addition, the result that the SP phenotype of ICM cells disappeared when the inhibitor verapamil was added into medium implies that the SP phenotype is directly associated with ABCG2. These results suggest that not all the stem cells demonstrate SP phenotype,; that SP phenotype might act as a purification marker for partial stem cells such as some pluripotent embryonic stem cells; multipotent adult stem cells, but not for all stem cells exampled by the totipotent stem cells in the very early stage of mouse embryos.
