The complementary DNA （cDNA） amplified fragment length polymorphism technique wasused to isolate transcript-derived fragments corresponding to genes involved in the flowering of tea *** sequence analysis of an approximately 300 bp differential fragment amplified by primer combi-nation E11M11revealed 80%-84% similarity to the corresponding part of an α-tubulin gene of other *** complete cDNA sequence of this α-tubulin was cloned by the rapid amplification of cDNA ends technique;its full length is 1537 bp and contains an open reading frame of 450 amino acid residues with two N-glycosylation sites and four protein kinase C phosphorylation *** deduced amino acid sequences didshow significant homology to the α-tubulin from other plants that has been reported to be a pollen-specificprotein and could be correlated with plant cytoplasm-nucleus-interacted male *** named this completecDNA *** nucleotide and amino acid sequence data of Tual have been recorded in the GenBanksequence database with the accession *** Tual gene was cloned into the pET-32aexpression system and expressed in Escherichia coli BL21trxB（DE3）.The molecular weight of expressedprotein was deduced to be approximately 49 *** blot analysis was used to identify the temporalexpression of Tual in tea *** studies of the effect of Tual protein on pollen tube growth indicatedthe Tual solution obviously promoted the growth of tea pollen tube.

Singularity Detecting with the SecondGeneration Wavelet

Adopting the lifting scheme, introduced by Sweldens, a new wavelet is constructed. It overcomes the shortcoming of the classical wavelets that has no flexibility in the number of vanishing moment. Two examples are given to prove the second-generation wavelets's potentialities in the singularity detection of signal: a wavelet with vanishing moment and symmetry can be constructed according to the problem. Taking Huize County as an example, the paper analyzed the landscape structure(landscape element structure, landscape type structure, landscape spatial structure, landscape succession structure) and the relations between landscape structure and land use. It was pointed out that the agriculture should be developed in harmony with the landscape structure in the study area.

b-DAZL:A novel gene in bovine spermatogenesis

The DAZL gene plays an important role in *** and mutation of this gene has been frequently associated with the sterility in both vertebrates and ***,a cDNA containing the complete open reading frame of DAZL gene,which encodes putative RRM RNA-binding protein,was isolated from bovine tissues by *** isolated gene was referred to as ***-based location analysis showed that b-DAZL is located near the end of chromosome *** b-DAZL gene and its vertebrate homologue shared a high nucleotide similarity (90-94%) in the coding region and in the genomic organization,each consisting of 10 exons and 9 *** protein products contained a highly conserved RNA-binding motif,a unique DAZ repeat, and PUM2 and PABP interacting ***-PCR assay revealed that b-DAZL was expressed in the testis and ***,no signals were detected in the testes of sterile male *** microstructure of the testes of sterile males showed very few spermat- ocytes but mostly somatic *** observations are consistent with the typical phenotype of testes with defective DAZL expression. Subsequent gene detection studies indicated that transcription arrest might lead to a b-DAZL transcript defect in testes of cattle-yaks. Our results suggest that b-DAZL may be involved in spermatogenesis,and that transcription arrest of the gene is associated with male sterility in cattle-yaks.

Effect of Silicon on Leaf Ultrastructure, Chlorophyll Content and Photosynthetic Activity of Barley Under Salt Stress

Two contrasting cultivars of barley (Horderm vykgare L.): Kepin No. 7 (salt sensitive), and Jian 4 (salt tolerant) were grown in a hydroponics system with 2 NaCl levels: 60 mmol NaCl L-1 and 120 mmol NaCl L-1, and 3 Si levels: 0 mmol Si L-1, 0.5 mmol Si L-1 and 1.0 mmol Si L-1 (as silicic acid). Compared with the plants treated with 60 mmol NaCl L-1 alone, the leaf chlorophyll contents of plants treated with salt and Si increased significantly for salt-sensitive cultivar at tillering stage, but for salt-tolerant cultivar,the addition of Si resulted in an obvious increase in the leaf chlorophyll content of plants exposed to 120 mmol NaCl L-1. However, this Si-enhancement of leaf chlorophyll content was also observed in the salttolerant plants at jointing stage, but not in the salt-sensitive plants. Moreover, leaf chlorophyll content was consistently higher for the salt-tolerant cultivar than for the salt-sensitive cultivar irrespective of salt and/or Si treatment. Compared with the plants treated with salt alone, net CO2 assimilation rate in plant leaves increased significantly for both cultivars when trested with salt and Si. The addition of Si to the salt treatment was found to improve the cell ultrastructure of leaves. Under salt stress condition, the double membranes of chloroplasts disappeared, but membrane integrity was markedly improved in the salt treatment supplemented with Si. Silicon was also found to ameliorate the damage to the ultrastructure of chloroplast granae which appeared to be disintegrated and vague in salt treatments without added Si. The results support previous work which showed that Si decreases the permeability of plasma membranes of salt-stressed barley, thus mitigating salt damage.

Advances on methods for mapping QTL in plant

Advances on methods for mapping quantitative trait loci (QTL) are firstly summarized. Then, some new methods, including mapping multiple QTL, fine mapping of QTL, and mapping QTL for dynamic traits, are mainly described. Finally, some future prospects are proposed, including how to dig novel genes in the germplasm resource, map expression QTL (eQTL) by the use of all markers, phenotypes and micro-array data, identify QTL using genetic mating designs and detect viability loci. The purpose is to direct plant geneticists to choose a suitable method in the inheritance analysis of quantitative trait and in search of novel genes in germplasm resource so that more potential genetic information can be uncovered.

Soil Fertility Indices of Citrus Orchard Land Along Topographic Gradients in the Three Gorges Area of china

In the Three Gorges Area of china, soil erosion and the resultant non-point source pollution and ecological degradation have endangered agricultural ecosystems and fresh water reservoirs. Although efforts have been undertaken to reduce soil and water losses from slope land used for citrus production, information on the effects of management practices on soil fertility indices is either limited or nonexistent. This study was conducted to compare the effects of 10 years of various management practices, citrus intercropped with white clover (WC), citrus mulched with straw (SM), citrus intercropped with contour hedgerows (CH), citrus orchard land with impermeable membrane (IM), and citrus intercropped with wheat (Triticum aestivum) and peanut (Arachris hypogaea) (WP), as treatments on soil fertility indices with that of the conventional citrus management (CM). Results showed that the soil organic carbon, total and available nitrogen, available potassium, and water-stable aggregate (> 0.25 mm) contents at the 0–5 cm depth were higher for the WC and SM treatments than the CM treatment. There was also spatial variation in soil fertility along slopes of the WC and SM treatments. The soil total and available nitrogen, phosphorus, and potassium, and water-stable aggregate (> 0.25 mm) contents at both the 0–5 and 5–20 cm depths were higher for the CH and IM treatments than the CM treatment. The average soil available nitrogen and available potassium contents were higher for the WP treatment than the CM treatment, but the WP treatment had little effect on the soil organic carbon, total nitrogen, and water-stable aggregate (> 0.25 mm) contents. These suggested that white clover intercropping and straw mulching were the most effective approaches to improve soil fertility in citrus orchard land of the Three Gorges area.

The Effect of Intra-cerebral Microinjection with Daidzein on the Plasma LH Level in the Castrated Boars

Daidzein (DA) was microinjected into mediobasal hypothalamus (MBH) and nucleus ventrome-dialis thalami (VM) of the castrated male Goettingen Mini pigs (n = 9, 10 μl·animal-1, 8 pgDA·μl-1 and plasma luteinizing hormone (LH) levels were compared between the prior and the posterior treatment. The result showed that LH levels after the cerebral administration (ad) tended to increase compared to the levels before ad. In MBH, LH levels in 4 cases (4/5), rose and were not changed in 1 case at 0.5 - 2 hours after ad compared to those before ad. There were no significant changes at 2.5 hours after ad. When it was injected in VM, LH levels in 3 cases (3/4) rose, and were not changed in 1 case after ad compared to those before ad. In the control, there were no changes in plasma LH levels between the pre-and post-treatment except 1 case in MBH. This study suggested that DA could up-regulate LH secretion through hypothalamus level in the castrated boars.

Application of Equilibrium Partitioning Approach to the Derivation of Sediment Quality Guidelines for Metals in Dianchi Lake

For the past 20 years, numerous studies have been carried out on the application of equilibrium partitioning approach （EqPA） for the derivation of sediment quality guidelines （SQGs）. However, for metals, few Equilibrium-partitioning- based numerical SQGs have been developed or are currently available because of the confounding factors mediating the bioavailability of metals. A study was conducted at Dianchi Lake, which is a heavily eutrophicated lake on the Yunnan- Guizhou Plateau, china with the focus on the measurement of partitioning coefficient （Kp） and SQGs derivation and normalization to acid volatile sulfide （AVS）, fine material, and organic carbon. Using new normalization methods, SQGs were formulated for seven metals including copper, zinc, lead, cadmium, chromium, mercury, and arsenic in Dianchi Lake. In Dianchi Lake sediments, the fine material contributed 25.4%-36.0% to the SQG values, with the largest contribution to the SQG value of mercury; AVS contributed 2.9%-75.0% to the SQG values, with the largest contribution to the SQG value of cadmium. This indicated that the fine material and the AVS were the most important controlling factors to the bioavailability of mercury and caximium, respectively. The contribution of total organic carbon （TOC） to the SQG values of copper and leaxi was 3.8% and 7.1%, respectively, indicating that at relatively lower concentrations, the contribution of TOC was not significant. In addition to normalization methods, appropriate procedures for the application of EqPA including sample collection, storage, and analysis are also essential to improve the reliability of SQGs. The normalized Dianchi Lake SQGs were higher than most of the empirically based SQGs developed in North America, but lower than Hong Kong interim SQGs except for cadmium and arsenic.
