Tibetan pig has been given more attention recent years for its ability to survive under adverse *** it is an ideal small experimental animal,for its small body size, extremely similarity with human in anatomy and physiology,*** small intestine is the main absorption place of peptides and free amino acids,which required many transporter *** b mediates apical uptake of basic amino acids,especially lystine,argine and *** aim of current study was to clone Tibetan porcine amino acid transporter bAT（SLC7A9） for comparing the sequences of tibetan and common pig,and investigating the tissue distribution and ontogenetic expression in the small intestine of Tibetan suckling *** Tibetan porcine SLC7A9 gene which cloned from the porcine small intestine was first described, encoding the amino acid transporter bAT. The entire open reading frame（ORF） of the SLC7A9 is 1464 bp and codified 487 amino acid residues,having a higher degree of sequence similarity with the horse counterpart （91.2%） than with monkey（89.5%） or human （88.7%）.The putative protein has 12 putative transmembrane *** this study, SLC7A9 mRNA was detected in brain,kidney, duodenum,jejunum,ileum,heart,liver,lung and muscle from Tibet pigs at 7 and 21 days by ***,we investigated the expression of Tibetan sucking piglets in duodenum, anterior jejunum,posterior jejunum,ileum and kidney from 1 to *** abundance of SLC7A9 mRNA in duodenum and jejunum was highest and lowest,*** has a similar pattern in duodenum and anterior jejunum,which the mRNA level was decreased in front of the sucking period and increased until *** jejunum expression was moving up steadily with age, except *** ileum has the highest expression at d14 and becomes steady after *** mRNA abundance in kidney has the trend in opposite to duodenum,raised until 14 and reduced after *** results showed the pattern of bAT expressed in small intestine of Tibetan pig and thi

Determination of 23 β_2-agonists and 5 β-blockers in animal muscle by high performance liquid chromatography-linear ion trap mass spectrometry

A high performance liquid chromatography-linear ion trap mass spectrometry method using isotope dilution technique has been developed for the simultaneous determination of 23 β2-agonists and 5 β-blockers in animal muscle tissues. Pork and chicken muscle samples were acid hydrolyzed and extracted with 5% trichloracetic acid in water, and then cleaned up using MCX solid phase extraction (SPE) cartridge. Methanol and 0.1% formic acid were used as mobile phases for gradient elution. A Waters AtlantisT3 column was used for separation. ESI positive ion scan mode was used with selective reaction monitoring. 9 β2-Agonists labeled by the deuterium isotope were used as internal standards for quantification. The linear ranges of 23 β2-agonists and 5 β-blockers were 5-200 μg/L, the coefficient of correlation was not less than 0.995, and the limit of detection for each compound in the muscle tissue was below 0.2 μg/kg. The recoveries of each compound in the spiked samples at three levels 5, 10, 20 μg/kg were in the range of 47.3%-123.7%, and the relative standard deviations were in the range of 3.2%-25.7%. The developed method is sensitive and specific for the determination of β2-agonists and β-blockers in pork and chicken muscle samples.

The genetic diversity of two natural populations(M,N) of beijing duck(Anas platyrhynchos) and 11 artificially selected lines of beijing duck(A,B,E-L,O) from china Gold Star Duck Production Ltd.,along with two Cherry Valley duck lines(C and D) from the British Cherry Valley Livestock Division,was evaluated using 18 microsatellite markers covering 16 linkage groups.A phylogenetic tree of the 15 populations of duck,formed of four main branches,was constructed from Nei’s DAgenetic *** mean genetic differentiation index(FST) in all loci,Nei’s standard genetic distance(Ds),and the genetic distance DAbetween the beijing duck and the Cherry Valley duck were 0.075,0.143 and 0.142,*** results demonstrated a high degree of genetic similarity between the two breeds and supported the hypothesis that the Cherry Valley duck was derived from the beijing *** FSTmatrix of seven clusters of beijing duck suggested that the efficiency of selection was not significant to some extent and should be supplemented by marker -assisted selection.

Recent Advances in Swine Amino Acid nutrition

Recent advances in swine protein nutrition are characterized by the development of functional a- mino acids （AA） in regulating fetal and postnatal survival, growth and development. These AA include arginine, glutamine, glutamate, proline, leucine, cyste- ine and tryptophan. Due to limited knowledge on AA nutrition, pork producers have traditionally paid little attention to supplementing the arginine family of AA to swine diets. Results of recent studies indicate that functional AA serve important regulatory functions in nutrient metabolism, protein turnover, and immune function, therefore enhancing efficiency of feed utili- zation by pigs. The underlying mechanisms include activation of nitric oxide, mammalian target of rapam- ycin, gaseous signaling, and AMP-activated protein ki- nase pathways, as well as anti-oxidative function. Di- etary supplementation with arginine, glutamine, pro- line or leucine to weanling piglets enhances theirgrowth performance. Arginine or glutamine is also ef- fective in increasing milk production by lactating sows. Furthermore, supplementing arginine to the diet of pregnant gilts between days 30 and 114 of gestation increases the number of live-born piglets and litter birth-weight. Availability of feed-grade functional AA holds great promise for improving animal health and nutrient utilization in pig production worldwide. Addi- tionally, feedstuffs of animal origin [ e. g. , blood meal （ ring dried ）, feather meal （ hydrolyzed ）, meat and bone meal, porcine protein meal, and poultry by-prod- uct meal （both feed- and petfood-grades） ] are excel- lent and cost-effective sources of both essential and functional AA for formulating balanced swine diets. New knowledge on AA nutrition provides a much needed scientific basis for revising the next edition of swine nutrient requirements.

＜正＞Each country has its own feeding standard, e. g. national Research Council（NRC） Requirements of United States and agricultural Research Council（ARC） Requirements of United Kingdom. Poultry Nutrient Requirements of NRC is one of the most influential feeding standards in the world. Since the first edition was published in 1944, it was revised in 1954、1960、1962、1966、1971、1977, 1984 and 1994 respectively. Frequent revision of Poultry Nutrient Requirements reflected the need of development of backyard chicken raising to modem poultry industry. It also reflected continually in-depth studying and gradually perfecting process of animal nutrition feed science, poultry breeding science and all the other relevant sciences during the past forty years.

肌肉生长抑制素(myostatin,MSTN)属于转化生长因子-β(transforming growth factor-β,TGF-β)超家族,主要功能为负向调节骨骼肌的生长.肌肉生长抑制素基因敲除小鼠肌肉出现显著增加,而将干涉该基因的短发夹RNA注射并电击转化入大鼠胫前肌则引起肌肉重量、肌纤维以及MHCⅡ表达的增加.通过与小鼠肌肉生长抑制素基因表达载体共转染HEK293细胞,筛选到两条能够高效抑制小鼠肌肉生长抑制素基因表达的小干涉RNA.构建了这两条小RNA的表达载体Mst-shRNA1和Mst-shRNA2,用其分别转染小鼠C2C12成肌细胞,并通过G418药物筛选和流式细胞仪富集整合了短发夹RNA表达载体的阳性细胞.通过采用Real-time PCR和Western blot分析,检测到在分别整合了Mst-shRNA1和Mst-shRNA2的C2C12细胞中,内源性肌肉生长抑制素基因的mRNA水平分别下降了10.2%和35.5%,蛋白质表达则分别下降了29.3%和64.7%.同时,在这两组中MyoD的表达上升了24.4%和40.4%,证明通过RNA干涉实现的肌肉生长抑制素基因的抑制导致了下游MyoD基因表达的上调.这些结果表明内源性产生的肌肉生长抑制素基因小干涉RNA能够有效抑制该基因的表达.这种以RNA干涉技术为基础促进肌肉生长的方法为遗传育种提供了一种新技术,能够在对家畜经济性状的改良中发挥重要作用.

Avian influenza virus（AIV） nonstructural 1（NS1） gene was amplified by real-time polymerse chain reac tion（RT-PCR） and inserted into pET28a, then transformed into E. coli BL21（DE3） competent cell. With the induction of isopropyl-β-D-thiogalactoside（IPTG） and the purification of Ni-NTA column, we finally obtained purified NS1 protein. T7-phage display system was used to screen the proteins that interacted with NS1 from lung cell cDNA li brary. The selected positive clones were identified by DNA sequencing and analyzed by BLAST program in Gene Bank. Two proteins were obtained as NS1 binding proteins, Homo sapiens nucleolar and coiled-body phosphoprotein 1（NOLC1） and Homo sapiens similar to colon cancer-associated antigen. By co-immunoprecipitation and other me thods, Homo sapiens NOLC1 was found to interact with the NS1 protein, the results would provide the basis for fur ther studying biological function of NS1 protein.
