To clarify the phylogenetic relationships and species status of Pneumocystis, the 5.8S rRNA gene and the internal transcribed spacers (ITS, 1 and 2) of Pneumocystis rRNA derived from rat, gerbil and human were amplified, cloned and sequenced. The genetic distance matrix of six Pneumocystis species compared with other fungi like Taphrina and Saccharomyces indicated that the Pneumocystis genus contained multiple species including Pneumocystis from gerbil. The phylogenetic tree also showed that Pneumocystis from human and monkey formed one group and four rodent Pneumocystis formed another group. Among the four members, Pneumocystis wakefieldiae was most closely related to Pneumocystis murina and Pneumocystis carinii, and was least related to gerbil Pneumocystis.

Objective Hyperbranched Rolling circle amplification(HRCA),a simple,economical, isothermal method was optimized and set up to test Mycobacterium tuberculosis(MTB)in clinical sputum *** The sequences of the 5’ terminal and 3’ terminal of the padlock probe was designed according to the sequences of Insert Sequence 6110(IS6110),which was specific for the Mycobacterium tuberculosis *** padlock probe was used in HRCA to detect the sequence of IS6110 in H37Rv and clinical sputum *** Through the optimization of HRCA,the sensitivity of HRCA in detection of purified DNA from H37Rv was 10 aM/ml,and the sensitivity in detection of H37Rv culture suspension was 10 cfu/ *** testing the specificity of HRCA,the padlock probe was used to detect Mycobacterium other than tuberculosis(MOTT) included ***,***,***,***, ***,***,*** and Mycobacterium intercellulare,all the results were negative. The sensitivity in directly detection 42 clinical sputum samples is 73.81%(31/42),which is similar to real-time quantitative PCR(78.57%,33/42) and higher than smear microcopy(42.86%,18/42),which the difference was statistical *** The establishment of HRCA in detection of specific gene of Mycobacterium tuberculosis is useful for development of a cost-effective and reliable method in tuberculosis diagnosis.

The infectious laryngotracheitis virus (ILTV) glycoprotein G (gG) gene of E3 and Zhonghai strains was cloned, sequenced and compared with the gG gene of other Type I animal herpesviruses. To find the localization and the function of the gG in the infected cells, the 35 kD fusion pro-tein (His-GG) was expressed by inserting the coding region of gG except for the signal peptide into pET30a (+). After puri-fication of the His-GG fusion protein, the rats’ antibody to the His-GG was prepared and purified by using the protein G Sepharose. Results of laser scanning confocal microscopy (LSCM) detection showed that the ILTV gG was in the peri-nuclear region and membrane of chicken embryo liver (CEL) and kidney (CEK) cells, and that the gG accumulated more in the coalescent part than in the other parts of the adjacent CEL or CEK cells. The plaque size and the one-step growth curve tests suggested that the ILTV gG was required for viral growth by cell-to-cell direct infection in tissue-cultured CEL cells.

目的世界银行贷款中国结核病控制项目首次将全间歇短程化疗（短化）方案［２Ｈ３Ｒ３Ｚ３Ｓ３（Ｅ３）／４Ｈ３Ｒ３］应用于国家结核病控制规划，取得了满意的近期疗效。为评价方案的有效性，特进行远期效果观察。方法对１２个项目省、自治区５６个示范县１９９２年全年登记的新发涂阳患者，在治愈后２年进行了结核分支杆菌涂片检查，以了解细菌学复发情况。结果４４００例治愈患者经痰涂片检查，发现１３５例痰菌复阳，复发率为３．１％。结论本研究与国内外短化每日疗法与继续期间歇的６个月短化方案相比，其复发率相近。证明本方案治愈率高（复发率在３％左右），不失为一个可优先推广应用于国家结核病规划的化疗方案之一。

Migration inhibitory factor-related protein 14 (MRP14) is one of calcium-binding proteins, referred as S100A9. The heterodimeric molecule formed by MRP14 with its partner MRP8 (S100A8) is the major fatty acid carrier in neutrophils. The MRP8/14 complex has been also implicated in the intracellular transport of arachidonic acid and its precursors in keratinocytes. We show here the involvement of MRP14 in human esophageal cancer. In an initial study, mRNA differential display-reverse transcription polymerase chain reaction (DD-PCR) was performed with two esophageal carcinomas, one esophageal adenocarcinoma and matched normal adjacent mucosa. DD-PCR with the arbitrary primer OPA3 showed that one cDNA band was highly expressed in normal tissues, but disappeared or substantially decreased in tumor counterparts. It was later identified to be the 3’-end of migration inhibitory factor-related protein 14 (MRP14). AIM:To investigate the relationship between human papillomavirus (HPV) infection and concurrent esophagus and gastric cardia cancer from the same patient (CC) and examine the significance of P16 INK4A protein ***:Polymerase chain reaction was used to detect the presence of HPV type16 (HPV16).The expression of P16 INK4A protein was detected using ***:Among the CC specimens,HPV16-DNA was found in eight cases of esophageal squamous cell carcinoma (ESCC) and five cases of gastric cardia adenocarcinoma (GCA),respectively (47% vs 29%),and two of both ESCC and GCA.P16 INK4A was highly expressed in both ESCC and *** the HPV-associated positive CC,higher P16 INK4A expression was observed in the GCA than in the ESCC (75% vs 25%,P < 0.05).CONCLUSION:HPV16 as a correlated risk factor may play an important role in the development of ESCC and GCA.P16 INK4A may be a screening index in the HPVassociated carcinoma of gastric cardia.

目的考核利福喷丁（Ｌ）的疗效；缩短疗程或全程间歇以减少用药次数；观察全程应用吡嗪酰胺（Ｚ）对疗效及毒副反应的影响。方法以利福平（Ｒ）为对照，采用５个月疗程方案（Ⅰ组２ＳＨＲＺ／３Ｒ２Ｈ２Ｚ２，Ⅱ组２ＳＨＲＺ／３Ｌ１Ｈ２Ｚ２）、６个月全间歇方案（Ⅲ组２Ｓ３Ｈ３Ｒ３Ｚ３／４Ｌ１Ｈ２Ｚ２，Ⅳ组２Ｓ３Ｈ３Ｒ３Ｚ３／４Ｌ１Ｈ２Ｅ２），观察Ｚ的全程应用结果，巩固期以乙胺丁醇（Ｅ）为对照。３６６例初治菌阳肺结核随机分入以上４组。结果（１）３３９例完成疗程者中３２９例治疗成功，满疗程时痰菌阴转率Ⅰ～Ⅳ组分别为９７０％、９４１％、１０００％、９７２％。Ｘ线病灶有效率依序为９６０％、９７６％、１０００％和９４４％。５个月组与６个月组空洞关闭率分别为７７％及７６％。各组相互比较均无显著性差异（Ｐ＞０．０５），未见严重副作用。（２）３０５例完成３年随访，Ⅰ、Ⅱ、Ⅲ、Ⅳ组细菌学加Ｘ线复发分别为２、３、６和３例。结论本研究结果进一步证明Ｌ是长效、高效、安全、便于督导的新药；巩固期用Ｚ无必要；现有基本药物合理联用有可能缩短疗程为５个月，值得进一步研究。
