Engineering infectious foot-and-mouth disease virus in vivo from a full-length genomic cDNA clone of the A/AKT/58 strain

Two full-length genomic cDNA clones, pTA/FMDV and pCA/FMDV, were constructed that contained three point-mutants [A174G and A308G (not present in pTA/FMDV); T1029G] in the genome compared with the wild type A/AKT/58 strain of foot-and-mouth disease virus. These two viruses were rescued by co-transfection of pCA/FMDV with pCT7RNAP, which can express T7 RNA polymerase in BHK-21 cell-lines, or by transfection of the in vitro transcribed RNA. Their biological properties were analyzed for their antigenicity, virulence in suckling-mice (LD50) and growth kinetics in BHK-21 cells. The in vivo rescued viruses showed high pathogenicity for 3-day-old unweaned mice (LD50=10?7.5). However, the in vitro transcribed RNA derived from pTA/FMDV had lower pathogenicity for suckling-mice (LD50=10?6), and the in vivo transcribed RNA recovered from pCA/FMDV co-transfected with pCT7RNAP showed no significant differences from the wild type virus. These data showed that recovery of the infectious foot-and-mouth disease virus directly from the use of in vivo techniques was better than from in vitro methods. Furthermore, the reverse genetic procedure technique was simplified to a faster one-step procedure based on co-transfection with pCT7RNAP. These results suggest that in vivo RNA tran- scripts may be more valuable for engineering recombinant foot-and-mouth disease virus than in vitro RNA transcripts, and may contribute to further understanding of the biological properties, such as replication, maturation and quasispecies, of the foot-and-mouth disease virus.

Chiroptera are the natural reservoirs of lyssaviruses and play an important role in the transmission of animal and human *** this present study,the prevalence of serum antibodies against rabies virus(RABV) in bats was investigated in china.A total of 685 bats belonging to 8 species in 4 families collected from 10 sites of 4 southern provinces were tested by the indirect fluorescent antibody test(iFAT) using FITC-conjugated protein A/G mixture and viral neutralization *** total positive rate of serum antibodies against RABV was 2.2%(15/685),involving 3 of the 8 species *** is the first report showing the seroprevalence of rabies virus in Chinese bats and the results obtained suggest that bats may play a role in the ecology of lyssaviruses in china and further surveillance for the presence of lyssaviruses in bats should be undertaken throughout china.

Multi-Residue Determination of Eight Anabolic Steroids by GC-MS in Muscle Tissues from Pigs

A gas chromatography-mass spectrometry （GC-MS） method to determine eight anabolic steroids （diethylstilbestrol, methyltestosterone, norethindrone, 17α-ethynylestradiol, estradiol, 6α-methyl-17α-hydroxy-progesterone, estradiol benzoate, and chlormadinone acetate） was developed. Muscle samples were extracted with liquid-liquid extraction and clean-up was performed in two steps, the extracts obtained were derivatized with heptafluorbutyric （HBF） anhydride and analyzed by GC-MS. In the above method, the linear scope was 2.5-50 μg kg^-1. The range of the recoveries was 78.5-148% for diethylstilbestrol, 70.8-109% for methyltestosterone, 69.8-87.2% for norethindrone, 67.7-120% for 17α-ethynylestradiol, 82.8-103% for estradiol, 70.3-99.2% for 6α-methyl-17α-hydroxy-progesterone, 73.0-104% for estradiol benzoate, and 72.9- 91.8% for chlormadinone acetate. The range of the coefficients of variation within batches was 0.4-12%; the range of the coefficients of variation between batches was 6.4-11%. The limit of detections and the detection capability were 0.99 and 3.30 μg kg^-1 for diethylstilbestrol, 1.05 and 3.50 μg kg^-1 for methyltestosterone, 1.19 and 3.97 μg kg^-1 for norethindrone, 0.94 and 3.13 μg kg^-1 for 17α-ethynylestradiol, 1.45 and 4.83 μg kg^-1 for estradiol, 1.56 and 5.20 μg kg^-1 for 6α-methyl-17α- hydroxy-progesterone, 1.92 and 6.40 μg kg^-1 for estradiol benzoate, and 2.41 and 8.03 μg kg^-1 for chlormadinone acetate, respectively. These results showed that the method was widely available, accurate, and sensitive.

Clostridium perfringens（***）is an important pathogen of animals and humans and is the causative agent of necrotic enteritis（NE）in *** study focused on the typing of intestinal *** isolates（n = 78,54 from chickens with NE,24 from healthy chickens）from china’s southern guangdong *** isolates were classified as type A using multiplex PCR *** recently described toxin gene associated with NE in chickens,netB,was found in 21（26.9 %）of the isolates（18 from chickens with NE,and 3 from unrelated chickens with no evidence of NE）.The in vitro susceptibility results showed that ceftriaxone showed highest activity with lowest MIC50 values（0.25-0.5 μg/ml）followed by cefotaxime（0.25-4 μg/ml）,penicilin（0.5-2μg/ml）.However,most of the isolates were resistant to lincomycin,tetracycline,metronidazole,and neomycin as evident by the high ***,70.5% of the isolates（n=55）were simultaneously resistant to more than four antibiotics,and 30.8% of the isolates（n=24）were resistant to all the antimicrobials *** our knowledge,this is the first study of the prevalence of *** in broiler chickens in china,and the first to examine the isolates for the recently identified novel toxin genes,netB.

＜正＞Immunity against chicken coccidiosis by in ovo vaccination of Eimeria necatrix invasive stages was investigated. The fertile Lingnanhuang broiler eggs were injected through amniotic sac on day 15, 17 or 19 of incubation with oocysts, sporocysts or sporozoites of ***, respectively. Vaccination doses were in the range of 3×103-1×104 oocysts, 4×103-4×104 sporocysts, or 8×103-8×104 sporozoites per egg. Hatching rates were generally unaffected when vaccinated. Tumor suppressor p53,the major cellular defense against tumor development,has recently been implicated in host antiviral *** studies have shown that p53 was induced at the apoptotic stage of influenza virus-infected ***,we found that p53 was induced not only at the apoptotic stage but also at the beginning phase of infection, showing a biphasic pattern with a first transient elevation apparent at the beginning phase of infection and a second elevation observable at the middle-late phase of *** up-regulation of p53 was independent of increased p53 transcription, but dependent on virus adsorption and *** increased p53 was active and able to transactivate its downstream target genes,such as interferon regulatory factor 9(IRF9) and *** our knowledge,this is the first report to describe a biphasic pattern of p53 accumulation in influenza virus-infected cells.

To investigate the influence of selenium on body weight and immune organs index in ducklings administrated with aflatoxin B(AFB),ninety 7-day-old ducklings were randomly divided into three groups(groupsⅠ-Ⅲ).groupⅠwas used as a blank ***Ⅱwas administered with AFB(0.1 mg/kg body weight).groupⅢwas administered with AFB](0.1 mg/kg body weight) plus selenium(sodium selenite,1 mg/kg body weight).All treatments were given once daily for 21 *** showed that the ducklings’ bursa of fabricius and thymus index,body weight in groupⅡsignificantly decreased when compared with groupⅠ(P<0.01).Furthermore,the spleen index significantly decreased(P<0.01).However,the ducklings’ bursa of fabricius and thymus index,body weight in groupⅢducklings significantly increased when compared with groupⅡ(P<0.05).In addition,the spleen index significantly decreased(P<0.01).These results revealed that AFB]significantly affect ducklings’ growth and immune organ ***,sodium selenite could significantly ameliorate the negative effect induced by AFB,.

Avian Infectious Bronchitis Virus(IBV) strain D41 is an attenuated strain of an isolate from guangdong province, china. In this study, the 3’ end of the D41 genome was amplified by RT-PCR. After sequencing, we got the 3’ end of the D41 genome(1529 bases) including the nucleocapsid(N) gene and part of 3’ untranslated region(UTR). The deduced sequence of the D41 N gene was 1227bp. It predicted a polypeptide of 409 amino acids with a molecular weight of 45KDa. It was identical in size and had a high degree of amino acid identity(91.2—98.5％) with some previously published sequencing data of other IBV strains. But the variation of nucleotide sequence occurred throughout the N gene and involved regions previously identified

The aim of the study was to investigate the effect of selenium on hepatic mitochondrial antioxidant capacity in ducklings administrated with aflatoxin B(AFB).90 seven-day-old ducklings were randomly divided into three groups(groupsⅠ-Ⅲ).groupⅠwas used as a blank ***Ⅱwas administered with AFB(0.1 mg/kg body weight).groupⅢwas administered with AFB(0.1 mg/kg body weight) plus selenium(sodium selenite,1 mg/kg body weight).All treatments were given once daily for 21 *** results showed that the activities of mitochondrial superoxide dismutase(SOD),catalase(CAT),glutathione peroxidase(GSH-Px) and glutathione reductase(GR) in groupⅡducklings significantly decreased when compared with groupⅠ(P<0.01).Furthermore,the content of hepatic mitochondrial malondialdehytra(MDA) significantly increased(P<0.01).However,the activities of hepatic mitochondrial SOD,CAT,GSH-Px and GR in groupⅢducklings significantly increased when compared with groupⅡ(P<0.05).In addition,the content of hepatic mitochondrial MDA significantly decreased(P<0.01).These results revealed that AFB significantly induced hepatic mitochondrial antioxidant ***,sodium selenite could significantly ameliorate the negative effect induced by AFB.
