Effects of Changtai granules, a traditional compound Chinese medicine, on chronic trinitrobenzene sulfonic acid-induced colitis in rats

AIM: To study the effects of Changtai granules (CTG), a traditional compound Chinese medicine, on chronic trinitrobenzene sulfonic acid-induced colitis in rats. METHODS: Healthy adult Sprague-Dawley (SD) rats of both sexes, weighing 250-300 g, were employed in the present study. The rat colitis models were induced by 2, 4,6-trinitrobenzene sulfonic acid (TNBS) enemas at a concentration of 100 mg/kg in 50% ethanol. The experimental animals were randomly divided into dexamethasone (DX) treatment, CTG treatment, and model control groups, which were intracolicly treated daily with DX (0.2 mg/kg), CTG at doses of 2.9, 5.7 and 11.4 g crude drug/kg, and the equal amount of saline respectively from 6 h following induction of the colitis in rats inflicted with TNBS to the end of study. A normal control group of rats treated without TNBS but saline enema was also included in the study. After 3 wk of treatment, the animals were assessed for colonal inflammatory and ulcerative responses with respect to mortality, frequency of diarrhea, histology and myeloperoxidase activity (MPO).RESULTS: The therapeutic effect of CTG on ulcerative colitis (UC) was better than DX. CTG effectively inhibited the activity of granulocytes, macrophages and monocytes in a dosedependent manner. Also it reduced MPO and formation of inflammation in colonic mucosal tissue. Furthermore, administration of CTG significantly prevented body mass loss and death, and decreased frequency of diarrhea in UC rats, when compared with the model control group ***: CTG would prove to be an ideal drug for chronic UC, and is warranted to be studied further.

Surgical treatment of pancreatic carcinoma

OBJECTIVE: To evaluate the surgical treatment of pancreatic carcinoma. METHODS: 101 patients with pancreatic carcinoma admitted from 1995 to 2002 were studied retrospectively. Of 83 patients undergoing surgery, 56 (64.5%) were subjected to tumor resection. Whipple' s procedure was performed in 48 patients, extended regional pancreatectomy and autograft intestinal transplantation in 2, combined resection of pancreatic body or tail carcinoma in 6, and 27 patients with unresectable pancreatic carcinoma were given inner drainage. RESULTS: Wound dehiscence and digestive bleeding were found in one patient respectively. Pancreatic fistula was found in one patient undergoing extended regional pancreatectomy and autograft intestinal transplantation. Other patients recovered uneventfully. CONCLUSIONS: Most patients with pancreatic carcinoma could undergo tumor resection. Blood vessel repair or transplantation can be used if the tumor adherent to the superior mesenteric-portal vein. Extended regional pancreatectomy and autograft intestinal transplantation are feasible. Simple inner drainage should be used in the patient whose neoplasm is unresectable.

The Regulating Effect of CCK and Gastrin on Apoptosis of Bile Duct Carcinoma Cells

Objective Probe into the influence and the mechanisms of CCK and gastrin on the apoptosis of bile duct carcinoma cells. Methods By taking beauvericin as the revulsant to the apoptosis of bile duct carcinoma cells, the influence of CCK and gastrin on the apoptosis of bile duct carcinoma cells was investigated by using the techniques such as TUNEL fluorescent staining, stream mode cell detecting instrument and reverse bcl-2 oligonucleotide. Techniques of immunohistochemistry, in situ hybridization, flow cytometry (FCM) , RT-PCR were used to study the roles of apoptosis-related genes bcl-2 and baxResults After beauvericin 40 uM worked for 12 h, the survival rate of QBC939 bile duct carcinoma cells was decreased by 35% ?40% . About 80% of the bile duct carcinoma cells showed various degrees of apoptosis. CCK and gastrin could upregulate the threshold value of the apoptosis of bile duct carcinoma cells, which could be inhibited by L60, L18 and reverse bcl-2 oligonucleotide. In terms of both transcription and translating levels, CCK and gastrin could obviously promote the genetic expression of bcl-2, but had no influence on the genetic expression of bax. Addition of CCK-A receptor or CCK-B/gastr in receptor antagonist could remarkably inhibit the expression of bcl-2 boosted by gastrin-17 and CCK-8S.Conclusion CCK and gastrin inhibited the apoptosis of bile duct carcinoma cells through upregulating the genetic expression of bcl-2. Theoretically, this research has expanded our understanding to the mechanism of CCK and gastrin in controlling the growth of tumors, enriched our view to the mechanism of apoptosis of alimentary tract tumors, and has provided a new thinking for the assistant treatment to bile duct carcinoma cells as well.

In the present study expression of estrogen receptor subtype -α(ERα) and -β (ERβ) in the cerebral cortex, cerebellum, and olfactory bulb was investigated and compared between neonatal (1～3-days-old) and adult (250～350g) rats, using reverse transcription-polymerase chain reaction (RT- PCR). No ERα transcripts were detectable in the adult cerebellum and olfactory bulb, whereas very weak expression of ERα was present in the adult cerebral cortex. No significant difference in ERβ transcripts was detectable between the neonatal and adult rats. While transcripts for both ER subtypes were co-expressed in these brain areas of neonatal rats, although ERα expression was significantly weaker than ERβ. Even in the cerebral cortex known to contain both ER subtypes in adult rats, ERα transcripts in neonatal rats were much higher than in adult. These observations provide evidence for the existence of different expression patterns of ERcα/ERβ transcripts in these three brain areas between the neonatal and adult rats, suggesting that each ER subtype may play a distinct role in the regulation of differentiation, development, and functions of the brain by estrogen.

Endoscopic management of biliary complications after orthotopic liver transplantation

BACKGROUND: Biliary complications are a serious problem in patients after liver transplantation and often require reoperation. This study was conducted to summarize the endoscopic diagnosis and management of biliary complications after orthotopic liver transplantation (OLT). METHODS: From December 2000 to November 2003, twelve endoscopic retrograde cholangiopancreatographies(ERCPs) were performed in 7 patients after OLT at Digestive Endoscopic Center of Changhai hospital in shanghai, china. The therapeutic maneuvers included endoscopic sphincterotomy (EST), biliary stent placement, balloon and basket extraction, irrigation, and nasobiliary tube placement. A retrospective study was made to determine the types of biliary tract complications after OLT. The success of ERCP and therapeutic maneuvers was also evaluated. RESULTS: Biliary tract complications including biliary stricture, biliary leak, biliary sludge, and stump leak of the cyst duct were treated respectively by endoscopic sphincterotomy with sludge extraction, stricture dilation or endoscopic retrograde biliary drainage. Two of the 3 patients with proximal common bile duct stricture were successfully treated with ERCP and stent placement. Four patients with anastomotic stricture and/without bile leak were treated successfully by dilation and stent placement or endoscopic nosobiliary drainage. No severe ERCP-related complications occurred. CONCLUSIONS: ERCP is an effective and accurate approach for the diagnosis of biliary tract complications after OLT, and placement of a stent is a safe initial treatment for biliary complications after liver transplantation.

Skin graft rejection is a typical cellular immune response,mainly mediated by T *** lymphocyte associated antigen 4-immunoglobin (CTLA4-Ig) extends graft survival by blocking the T cellco-stimulation pathway and inhibiting T cell *** investigate the efficacy of CTLA4-Ig in prolong-ing skin graft survival,human CTLA4-Ig (hCTLA4-Ig) was engineered to overexpress in mouse skin bytransgenesis using the K14 *** transcription-polymerase chain reaction (RT-PCR) and West-ern blot assay indicated that the expression of CTLA4-Ig remained skin-specific and relatively constantcompared to the internal control protein,AKT,through seven *** presence and concentrationof the hCTLA4-Ig protein in transgenic mouse sera was determined by enzyme-linked immunosorbent assay(ELISA),and the results indicated that the serum CTLA4-Ig concentration also remained constant *** of transgenic mouse skins grafted onto rat wounds was remarkably prolonged com-pared to that of wild-type skins from the same mouse strain,and remained comparable among all *** suggested that the bioactive hCTLA4-Ig protein was stably expressed in transgenical micethrough at least seven generations,which was consistent with the stable skin-specific CTLA4-Ig *** results demonstrated that the transgenic expression of hCTLA4-Ig in skin driven by the K14 promoterremained constant through generations,and a transgenic line can be established to provide transgenic skinwith extended survival reproducibly.

Aim:The role of caveolin-1(Cav-1)in angiogenesis remains poorly *** endothelial nitric oxide(NO)synthase(eNOS),a caveolin-interacting protein,was demonstrated to play a predominant role in vascular endothelial growth fac-tor(VEGF)-induced *** purpose of our study was to examine therole of Cav-1 and the eNOS complex in NO-mediated ***:Human umbilical vein endothelial cells(HUVEC)were isolated and cultured in 3-Dfibrin gels to form capillary-like tubules by VEGF *** expression ofCav-1 and eNOS was detected by semiquantitative *** HUVEC weretreated with antisense oligonucleotides to downregulate Cav-1 *** and non-infected HUVEC were cultured in fibrin gels in the presenceor absence of VEGF(20 ng/mL)and N~G-nitro-L-arginine methyl ester(L-NAME;5 mmol/L).NO was measured using a NO assay kit and capillary-like tubules werequantified by tubule formation index using the Image J ***:RT-PCRanalysis revealed that Cav-1 levels steadily increased in a time-dependent mannerand reached their maximum after 5d of incubation,but there were no obviouschanges in eNOS mRNA expression in response to VEGF in the fibrin gel ***(20 ng/mL)can promote NO production and the formation of capillary-liketubules,and this promoting effect of VEGF was blocked by the addition ofL-NAME(5 mmol/L).When transduced HUVEC with the antisense Cav-1 oligo-nucleotides were plated in the fibrin gels,the capillary-like tubules were signifi-cantly fewer than those of the non-infected *** capillary-like tubules forma-tion and NO production of transduced HUVEC with the antisense Car-1 oligo-nucleotides cultured in fibrin gels showed no responses to the addition of VEGF(20 ng/mL)and L-NAME(5.0 mmol/L).Conclusion:NO was a critical angiogenicmediator in this ***-1 was essential for NO-mediated angiogenesis andmay be an important target of anti-angiogenesis therapy.
