＜正＞Objective pain is known to be processed by a complex neural network（neuromatrix） in the *** is hypothesized that under pathological state,persistent or chronic pain can affect various higher brain functions through ascending pathways,leading to co-morbidities or mental disability of ***,so far the influences of pathological pain on the higher brain functions are less clear and this may hinder the advances in pain *** In the current study,by virtue of a well-developed multi-electrode array technique,we established an in vitro ’neural network’ model for studying spatiotemporal plasticity of synaptic connection and function in the hippocampal formation（HF） in response to persistent *** waveform of the field excitatory postsynaptic potential（fEPSP）,induced by perforant path electrical stimulation and pharmacologically identified as being activity-dependent and mediated by ionotropic glutamate receptors,was consistently positive-going in the dentate gyrus（DG）,while that in the CA1 was negative-going in shape in naive and saline control *** For the spatial characteristics of synaptic plasticity,persistent pain significantly increased the number of detectable fEPSP in both DG and CA1 areas at the network level and distinctly enhanced the input-output function of individual synaptic efficacy at the cellular *** the temporal plasticity,long-term potentiation produced by theta burst stimulation conditioning was also remarkably enhanced by ***,it is strikingly noted that the shape of fEPSP waveform was drastically deformed or split by the conditioning stimulus under the condition of persistent nociception,while that in naive or saline control state was not *** these changes in synaptic connection and function were further confirmed by the 2-dimentional current source density *** an initial step in dissecting the mechanisms underlying this phenomenon,we performed infrared differential interferen

Objective Melittin is the main peptide in bee *** studies demonstrated that this peptide could cause persistent spontaneous pain,primary heat and mechanical hyperalgesia,and enhance the excitability of spinal nociceptive ***,the underlying mechanism of melittin-induced cutaneous hypersensitivity is still not *** In the present study,melittin was applied topically to acutely dissociated rat dorsal root ganglion(DRG)neurons to examine its effects on the excitability of sensory neurons and the underlying cellular mechanisms using whole-cell patch clamp and calcium imaging *** It was found that melittin induced intracellular calcium([Ca])increases in 60%of small(25μm)and medium(2) of small and medium neurons *** sodium channel blocker tetrodotoxin(TTX)failed to eliminate firing evoked by *** clamp experiments showed that melittin evoked inward currents in sensory neurons in a concentration-dependent *** application of melittin caused increased amplitude of the inward currents. Most of the melittin-sensitive neurons were capsaicin-sensitive,and 65%were isolectin B4(IB4)*** results showed that capsazepine,the TRPV1 receptor inhibitor,completely abolished the melittin-induced inward currents and intracellular calcium *** signaling pathways involved in the melittin-induced inward current were also ***,but not PLC,was involved in producing the melittin-induced inward *** of cyclooxygenase(COX)and lipoxygenase(LOX),two key components of the arachidonic acid metabolism pathway, each partially suppressed the inward current evoked by *** of protein kinase A(PKA),but not of PKC, also abolished the melittin-induced inward *** These results indicate that melittin can directly excite smal

＜正＞Objective In spite of overwhelming evidence derived from human and animal studies showing active involvement of primary somatosensory cortex（S1） in subserving the somatosensory-discriminative aspects of pain,it remains unclear whether persistent pain could produce neuronal and synaptic plasticity in this *** the present study,we sought to determine,at various levels,possible plastic changes in neuronal activities,protein expression（activation） and synaptic connection and transmission in S1 area of rats subjecting to peripheral persistent *** and Results First,our immunohistochemical experiments demonstrated that subcutaneous bee venom injection,a wellestablished animal model of persistent inflammatory pain,could evoke increased populational neuronal activities in the S1 region with predominant localization in layersⅡ-Ⅲ.Subsequently,we used Western blot technique to unravel regionrelated differential activation of mitogen-activated protein kinase subfamily members in spinal cord and S1 area in re- sponse to peripheral persistent pain *** the microscopic synaptic unit level,our whole cell patch clamp recording results showed that persistent painful stimulation in the periphery exerted a significant impact on the intrinsic synaptic activities of cortical neurons,while at the macroscopic synaptic circuit level,combination of multi-electrode array recording with current source density imaging revealed pain-elicited,layer-dependent spatial and temporal plasticity of synaptic connection,transmission and function within the S1 ***,our ongoing work using somatosensoryevoked potentials in vivo has shown nociception specific,long-term potentiation-like phenomenon following bee venominduced peripheral inflammatory *** The present results add a new line of evidence showing multiple levels of plastic changes in the S1 area in response to peripheral persistent nociception.

＜正＞Objective There is substantial evidence supporting the notion that the anterior cingulate cortex（ACC） is an important limbic structure involved in the affective-emotional component of ***,neuronal and synaptic plasticity occurring within the ACC after injury has been proposed to contribute substantially to the pathophysiogenesis of chronic ***,fewer reports have been conducted to investigate simultaneously spatial and temporal features of synaptic plasticity in the ACC in response to persistent pain at cellular and network *** present study was thus undertaken to evaluate,at the local network level,potential changes in synaptic connection and transmission in the ACC following peripheral persistent nociception in *** and Results An initial probe of underlying mechanisms was also attempted at the cellular *** using multi-electrode array recording coupled with two-dimensional current source density imaging,we found that persistent pain produced enlargement of synaptic connection size in the ACC slices, reflecting increased spatial plasticity by pain,with the temporal plasticity,namely long-term potentiation,being unaffected. Subcutaneous injection of bee venom causes long-term neural activation and hypersensitization in the dorsal horn of the spinal cord,which contributes to the development and maintenance of various pain-related *** unique behavioral 'phenotypes' of nociception and hypersensitivity identified in the rodent bee venom test are believed to reflect a complex pathological state of inflammatory pain and might be appropriate to the study of phenotype-based mechanisms of pain and *** this review,the spinal processing of the bee venom-induced different 'phenotypes' of pain and hyperalgesia will be *** accumulative electrophysiological,pharmacological,and behavioral data strongly suggest that different 'phenotypes' of pain and hyperalgesia are mediated by different spinal signaling *** the phenotype-based mechanisms of pain might be useful in development of novel therapeutic drugs against complex clinic pathological pain.

＜正＞It has long been known that the brain governs the body functions with feedforward and feedback neural signals conducted through the peripheral nervous system（PNS）,while it also regulates its own functions through structural connections among different parts（nuclear group） of the central nervous system（CNS）（brain and spinal cord） via ’cables’ formed by nerve *** the PNS,nerve fibers are classified into thickly-myelinated,thinly-myelinated and unmyelinated fibers,while in the CNS it is believed that all of the nerve fibers are concentrically wrapped by myelin sheath,the ’cablecoating’. The major function of myelination is to speed up the conduction of neural signals（series of action potentials） in the nervous *** myelinating cells in the PNS are Schwann cells,while those in the CNS are oligodendroglial cells. In the past century,attention has been mainly paid to the structures and functions of both the nerve cell bodies（soma） and synapses（’contact’ between nerve cells） due to the influence of Cajal’s Doctrine of Neuron,while little attention has been paid to the structures and functions of the ’cables’,especially the ’cable-coating’ in the nervous *** *** is no multiple time-points study describing age-related change in myelination and demyehnation of nerve *** this presentation,I would like to show some new data about the ultrastructural features of the myelinated nerve fibers in both the PNS and the CNS in terms of postnatal development,growth,maturity and aging which are temporally associated with the nature of life *** new findings are summarized as follows:（1） Myelination of nerve fibers in both the PNS and the CNS are not congenital but likely to be acquired with the need of *** electron microscopic observation of both the PNS and CNS in rats with postnatal ages of 1W,2W,3W,4W,8W,28 W,56W,68 W,for instance, no myelinated fibers could be observed in the optic nerve（ON） at 1W when the rat does not open its e

＜正＞Objective According to the different firing properties to a long depolarizing current pulse,two classes of neurons can be identified in the dorsal root ganglion（DRG）.Tonic firing neuron generates multiple action potentials（Aβs）, whereas phasic firing neuron fires a few Aβ***,it is still unclear which type of the two neurons is really involved in mediation of pain *** this report,we identified the functional roles of the two types of neurons by gene regulation of TTX-resistant sodium channels,NaV1.8 and NaV1.9,which have been demonstrated to be differentially expressed in tonic and phasic *** Complete freund’s adjuvant（CFA） was used to induce peripheral inflammatory *** cell patch-clamp recording was used to identify tonic and phasic types of DRG ***-PCR, Western blot and immunocytochemistry were used to evaluate the expression levels of NaV1.8 and *** （1） Under CFA-induced inflammatory pain state（heat and mechanical hypersensitivity）,electrophysiological properties of tonic and phasic neurons were compared and it was found that tonic neurons were with decreased rheobase and increased number of spikes,while phasic type showed no changes in excitability at *** membrane potential was altered in both types of neurons by CFA.（2） RT-PCR,Western blot and immunocytochemical staining showed parallel increase in gene and protein expression level of both NaV1.8 and NaV1.9,suggesting up-regulation of TTX-resistant sodium channels in small to medium-sized DRG cells by CFA.（3） Down-regulation of NaV1.8 and NaV1.9 in DRG cells was established, respectively,by intrathecal（i.t.） administration of antisense oligonucleotides that was confirmed by both gene and protein expression *** CFA-induced decrease in rheobase and increase in spike discharges of tonic neurons were completely restored by down-regulation of NaV 1.8,while electrophysiological properties of phasic neurons were not affected by either NaV 1.8

Objective:Tetrodotoxin-resistant(TTX-R) sodium channels Na1.8 and Na1.9 in sensory neurons were key pain *** with the well reported roles of Na1.8,evaluations of Na1.9 on inflammatory pain is poorly studied by antisense agents inducing specific gene ***,we used molecular,electrophysiological and behavioral methods to examine the effects of antisense oligodeoxynucleotide(AS ODN) targeting Na1.8 and Na1.9 on inflammatory *** Complete Freund’s adjuvant(CFA) was used to induce peripheral inflammatory ***-PCR,Western blot,immunocytochemistry and whole cell patch-clamp were used to evaluate the expressions and functions of Na1.8 and *** Following CFA inflammation treatment,Na1.8 and Na1.9 in rat dorsal root ganglion(DRG) were activated demonstrated in the up-regulations of mRNA and protein expression and increased sodium current *** data demonstrated that Na1.8 was mainly localized in medium and small-sized DRG neurons,whereas Na1.9 was only expressed in small-sized DRG ***(i.t.) delivery of AS ODN was used to down-regulate Na1.8 or Na1.9 expressions confirmed by immunohistochemistry and Western ***,behavioral tests showed that only Na1.8 AS ODN,but not Na1.9 AS ODN could reverse CFA-induced heat and mechanical *** Our data indicated that the key pain modulators Na1.8 and Na1.9 played distinct functional roles in the processing of inflammatory pain and antisense oligodeoxynucleotide-mediated blocking of Na1.8 over-expression might point toward a potential treatment strategy for certain type of pathological pain.

＜正＞Objective It is known that chronic pain affects various higher brain functions including perception, emotion,cognition,and ***,few studies have been performed to examine pain-induced synaptic plastic changes in the hippocampal formation（HF）,an important region subserving affective-motivational component of *** previous study has revealed a strong impact of peripheral persistent nociception on synaptic connection, transmission and function in the HF of rats,"in both temporal and spatial ***,the underlying signaling mechanisms for this pain-related spatial and temporal plasticity are still less *** present study attempted to examine potential different roles of the mitogen-activated protein kinase（MAPK）members in mediating this plastic *** Inhibitors targeting extracellular signal-regulated kinase（ERK）,p38 MAPK and c-Jun N-terminal kinase（JNK）were used *** and temporal components of entorhinal-hippocampal synaptic connections and efficacy of rats were recorded by using a newly developed 64-channels（8x8）multielectrode array technique（Med64 system）.Results（1）Pharmacological inhibition of the ERK-and JNK-mediated signaling pathway,at the plateau phase of the long-term potentiation（LTP）,significantly decreased pain-enhanced LTP maintenance whereas similar blockade of p38 MAPK pathway dramatically further increased the potentiation. （2）Regarding the spatial magnification of pain,ERK and p38 MAPK seemed to play opposing roles,with the former positively involved and the latter negatively involved,without any detectable effect of the JNK signaling pathway. Conclusion Together,these results suggest differential roles of the specific members of the MAPK family in mediating pain-associated spatial and temporal plasticity in the HF,which are in good agreement with previous observations.
