Idiopathic dilated cardiomyopathy (IDC) is characterized by dilation and impaired contraction of the left ventricle or both, and it is a relevant cause of heart failure and a common indication for heart transplantation. The major pathogenetic hypothesis in IDC involves autoimmune mediated damage to myocytes. The development of autoimmune inflammatory damage occurs only in patients with a predisposing genetic background. Changes in the immune system concerning cell-mediated and humoral immunity have been detected. The immune system is strictly related to human leukocyte antigen (HLA), which is located on the surface of antigen presenting cells. Its primary function is to restrict T-cell receptors in the process of recognizing auto- or exterior antigen, and thus participates in or mediates immunological recognition, immunological response and immune regulation at various levels. HLA is a genetic marker of susceptibility to autoimmune myocardial damage.~1 In the present study, the HLA-DQA1 and -DQB1 alleles in IDC patients were detected with the techniques of polymerase chain reaction-sequence specific primers (PCR-ssP) to explore the immunogenetic mechanisms involved in the pathogenesis of IDC.

Objective: To retrospectively analyze 85 hospitalizednonoperative patients with severe acute pancreatitis(sAP) and to find out the stages of the ***: We statistically calculated the time of onsetof complications in these patients from 1987 to ***: The 95% confidence interval of total averagefor the complications of acute respiratory distress syn-drome (ARDs), shock and kidney failure was between2 to 4 days, and for encephalopathy, hemorrhage ofthe digestive tract, bacterial infection, fungous infec-tion and abscess between 3 to 6 days, 3 to 5 days, 13to 16 days, 13 to 16 days, and 14 to 23 days respec-tively. The 95% confidence interval of total averagein 18 deaths (21%) was between 5 to 6 days. ARDs,kidney failure, and shock occurred within 4 days, en-cephalopathy within 6 days (average 4.8±0.9 days),abscesses after 14 days, systemic bacterial infectionand fungous infection within half a month (average14.6±1.1days, 14.8±0.9days respectively), and deathwithin 6 ***: According to the time of the occurrence ofcomplications, we divide the courses of the disease intothree stages: early phase(first 4 days) with ARDs,kidney failure, shock, encephalopathy and hemorrhageof the digestive tract; middle phase (5-15 days) withbacterial infection and fungous infection; late phase(15 days after the onset) with abscess.

AIM: To monitor the early responses to irradiation in primary and metastatic colorectal cancer (CRC) with 18 Ffluorothymidine ( 18 F-FLT) and 18 F-fluorodeoxyglucose ( 18 F-FDG) small-animal position emission tomography (micro-PET). METHODs: The primary and metastatic CRC cell lines, sW480 and sW620, were irradiated with 5, 10 and 20, l l l , Gy. After 24 h, the cell cycle phases were analyzed. A dual-tumor-bearing mouse model of primary and metastatic cancer was established by injecting sW480 and sW620 cells into mice. micro-PET with 18 F-FLT and 18 F-FDG was performed before and 24 h after irradiation with 5, 10 and 20 Gy. The region of interest (ROI) was drawn over the tumor and background to calculate the ratio of tumor to non-tumor (T/NT) in tissues. Immunohistochemical assay and Western blotting were used to examine the levels of integrin β3, Ki-67, vascular endothelial growth factor receptor 2 (VEGFR2) and heat shock protein 27 (HsP27). REsULTs: The proportion of sW480 and sW620 cells in the G 2 -M phase was decreased with an increasing radiation dose. The proportion of sW480 cells in the G 0 -G 1 phase was increased from 48.33% ± 4.55% to 87.09% ± 7.43% (P sW620 cells in the s-phase was elevated from 43.57% ± 2.65% to 66.59% ± 7.37% (P = 0.021). In micro-PET study, with increasing dose of radiation, 18 F-FLT uptake was significantly reduced from 3.65 ± 0.51 to 2.87 ± 0.47 (P = 0.008) in sW480 tumors and from 2.22 ± 0.42 to 1.76 ± 0.45 (P = 0.026) in sW620 tumors. 18 F-FDG uptake in sW480 and sW620 tumors was reduced but not significantly (F = 0.582, P = 0.633 vs F = 0.273, P = 0.845). Dose of radiation was negatively correlated with 18 F-FLT uptake in both sW480 and sW620 tumors (r = -0.727, P = 0.004; and r = -0.664, P = 0.009). No significant correlation was found between 18 F-FDG uptake and radiation dose in sW480 or sW620 tumors. HsP27 and integrin β3 expression was higher in sW480 than in sW620 tumors. Backgrounds:The anti-cancer effects of arsenic trioxide（ATO） are mainly via reactive oxygen species（ROs） generation,while ROs may induce the expression of heme oxygenase-1（HO-1）,a protective molecule against oxidative *** study was to investigate whether inhibition of HO-1 could enhance oxidative damage induced by ATO in glioma ***:HO-1 expression in glioma cells was modulated by the addition of its inducer cobalt protoporphyrin IX（CoPPIX）,its inhibitor zinc protoporphyrin IX（ZnPPIX）,or siRNA-mediated knockdown of nuclear factor（erythroid-derived 2）-like 2（Nrf2）.Cell death fraction,mitochondrial membrane potential（MMP） loss,and ROs generation were examined to show the occurrence of oxidative damage in U251MG ***:Our results clearly revealed that ATO induced cell death,MMP loss,ROs generation,and HO-1 expression in U251MG cells,all of which were significantly inhibited by ROs scavenger L-N-acetylcystein（LNAC）.CoPPIX induced HO-1 expression,and protected from ATO induced cell death and ROs generation,while ZnPPIX led to a significant increase in cell death and ROs generation induced by *** addition,Nrf2 knockdown strongly inhibited HO-1 expression induced by ATO,and significantly enhanced ATO induced oxidative ***:Our results for the first time demonstrated that HO-1 could play an important role in protection against ATO induced oxidative stress in glioma,and combination of HO-1 inhibition or Nrf2 knockdown and ATO had synergistic effects on glioma treatment.

Primary and secondary malignant liver cancer are some of most common malignant tumors in the world. Chemotherapy and radiotherapy are not very effective against them. surgical resection has been considered the only potentially curtive option, but the majority of patients are not candidates for resection because of tumor size, location near major intrahepatic blood vessels and bile ducts, precluding a margin-negative resection, cirrhotic, hepatitis virus infection or multifocial. Radiofrequence ablation (RFA), which is a new evolving effective and minimally invasive technique, can produce coagulative necrosis of malignant tumors. RFA should be used percutaneously, laparascopically, or during the open laparotomy under the guidance of ultrasound, CT scan and MRI. RFA has lots of advantages superior to other local therapies including lower complications, reduced costs and hospital stays, and the possibility of repeated treatment. In general, RFA is a safe, effective treatment for unresectable malignant liver tumors less than 7.0 cm in diameter. We review the principle, mechanism, procedures and experience with RFA for treating malignant liver tumors.

AIM: To determine the effect of cis -9, trans -11-conjugated linoleic acid (c9, t11-CLA) on the cell cycle of gastric cancer cells (sGC-7901) and its possible mechanism in inhibition cancer growth. METHODs: Using cell culture and immunocytochemical techniques, we examined the cell growth, DNA synthesis, expression of PCNA, cyclin A, B(1), D(1), p16(ink4a) and p21(cip/waf1) of sGC-7901 cells which were treated with various c9, t11-CLA concentrations (25, 50, 100 and 200 micromol.L(-1))of c 9, t 11-CLA for 24 and 48h, with a negative control (0.1% ethane). REsULTs: The cell growth and DNA synthesis of sGC-7901 cells were inhibited by c9, ***-7901 cells. Eight day after treatment with various concentrations of c9, t11-CLA mentioned above, the inhibition rates were 5.92%, 20.15%, 75.61% and 82.44%, respectively and inhibitory effect of c9, t11-CLA on DNA synthesis (except for 25 micromol.L, 24h) showed significantly less (3)H-TdR incorporation than that in the negative controls (Pstaining demonstrated that sGC-7901 cells preincubated in media supplemented with different c9, t11-CLA concentrations at various times significantly decreased the expressions of PCNA (the expression rates were 7.2-3.0%, 24h and 9.1-0.9% at 48h, respectively), Cyclin A (11.0-2.3%, 24h and 8.5-0.5%,48h), B(1) (4.8-1.8% at 24h and 5.5-0.6% at 48h)and D(1) (3.6-1.4% at 24h and 3.7%-0 at 48h) as compared with those in the negative controls(the expressions of PCNA, Cyclin A, B(1) and D(1) were 6.5% at 24h and 9.0% at 48h, 4.2% at 24h and 5.1% at 48h, 9.5% at 24h and 6.0% at 48h,respectively)(Ps the expressions of P16(ink4a) and P21(cip/waf1), cyclin-dependent kinases inhibitors(CDKI), were increased. CONCLUsION: The cell growth and proliferation of sGC-7901 cell is inhibited by c9, t11-CLA via blocking the cell cycle, with reduced expressions of cyclin A,B(1) and D(1) and enhanced expressions of CDKI(P16(ink4a) and p21(cip/waf1)).
