Insulin-responsive GLUT4(glucose transporter 4) translocation plays a major role in regulating glucose uptake in adipose tissue and *** or not there is a specialized secretory GSV(GLUT4 storage vesicle) pool,and more importantly how GSVs are translocated to the PM(plasma membrane) under insulin stimulation is still under *** the present study,we systematically analyzed the dynamics of a large number of single GLUT4-containing vesicles in 3T3-L1 adipocytes by TIRFM(total internal reflection fluorescence microscopy).We found that GLUT4-containing vesicles can be classified into three groups according to their mobility,namely vertical,stable,and lateral GLUT4-containing *** these groups,vertical GLUT4-containing vesicles exclude transferrin receptors and move towards the PM specifically in response to insulin stimulation,while stable and lateral GLUT4-containing vesicles contain transferrin receptors and show no insulin *** data demonstrate that vertical GLUT4-containing vesicles correspond to specialized secretory GSVs,which approach the PM directly and bypass the constitutive recycling pathway.

Two multigene superfamilies, named V1R and V2R, encoding seven-transmembrane-domain G-protein coupled receptors (GPCRs) have been identified as pheromone receptors in mammals. Three V2R gene families have been described in mouse and rat. Here we screened the updated mouse genome se- quence database and finally retrieved 63 putative functional V2R genes including three newly identified genes which formed a new additional family. We described the genomic organization of these genes and also characterized the conservation of mouse V2R protein sequences. These genomic and se- quence information we described are useful as part of the evidence to speculate the functional domain of V2Rs and should give aid to the functionality study in the future.

To study the historical temperature variation in Yili of Xinjiang,the tree-ring chronologies of maximum density(MXD) and tree-ring width(TRW) were established with the Schrenk Spruce(Picea schrenkiana *** Mey) collected from six sites to analyze the characteristics and ability of response to climate *** results suggest that the MXD chronology of the Schrenk Spruce from different sampling sites responded to climate change well and were positively correlated with the mean maximum temperature and the mean temperature from April to August in the study *** mean maximum temperature anomaly from April to August,spanning the years from 1848 to 2000,was reconstructed by three MXD chronologies which were selected by stepwise *** reconstructed function was stable and explained 56.2% of the *** reconstructed results indicated a cold span of 153 years in spring and summer from the beginning of the 1950s to the beginning of the 1970s in the Yili *** strongly increasing tendency was detected in the mean maximum temperature anomaly from April to August during this reconstructed period.

The Space multi-band Variable Object Monitor(SVOM) is a proposed chinese astronomical satellite,dedicated to the detection,localization and measurement of gamma-ray bursts(GRBs) on a cosmological *** efficient algorithm is developed for the purpose of onboard star extraction from the CCD images obtained with the Visible Telescope(VT) onboard the *** CCD pixel coordinates of the reference stars will be used to refine the astronomical position of the satellite,which will facilitate triggering rapid ground-based follow-up observations of the *** this algorithm,the image is divided into a number of grid cells and the "global" pixel-value maximum within each cell is taken as the first-guess position of a "bright" *** correct center position of a star is then computed using a simple iterative *** two additional strategies,i.e.,scanning the image only by even(or odd) lines or in a black-white chess board mode,we propose to further reduce the time to extract the *** examine the efficiency of the above algorithms,we applied them to the experimental images obtained with a ground-based *** find that the accuracy of the astronomical positioning achieved by our method is comparable to that derived by using the conventional star-extraction method,while the former needs about 25 times less CPU time than the *** will significantly improve the performance of the SVOM VT mission.

High-spin states in 161Er have been studied experimentally via the 150Nd(16O, 5n) reaction at a beam energy of 86 MeV. Three rotational bands built on the 5/2+[642], 3/2-[521], and 11/2-[505] configurations have been extended up to high-spin states, and particularly, the α = -1/2 branch of the ground state 3/2-[521] band has been revised significantly. It is found that signature inversion occurs in the 3/2-[521] band after the band crossing in 161Er. The systematics of the signature inversion associated with the 3/2-[521] configuration in the rare-earth region is discussed. The band properties are analyzed within the framework of a triaxial particle-rotor model, and a triaxial deformation is proposed to the 3/2-[521] band.

The flagella master regulatory gene flhDC of Yersinia pseudotuberculosis serotype III (YPIII) was mutated by deleting the middle region; replaced by a tetracycline resistant gene,; the subsequent mutant strain named YPIIIΔflhDC was obtained. Swimming assay showed that the swimming motility of the mutant strain was completely abolished. The promoter region of the flagella second-class regulatory gene fliA was fused with the lux box,; was conjugated with the mutant; the parent strains respectively for the first cross. LUCY assay result demonstrated that flhDC regulated the expression of fliA in YPIII as reported in E. coli. Biofilm formation of the mutant strain on abiotic; biotic surfaces was observed; quantified. The results showed that mutation of flhDC decreased biofilm formation on both abiotic; biotic surfaces,; abated the infection on Caenorhabdtis elegans. Our results suggest that mutation of the flagella master regulatory gene flhDC not only abolished the swimming motility, but also affected biofilm formation of YPIII on different surfaces. The new function of flhDC identified in this study provides a novel viewpoint for the control of bacterial biofilm formation.

Analyses of rare earth and trace element concentrations of native sulfur samples from the Kueishantao hydrothermal field were performed at the Seafloor Hydrothermal Activity Laboratory of the Key Labo-ratory of Marine Geology and Environment, Institute of Oceanology, chinese academy of sciences. Using an Elan DRC II ICP-MS, and combining the sulfur isotopic compositions of native sulfur samples, we studied the sources and formation of a native sulfur chimney. The results show, when comparing them with native sulfur from crater lakes and other volcanic areas, that the native sulfur content of this chimney is very high (99.96%), the rare earth element (REE) and trace element constituents of the chimney are very low (ΣREE<21×10?9), and the chondrite-normalized REE patterns of the native sulfur samples are similar to those of the Kueishantao andesite, implying that the interaction of subseafloor fluid-andesite at the Kueishantao hydrothermal field was of short duration. The sulfur isotopic compo-sitions of the native sulfur samples reveal that the sulfur of the chimney, from H2S and SO2, originated by magmatic degassing and that the REEs and trace elements are mostly from the Kueishantao ande-site and partly from seawater. Combining these results with an analysis of the thermodynamics, it is clear that from the relatively low temperature (<116 ℃ ), the oxygenated and acidic environment is favorable for formation of this native sulfur chimney in the Kueishantao hydrothermal field.

Layered cathode material LiCo1/3Ni1/3Mn1/3O2 was synthesized by Pechini process, and investigated using X-ray diffraction (XRD), scanning electron microscopy (SEM) and galvanostatic charge/discharge cycling. The sample is well-crystallized and has a phase-pure a-NaFeO2 structure. The particle sizes are uniform, and distributed in the range of 20-200 nm. The initial discharge capacity of the Li/LiCo1/3Ni1/3Mn1/3O2 cell was about 149 mAh·g-1 when it was cycled at a voltage range of 4.5-2.3 V with a specific current of 0.25 mA. The result is better in comparison with solid-state solution method. The synthetic procedure was discussed. Three major reactions: chelation, esterification, and polymerization successively occurred.
