Based on Web and CSCW(Computer Supported Cooperative Work) technologies,a cooperative design platform for architectural analog system for heat,electricity and cold supplies and evaluation was proposed to support those remote and distributive design tasks *** mainly focused on the architectural design and implementation methods,and also presented the system structure and pointed out some key problems in *** platform can help people in different places join the project effectively so that they can allocate resources reasonably, strengthen the communication,decrease the cost and improve their efficiency.

探讨了肝细胞在胚胎干细胞 (EScell)体外诱导分化系统中成熟分化的条件、机制及其鉴定方法 .利用TGF ,bFGF、HGF等细胞生长因子进行BALB/c小鼠ES细胞向肝细胞方向的定向诱导 .利用反转录聚合酶链反应 (RT PCR)、免疫细胞化学 (ICC)和放... 详细信息

探讨了肝细胞在胚胎干细胞 (EScell)体外诱导分化系统中成熟分化的条件、机制及其鉴定方法 .利用TGF ,bFGF、HGF等细胞生长因子进行BALB/c小鼠ES细胞向肝细胞方向的定向诱导 .利用反转录聚合酶链反应 (RT PCR)、免疫细胞化学 (ICC)和放射免疫法 (RIA)动态检测肝细胞特异性基因和蛋白AFP ,ALB ,G6P ,TAT ,CK8,CK1.等在培养体系中的表达 ,并测定肝细胞的尿素合成功能 ,最后测定肝细胞分化率 .结果 ,肝细胞特异基因AFP ,ALB ,G6P和TAT最早分别于第 3、 9、 1.、 1.天表达 ,肝细胞特异蛋白AFP ,CK8,CK1.和ALB最早分别于第 7、 9、 9和 1.天开始表达 .第 1.天开始检测到尿素出现 ,浓度为 8 3μmol/L ,并随培养时间延长而浓度逐渐增加 .最后 ,测得生长因子诱导组肝细胞的分化率为 32 % ,对照组肝细胞分化率为 8% .说明肝细胞可以在ES细胞体外诱导分化系统中出现并成熟分化 ,bFGF、HGF、OSM等可以明显提高细胞分化率和成熟度 ,有望成为解决肝功能替代疗法中细胞来源问题的新希望 .

Aim:To investigate the effects of the wingless-related MMTV integration site 3A(Wnt3a) signaling on the proliferation,migration,and the myogenic and adipogenicdifferentiation of rat bone marrow mesenchymal stem cells (rMSC).Methods:Primary MSC were isolated and cultured from Sprague-Dawley rats and character-ized by flow *** L cells were transfected with Wnt3a cDNA,andconditioned media containing active Wnt3a proteins were *** prolif-eration was evaluated by cell count and 5-bromodeoxyuridine incorporation *** migration of rMSC was performed by using a transwell migration and woundhealing *** myogenic and adipogenic differentiation in rMSC were exam-ined by light microscopy,immunofluorescence,and RT-PCR at different timepoints after myogenic or adipogenic ***:Wnt3a signaling in-duced β-catenin nuclear translocation and activated the Wnt pathway in *** the presence of Wnt3a,rMSC proliferated more rapidly than the control cells,keeping their differentiation ***,Wnt3a signaling induced 2.62%and 3.76% of rMSC-expressed desmin and myosin heavy chain after being cul-tured in myogenic *** myogenic differentiation genes,including Pax7,MyoD,Myf5,Myf4,and myogenin,were activated after Wnt3a *** theother hand,Wnt3a inhibited the adipogenic differentiation in rMSC through thedownregulated expression of CCAAT/enhancer-binding protein alpha (C/EBPalpha)and peroxisome proliferator-activated receptor gamma (PPARgamma).Furthermore,Wnt3a promoted the migration capacity of ***:The results indi-cate that Wnt3a signaling can induce myogenic differentiation in ***3asignaling is also involved in the regulation of the proliferation and migration *** results could provide a rational foundation for cell-based tissuerepair in humans.

A series of prodrugs of amylose grafted with in-domethacin （IND） were mildly synthesized at room tempera-ture and their chemical structures were characterized by FTIR and 1. NMR spectroscopy. The results showed that IND was conjugated with amylose through the ester bond. The loaded IND content in each prodrug was determined to be 1.8, 2.7, 4.8, 1..0, 1..6 and 21.6 mg/g from UV spectroscopy. For in vitro experiment, the simulated medium for stomach and small intestine was a chloric acid solution （pH 1.2）, and a sodium phosphate buffer solution （pH 6.8） containing porcine pan-creatin. A culture fermentation system was used to simulate the colonic condition. The results indicated that IND was hardly released in the simulated stomach medium, but could be controlled released by hydrolysis of the prodrug through α-amylase and lipase in the simulated small intestine and colonic conditions. The pharmacokinetic analysis indicated that Tmax, the time at which a maximum concentration of drug in the plasma, was 2.1.h for free IND. But it was prolonged to 5.2 - 8.8 h for the prodrugs. Meanwhile, the maximum drug concentration in the plasma （Cmax） decreased from 25.8 μg/mL for free IND to 0.5 - 7.0 μg/mL for the prodrugs. It further proved that the prodrug could realize controlled release IND.

Aim:To investigate whether oxidized low-density lipoprotein(ox-LDL)modulatesperoxisome proliferator-activated receptorγ(PPARγ)activity through phosphory-lation in macrophages,and the effect of PPARγ phosphorylation on macroph-ages-derived foam cell ***:After exposing the cultured THP-1.ells to ox-LDL in the presence or absence of different mitogen-activated proteinkinase(MAPK)inhibitors,PPARγ and phosphorylated PPARγ protein levels weredetected by Western *** activity was analyzed using MAP Kinase As-say *** cholesterol accumulation was assessed by Oil red O stainingand cholesterol oxidase enzymatic *** mRNA level of PPARγtarget genewas determined by reverse transcription-polymerase chain reaction(RT-PCR).Results:ox-LDL evaluated PPARγ phosphorylation status and subsequently de-creased PPARγ target gene expression in a dose-dependent manner,ox-LDL alsoinduced MAPK *** of THP- 1.cells with c-Jun N-terminal kinase-,but not p38- or extracellular signal-regulated kinase-MAPK inhibitor,significantlysuppressed PPARγ phosphorylation induced by ox-LDL,which in turn inhibitedfoam cell ***:In addition to its ligand-dependent activation,ox-LDL modulates PPARγ activity through phosphorylation,which is mediatedby MAPK ***γ phosphorylation mediated by MAPK facilitatesfoam cell formation from macrophages exposed to ox-LDL.
