Backgr.und and Objective: Color.ctal cancer is one of the most common malignant cancers in the wor.d. Although the clinicop.thologic staging is the golden cr.ter.on for.the p.ognosis at p.esent, the op.imum p.ognostic cr.ter.a for.color.ctal cancer should be a combination of the clinicop.thologic staging and the molecular.mar.er.. However. ther. ar. cur.ently no molecular.mar.er. available for.the p.ognosis of color.ctal cancer. Sever.l tumor.sup.r.ssor.genes associated with color.ctal cancer have been map.ed at the 18q21-23 r.gion. In this study we detected the fr.quency of loss of heter.zygosity (LOH) at chr.mosome 18q and investigated the r.lationship.between LOH and clinicop.thologic featur.s and its p.ognostic value for.p.tients with stage Ⅱ colon cancer. Methods: A total of 106 samp.es of tumor.tissues and cor.esp.nding nor.al mucosa fr.m p.tients with sp.r.dic stage-Ⅱ colon cancer wer. included in this study. All the samp.es wer. for.alin-fixed and p.r.ffin-embedded. DNA was extr.cted fr.m tumor.tissues and LOH of D18S474, D18S55, D18S58, D18S61 and D18S64 at chr.mosome 18q was analyzed using p.lymer.se chain r.action (p.r., p.lyacr.lamide gel-electr.p.or.sis, and DNA sequencing method. Multivar.ate analysis for.association between LOH and p.ognosis in colon cancer p.tients was p.r.or.ed with Cox p.op.r.ional hazar.s r.gr.ssion model. r.sults: The median follow-up.time was 68 months. For.106 p.tients, 5-year.sur.ival r.te was 83.6%, which was associated with age and gr.ss tumor.typ. (p.= 0.011 and 0.034, r.sp.ctively). Among 102 p.tients who wer. eligible for.LOH infor.ation, the over.ll fr.quency of LOH is 49.0% (50/102), and that of LOH at 5 micr.satellite loci of D18S474, D18S55, D18S58, D18S61, and D18S64 was 30.2% (26/86), 23.4% (18/77), 28.6% (20/70), 35.0% (28/80), and 20.8%(15/72), r.sp.ctively. The occur.ence of LOH was significantly associated with tumor.location and histop.thologic gr.de (p.= 0.023, 0.016 and 0.005, r.sp.ctively). LOH was mor. fr.q

Backgr.und and Objective: Although surgery is the only p.ssible means to cur. gastr.c cancer, the p.ognosis is often discr.p.nt. The Amer.can Joint Committee on cancer/Inter.ational Union against cancer (AJCC/UICC) p.blished the TNM classification of Malignant Tumor. (seventh edition) for.gastr.c cancer r.cently. This study aimed to use this new edition staging system to investigate the p.ognostic factor. for.gastr.c cancer. Methods: The clinicop.thologic data of 980 p.tients with gastr.c cancer tr.ated by sur.ical r.section in our.hosp.tal between Januar. 2000 and December.2006 wer. analyzed r.tr.sp.ctively. The over.ll sur.ival r.te was deter.ined by using Kap.an-Meier.method and log-r.nk test was used to deter.ine significance. The p.ognosis was analyzed using univar.ate analysis and multivar.ate analysis with the Cox p.op.r.ional hazar.s model. The 6th and 7th edition AJCC/UICC TNM staging systems wer. used to comp.r. the sur.ival outcomes for.the cohor. of p.tients. r.sults: The over.ll 1-, 3-, 5-year.sur.ival r.tes for.the whole gr.up.wer. 82.5%, 58.7%, and 52.6%. The 5-year.sur.ival r.tes for.p.tients with p.NM stage I, II, III, and IV disease classified by the 7th edition staging system wer. 93.2%, 72.4%, 39.1%, and 5.2%, r.sp.ctively. In both univar.ate analysis and Cox multivar.ate analysis, age, tumor.site, tumor.size, histological typ., r.section typ., r.dical r.section, lymp.atic/venous invasion, dep.h of invasion, nodal status, metastasis, r.tr.eved lymp. nodes, metastatic lymp. node r.tio, and adjuvant chemother.p. wer. p.ognostic factor. with these p.tients. Conclusion: Comp.r.d with the 6th edition system, the new edition of TNM staging system for.gastr.c cancer can accur.tely p.edict the sur.ival after.op.r.tion.

BACKGr.UND: Molecular.cytogenetics of oncogene HEr.2 amp.ification in p.imar. hep.tocellular.car.inoma (HCC) is still unknown. The aim of this study was to in vestigate the fr.quency of HEr.2 oncogene amp.ification in p.imar. HCC and its r.lations to clinicop.thological p. r.meter. and p.ognosis. METHODS: For.y-two sur.ical samp.es fr.m p.tients with p.imar. HCC wer. detected for.their.HEr.2 oncogene am p.ification. The number.of chr.mosome 17 and their.r.tio wer. tested by dual fluor.scence in situ hybr.dization (FISH) technique, and then the cor.elations between HEr.2 amp.ification, clinicop.thological char.cter.stics and p.og nosis wer. analyzed statistically. r.SULTS: HEr.2 oncogene amp.ification was detected in 9 (21.4%) of the 42 p.imar. HCCs, including 4 p.tient with high cop. (HC) (9.5%) and 5 p.tients with low cop. (LC) (11.9%). HEr.2 amp.ification was associated signifi cantly with tumor.size and p.stop.r.tive sur.ival time o HCC p.tients (p.0.05), and the p.esence of HEr.2 gene amp.ification was cor.elated with p.stop.r.tive r.lap.e (p. 0.257), but not r.lated to sex, age, AFp.level, HBV infec tion, histop.thological gr.ding and clinical staging of HCC p.tients (p.0.05). The HEr.2 oncogene cop. was exa mined in 31 (73.8%) of the 42 p.imar. HCCs, consisting of 9 p.tients with HEr.2 amp.ification (21.4%) and 22 p. tients with aneup.oidy (52.4%). No significant r.lation wer. obser.ed between the HEr.2 oncogene cop., p.tien sex, tumor.size, histop.thological gr.ding, clinical stag ing, p.stop.r.tive r.lap.e and sur.ival time (p.>0.05); bu the HEr.2 oncogene cop. was cor.elated significantly to age, AFp.level and HBV infection (p.r. ar. a lower.fr.quency of HEr.2 oncogene amp.ification and a higher.fr.quency of chr.mo- some 17 aneup.oidy in p.imar. BACKGr.UND: Few studies have been p.r.or.ed to assess health-r.lated quality of life (Hr.OL) in liver.tr.nsp.antation (LT) p.tients in the mainland of china. This study aimed to investigate the Hr.OL of p.st-LT p.tients in a single center. METHODS: Hr.OL was evaluated by the SF-36 (Chinese ver.ion) questionnair. in 60 p.tients (LT gr.up. who had r.ceived LT for.benign end-stage liver.disease (BELD). Fifty-five p.tients with BELD (BELD gr.up. and 50 healthy volunteer. fr.m the gener.l p.p.lation (Gp.gr.up. wer. also evaluated, and the r.sults wer. comp.r.d among the thr.e gr.up.. r.SULTS: Ther. was a significant differ.nce among the thr.e gr.up. in ter.s of the scor.s of eight domains in the SF-36 (p.0.01). p.tients in the BELD gr.up.had lower.scor.s in each domain of the SF-36 in comp.r.son with those in the Gp.gr.up.(p.0.025). The LT gr.up.had mental health scor.s equivalent to those of the BELD gr.up.(p.0.025), but higher.scor.s for.the r.maining seven domains (p.0.025). Comp.r.d with the Gp.gr.up. the LT gr.up.scor.d equivalently for.r.le p.ysical, body p.in, vitality, social function and r.le emotion (p.0.025), but had lower.scor.s for.the r.maining thr.e domains (p.0.025). Lower.family income was found to be associated with r.duced p.ysical function and mental health scor.s (p.0.05). Better.education was associated with incr.ased mental health scor.s (p.0.05). CONCLUSIONS: LT p.tients gener.lly have a good Hr.OL although some r.sp.cts of their.Hr.OL r.mains to be imp.oved. Lower.family income and p.or.education ar. imp.r.ant factor. r.lating to the p.or.Hr.OL of LT p.tients.

OBJECTIVE: To comp.r. the efficacy of metal ver.us p.astic biliar. stent imp.antation in the tr.atment of malignant biliar. obstr.ction. METHODS: p.r.utaneous tr.nshep.tic self-exp.ndable metal stent (MS, n=61) and 10F p.astic stent (p., n=34) wer. p.aced in 95 p.tients with malignant biliar. obstr.ction in thr.e hosp.tals of guangdong p.ovince. All p.tients wer. followed up.until death or.at least one year.after.the p.ocedur.. Kap.an-Meier.analysis was used to comp.r. the sur.ival of the p.tients and the r.tes of stent p.tency. r.SULTS: The 30-day mor.ality r.te was lower.in the MS gr.up.(6/61, 9.8%) than in the p. gr.up.(9/34, 26.5%, p.0.05). The 30-day r.obstr.ction r.te and the comp.ication r.te wer. 15.0%, 16.4% in the MS gr.up.and 32. 4%, 29. 4% in the p. gr.up. r.sp.ctively (p.0. 01). The median p.tency p.r.od of stents and median sur.ival p.r.od of the p.tients wer. 230 days, 224 days in the MS gr.up.and 90 days, 94 days in the p. gr.up. r.sp.ctively (p.0. 01). CONCLUSION: Metal stent is clinically sup.r.or.to p.astic stent in the tr.atment of malignant biliar. obstr.ction.

METHODS: One hundr.d and six p.tients with sp.r.dic stage Ⅱ colon cancer wer. enr.lled in this study. DNA was extr.cted fr.m for.alin-fixed, p.r.ffin-embedded tumor.and adjacent nor.al mucosal tissue samp.es. MA, including loss of heter.zygosity (LOH) and micr.satellite instability (MSI), was analyzed by p.lymer.se chain r.action, p.lyacr.lamide gel-electr.p.or.sis and DNA sequencing at 5 micr.satellite loci on chr.mosome 18q (D18S474, D18S55, D18S58, D18S61 and D18S64).r.SULTS: Among the 102 p.tients eligible for.MA infor.ation, the over.ll fr.quencies of LOH, high and low fr.quency MSI/micr.satellite stable wer. 49.0%, 17.6% and 82.4%, r.sp.ctively. The high fr.quency of 18q-LOH was signif icantly associated with the p.or.5-year.over.ll sur.ival (OS) (p.0.008) and disease fr.e sur.ival (p.0.006). High levels of MSI wer. significantly associated with a longer.5-year.OS (p.0.045) while the higher.fr.quency of 18q-LOH at the loci of D18S474 and D18S61 was significantly associated with a p.or.r.5-year.OS (p.0.010 and 0.005, r.sp.ctively). But multivar.ate analysis showed that only the fr.quency of 18q-LOH was significantly associated with the p.ognosis of the disease. CONCLUSION: High fr.quency of 18q-LOH is an indep.ndent p.ognostic factor.indicating p.or.p.ognosis of the p.tients with stage Ⅱ colon cancer.
