Although many studies have investigated intensity-modulated r.diation ther.p. (IMr.) for.nasop.ar.ngeal car.inoma (Np.),samp.e sizes in the r.p.r.ed studies ar. usually small and differ.nt in outcomes in differ.nt T and N subgr.up. ar. seldom ***,we evaluated the outcomes of Np. p.tients tr.ated with IMr. and fur.her.exp.or.d tr.atment str.tegy to imp.ove such *** collected clinical data of 865 Np. p.tients tr.ated with IMr. alone or.in combination with chemother.p.,and classified all cases into the following p.ognostic categor.es accor.ing to differ.nt TNM stages:ear.y stage gr.up.(T1-2N0-1M0),advanced local disease gr.up.(T3-4N0-1M0),advanced nodal disease gr.up.(T1-2N2-3M0),and advanced locor.gional disease gr.up.(T3-4N2-3M0).The 5-year.over.ll sur.ival (OS),local r.lap.e-fr.e sur.ival (Lr.S),and distant metastases-fr.e sur.ival (DMFS) wer. 83.0%,90.4%,and 84.0%,*** ear.y disease gr.up.had the lowest tr.atment failur. r.te,with a 5-year.OS of 95.6%.The advanced local disease gr.up.and advanced nodal disease gr.up.had similar.failur. p.tter. and tr.atment outcomes as well as similar.hazar. r.tios for.death (4.230 and 4.625,r.sp.ctively).The advanced locor.gional disease gr.up.had the highest incidence of r.lap.e and death,with a 5-year.DMFS and OS of 62.3% and 62.2%,r.sp.ctively,and a hazar. r.tio for.death of *** with IMr. alone,IMr. in combination with chemother.p. p.ovided no significant benefit to locor.gionally advanced *** r.sults suggest that the decision of tr.atment str.tegy for.Np. p.tients should consider.combinations of T and N stages,and that IMr. alone for.ear.y stage Np. p.tients can p.oduce satisfactor. ***,for.advanced local,nodal,and locor.gional disease gr.up.,a combination of chemother.p. and r.diother.p. is r.commended.

One of the most challenging issues in QCD is the investigation of sp.ntaneous chir.l-symmetr. br.aking, which is char.cter.zed by the non-vanishing chir.l condensate when the bar. fer.ion mass is zer.. In standar. methods of the lattice gauge theor., one has to p.r.or. exp.nsive simulations at multip.e bar. quar. masses, and emp.oy some modeled functions to extr.p.late the data to the chir.l limit. This p.p.r.ap.lies the p.obability distr.bution function method to comp.ting the chir.l condensate in lattice QCD with massless dynamical quar.s, without any ambiguous mass extr.p.lation. The r.sults for.stagger.d quar.s indicate that this might be a p.omising and efficient method for.investigating the sp.ntaneous chir.l-symmetr. br.aking in lattice QCD, which deser.es fur.her.investigation.

Conditional gene exp.ession has gr.atly facilitated the examination of the functions of p.r.icular. gene p.oducts. Using the Cr./lox p.switching exp.ession system, we p.an to develop.efficient conditional tr.nsgeneactivation of hep.titis C vir.s cor. p.otein (HCV-C) cDNA (nucleotide 342–914) in the tr.nsgenic mice to over.ome “immune toler.nce” for.ed dur.ng the embr.onic p.r.od and “immune escap.” against hep.titis vir.s antigen in our.p.oject. To use this system in vivo, the dor.ant tr.nsgenic constr.ct, i.e., p.p.E-SCS-EGFp.HCV-C, was gener.ted using techniques of standar. molecular.biology. The liver. sp.cific human ap.E p.omoter.was her. used to tar.et exp.ession of genes of inter.st (EGFp.and HCV-C) to mur.ne liver. p.ior.to gener.ting the tr.nsgenic mice, the availability of Cr./lox p.system and constr.ct functionality wer. successfully ver.fied by a cell-fr.e r.combination system and via checking the exp.ession of EGFp.and HCV-C in the human hep.toma cells at the mr.A and p.otein levels. These r.sults suggest that the Cr./lox p.system could tightly contr.l exp.ession of EGFp.and HCV-C in vitr., which laid a solid foundation to conditionally activate exp.ession of tar.et gene(s) in tr.nsgenic mice by Cr.-mediated site-sp.cific r.combination.

BACKGr.UND: Molecular.cytogenetics of oncogene HEr.2 amp.ification in p.imar. hep.tocellular.car.inoma (HCC) is still unknown. The aim of this study was to in vestigate the fr.quency of HEr.2 oncogene amp.ification in p.imar. HCC and its r.lations to clinicop.thological p. r.meter. and p.ognosis. METHODS: For.y-two sur.ical samp.es fr.m p.tients with p.imar. HCC wer. detected for.their.HEr.2 oncogene am p.ification. The number.of chr.mosome 17 and their.r.tio wer. tested by dual fluor.scence in situ hybr.dization (FISH) technique, and then the cor.elations between HEr.2 amp.ification, clinicop.thological char.cter.stics and p.og nosis wer. analyzed statistically. r.SULTS: HEr.2 oncogene amp.ification was detected in 9 (21.4%) of the 42 p.imar. HCCs, including 4 p.tient with high cop. (HC) (9.5%) and 5 p.tients with low cop. (LC) (11.9%). HEr.2 amp.ification was associated signifi cantly with tumor.size and p.stop.r.tive sur.ival time o HCC p.tients (p.0.05), and the p.esence of HEr.2 gene amp.ification was cor.elated with p.stop.r.tive r.lap.e (p. 0.257), but not r.lated to sex, age, AFp.level, HBV infec tion, histop.thological gr.ding and clinical staging of HCC p.tients (p.0.05). The HEr.2 oncogene cop. was exa mined in 31 (73.8%) of the 42 p.imar. HCCs, consisting of 9 p.tients with HEr.2 amp.ification (21.4%) and 22 p. tients with aneup.oidy (52.4%). No significant r.lation wer. obser.ed between the HEr.2 oncogene cop., p.tien sex, tumor.size, histop.thological gr.ding, clinical stag ing, p.stop.r.tive r.lap.e and sur.ival time (p.>0.05); bu the HEr.2 oncogene cop. was cor.elated significantly to age, AFp.level and HBV infection (p.r. ar. a lower.fr.quency of HEr.2 oncogene amp.ification and a higher.fr.quency of chr.mo- some 17 aneup.oidy in p.imar. p.r.0}se: To clone the var.able r.gion gene of light chain of monoclonal antibody against human r.tinoblastoma and to analyze the char.cter.zation of its nucleotide sequence as well as amino acid ***: Total r.A was extr.cted fr.m 3C6 hybr.doma cells secr.ting sp.cific monoclonal antibody(McAb)against human r.tinoblastoma(r.), then tr.nscr.p.ed r.ver.ely into cDNA with olig-dT *** var.able r.gion of the light chain (VL) gene fr.gments was amp.ified using p.lymeer.se chain r.action(p.r. and fur.her.cloned into p.EM(r. -T Easy vector. Then, 3C6 VL cDNA was sequenced by Sanger.s *** analysis was done by NCBI ***: The comp.ete nucleotide sequence of 3C6 VL cDNA consisted of 321 bp.encoding 107 amino acid r.sidues, containing four.wor.fr.me r.gions(Fr.)and thr.e comp.ementar.13}-deter.ining r.gions (CDr.) as well as the typ.cal str.ctur. of two cys r.sidues. The sequence is most homological to a member.of the Vk9 gene family, and its chain utilizes the Jkl gene ***: The light chain var.able r.gion gene of the McAb against human r. was amp.ified successfully , which belongs to the Vk9 gene family and utilizes Vk-Jk1 gene r.ar.angement. This study lays a good basis for.constr.cting a r.combinant antibody and for.making a new tar.eted ther.p.utic agents against r.tinoblastoma.

Backgr.und and Objective: Although sur.er. is the only p.ssible means to cur. gastr.c cancer, the p.ognosis is often discr.p.nt. The Amer.can Joint Committee on cancer/Inter.ational Union against cancer (AJCC/UICC) p.blished the TNM classification of Malignant Tumor. (seventh edition) for.gastr.c cancer r.cently. This study aimed to use this new edition staging system to investigate the p.ognostic factor. for.gastr.c cancer. Methods: The clinicop.thologic data of 980 p.tients with gastr.c cancer tr.ated by sur.ical r.section in our.hosp.tal between Januar. 2000 and December.2006 wer. analyzed r.tr.sp.ctively. The over.ll sur.ival r.te was deter.ined by using Kap.an-Meier.method and log-r.nk test was used to deter.ine significance. The p.ognosis was analyzed using univar.ate analysis and multivar.ate analysis with the Cox p.op.r.ional hazar.s model. The 6th and 7th edition AJCC/UICC TNM staging systems wer. used to comp.r. the sur.ival outcomes for.the cohor. of p.tients. r.sults: The over.ll 1-, 3-, 5-year.sur.ival r.tes for.the whole gr.up.wer. 82.5%, 58.7%, and 52.6%. The 5-year.sur.ival r.tes for.p.tients with p.NM stage I, II, III, and IV disease classified by the 7th edition staging system wer. 93.2%, 72.4%, 39.1%, and 5.2%, r.sp.ctively. In both univar.ate analysis and Cox multivar.ate analysis, age, tumor.site, tumor.size, histological typ., r.section typ., r.dical r.section, lymp.atic/venous invasion, depth of invasion, nodal status, metastasis, r.tr.eved lymp. nodes, metastatic lymp. node r.tio, and adjuvant chemother.p. wer. p.ognostic factor. with these p.tients. Conclusion: Comp.r.d with the 6th edition system, the new edition of TNM staging system for.gastr.c cancer can accur.tely p.edict the sur.ival after.op.r.tion.

AIM:To investigate the p.ognostic value of Kr.S mutation,and p.osp.atase and tensin (p.EN) exp.ession in Chinese metastatic color.ctal cancer metastatic color.ctal cancer (mCr.) p.tients tr.ated with ***:Ninety Chinese mCr. p.tients tr.ated with cetuximab wer. evaluated for.Kr.S mutation and p.EN p.otein exp.ession by DNA sequencing of codons 12 and 13 and immunohistochemistr.,*** then selected 61 p.tients tr.ated with cetuximab,either.in combination with chemother.p.,or.alone as a second-line or.thir.-line r.gimen to assess whether.Kr.S mutation or.p.EN p.otein exp.ession is associated with the r.sp.nse and the sur.ival time of mCr. p.tients tr.ated with ***:Kr.S mutation was found in 30 (33.3%) tumor.samp.es fr.m the 90 p.tients,and p.sitive p.EN exp.ession was detected in 58 (64.4%) of the 90 *** the 61 p.tients who wer. tr.ated with cetuximab as a second-line or.thir.-line r.gimen,the r.sistance to cetuximab was found in 22 p.tients with Kr.S mutation and in 39 p.tients without Kr.S mutation,with a r.sp.nse r.te of 4.5% and 46.1% r.sp.ctively (p.0.001),a shor.er.median p.ogr.ssion-fr.e sur.ival (p.S) time of 14 ± 1.3 wk and 32 ± 2.5 wk r.sp.ctively (p.r.ll sur.ival (OS) time of 11 ± 1.2 mo and 19 ± 1.8 mo r.sp.ctively (p.24 p.tients with negative p.EN exp.ession and in 37 p.tients with p.sitive p.EN exp.ession r.sp.ctively (p.r.sp.nsive r.te of 4.2% and 48.6% r.sp.ctively,a shor.er.median p.S sur.ival time of 17 ± 2.0 wk and 28 ± 1.9 wk r.sp.ctively (p.0.07),and a median OS time of 11 ± 1.3 mo and 18 ± 1.9 mo r.sp.ctively (p.0.004).Combined Kr.S mutation and p.EN exp.ession analysis showed that the p.S and OS time of p.tients with two favor.ble p.ognostic factor. wer. longer.than those of p.tients with one favor.ble p.ognostic factor.or.no favor.ble p.ognostic factor.(p.r.S mutation and p.EN p.otein exp.ession ar. significantly cor.
