While it has been well demonstrated that quantum dots (QDs) play an important role inbiological labeling both in vitro and in vivo,there is no report describing the cellular nanostructure basis ofreceptor-mediated ***,nanostructure evolution responses to the endocytosis of transferrin(Tf)-conjugated QDs were characterized by atomic force microscopy (AFM).AFM-based nanostructureanalysis demonstrated that the Tf-conjugated QDs were specifically and tightly bound to the cell receptorsand the nanostructure evolution is highly correlated with the cell membrane receptor-mediated ***,confocal microscopic and flow cytometry results have demonstrated the specificity anddynamic property of Tf-QD binding and *** found that the internalization of Tf-QD is linearlyrelated to ***,while the nanoparticles on the cell membrane increased,the endocytosis was stillvery active,suggesting that QD nanoparticles did not interfere sterically with the binding and function ***,ligand-conjugated QDs are potentially useful in biological labeling of cells at a nanometerscale.

Mouse Pem,a homeobox gene,encodes a protein consisting of 210 amino acid *** the function of mouse Pem protein,we used the yeast two-hybrid system to screen the library of 7-daymouse embryo with full-length mouse Peru ***-two colonies were obtained after 1.57×10~8 colonieswere screened by nutrition limitation and β-galactosidase *** individual insert fragments wereobtained from the library,and three of them were identified,one of which was confirmed to be the celldivision cycle 37(Cdc37)homolog gene by *** interaction between mouse Pem and Cdc37homolog was then confirmed by glutathione S-transferase pull-down assay,and the possible interaction modelwas suggested.

Diversity, distribution, and new phylogenetic information of calcareous dinoflagellates from the china Sea

Calcareous dinoflagellates have been recorded from subarctic to tropical areas, inhabiting both neritic and oceanic regions. In the present study we analyse 12 sediment samples from the china Sea to establish the existence of calcareous dinoflagellates in this area. A total of 11 calcareous dinoflagellate species were recorded, among them, three genera (Calciodinellum, Leonella, Posoniella) and seven species (Calciodinellum albatrosianum, C. levantinum, C. operosum, Leonella granifera, Posoniella tricarinelloides, Scrippsiella irregularis, and S. regalis) were reported from the china Sea for the first time. Cysts of S. irregularis and S. precaria are identical, with a mesoepicystal archeopyle, representing the loss of 2–4 and 1–3a paraplates; however, their thecal plates vary slightly and differ from each other in 78 positions of the internal transcribed spacer sequence. Scrippsiella trochoidea is dominant in coastal areas, whereas Calciodinellum albatrosianum is the most abundant offshore. Scrippsiella irregularis is distributed widely along the coast of china, whereas S. precaria is recorded only in northern china. Leonella granifera is the second most abundant species offshore, comprising 15–40% of the cyst assemblage.

In Vitro Construction of Effective M1GS Ribozymes Targeting HCMV UL54 RNA Segments

Seven sequence-specific ribozymes(M1GS RNAs)derived in vitro from the catalytic RNAsubunit of Escherichia coli RNase P and targeting the mRNAs transcribed by the UL54 gene encoding theDNA polymerase of human cytomegalovirus were screened from 11 ribozymes that were designed based onfour rules:(1)the NCCA-3′ terminal must be unpaired with the substrate;(2)the guide sequence(GS)mustbe at least 12 nt in length;(3)the eighth nucleotide must be U,counting from the site-1;and(4)around thecleavage site,the sites-1/+1/+2 must be U/G/C or C/G/*** investigation of the factors affecting thecleavage effect and the optimal ratio for M1GS/substrate was carried *** was determined that the optimalratio for M1GS/substrate was 2:1 and too much M1GS 1ed to substrate *** indicated *** M1GS that cleaved HCMV UL54 RNA segments in vitro were successfully designed and *** studies support the use of ribozyme M1GS as antisense molecules to silence HCMV mRNA in vitro,andusing the selection procedure as a general approach for the engineering of RNase P ribozymes.

Microwave peak absorption frequency of liquid

Microwave-assisted extraction is a new effective method which has practical ap-plications in many fields. Microwave heating is one of its physical mechanisms,and it also has the characteristic of selectivity. When the applied microwave fre-quency equals a certain absorption frequency of the material (or specific compo-nent),the material will intensively absorb microwave energy. This is also known as resonant absorption,and the frequency is called the peak absorption frequency which depends on the physical structure of the material. In this work,dynamic hy-drogen bond energy was included in molecular activation energy; with the liquid cell model,the expression of interaction energy between dipolar molecules was derived. The rotational relaxation time was gotten from the Eyring viscosity formula. Then based on the relationship between dielectric dissipation coefficient and re-laxation time,the expression of microwave peak absorption frequency as a func-tion of the material physical structure,rotational inertia and electrical dipole mo-ment of molecules was established. These theoretical formulas were applied to water and benzene,and the calculated results agree fairly well with the experi-mental data. This work can not only deepen the study of the interaction between microwave and material,but also provide a possible guide for the experiment of microwave-assisted extraction.

Investigation on the Surface Coating of Grinding Balls

The surface coating of grinding balls was investigated experimentally. The results show that a coating may form on the surface of grinding balls when Cr or Al powders are subjected to ball grinding. The plastic deformation of the ball surface plays an important role during the coating formation, and the strong binding force between the powders and the balls is a necessary pre-condition. The thickness of coating increases with the plasticity of the powders and the balls. Annealing the balls with coating will result in an obvious diffusion of the elements in the bonding zone of interface.

Phylogeny of Ptychostomum (Bryaceae,Musci) inferred from sequences of nuclear ribosomal DNA internal transcribed spacer (ITS) and chloroplast rps4

The phylogeny of Ptychostomum was first undertaken based on analysis of the internal transcribed spacer(ITS)region of the nuclear ribosomal(nr)DNA and by combining data from nrDNA ITS and chloroplast DNA rps4 *** parsimony,maximum likelihood,and Bayesian analyses all support the conclusion that the reinstated genus Ptychostomum is not *** funkii(Schw(a|¨)gr.)*** (≡Bryum funkii Schw(a|¨)gr.)is placed within a clade containing the type species of Bryum,*** Hedw. The remaining members of Ptychostomum investigated in the present study constitute another well-supported clade. The results are congruent with previous molecular *** the basis of phylogenetic evidence,we agree with transferring *** Mü***.(≡***(Brid.)Turton≡Bryum archangelicum Bruch & Schimp.), Bryum lonchocaulon Mü***.,Bryum pallescens *** Schw(a|¨)gr.,and Bryum pallens *** Ptychostomum.

Effects of temperature and organic and inorganic nutrients on the growth of Chattonella marina (Raphidophyceae)from the Daya Bay,South china Sea

The effects of temperature and different forms of nutrients on Chattonella marina growth have been investigated in strains isolated from the Daya Bay, the South china Sea. The strain of C. marina preferred high temperatures, with an optimal temperature of 25℃, and 18℃ was the minimum for its survival. Higher cell number and growth rate were obtained in high nitrogen and phosphorus concentrations （500 μg/L, 74μg/L） than under nutrient limitation. Nitrogen influenced the growth most, as the specific growth rate and maximum cell density were lower in nitrogen- limited cultures than noted under phosphorus limitation or under limitation from both. C. marina was capable of using many kinds of organic nitrogen sources including L-serine （L-Ser）, glycine （Gly）, alanine （Ala）, L-threonine （L-Thr）, glutamic acid （Glu） and urea, but could not utilize uric acid. Various forms of organic phosphorus compound such as glucose-6-phosphate （G6P）, sodium glycerophosphate （GYP）, adenosine triphosphate （ATP）, adenosine monophosphate （AMP）, cyti- dine monophosphate （CMP）, guanosine monophosphate （GMP）, uridine monophosphate （UMP）, 4-nitrophenylphosphate （NPP） and triethyl phosphate （TEP） supported the growth as well. Algal cells had the ability to sustain growth under nitrogenand/or phosphorus-free conditions particularly under phosphorus depleted condition. These results led to the hypothesis that high loading of nitrogen has played an important role in frequent C. marina blooms in the past decade, and its capability for utilization of diverse forms of organic nutrients and growth in low nutrient conditions make this species a likely recurrent dominant in the Daya Bay phytoplankton assemblages, visible as more frequent blooms.

Protokeelia is described as a new and rare pennate diatom genus in *** valve includes two parts:ventral valve and dorsal *** ventral valve is more or less convex or *** raphe runs in a distinct elevated and thickened siliceous *** the internal valve,beneath the raphe are fibulae or fibulae clusters which connect the ventral and dorsal *** species,*** Round & Basson,is observed in the water samples from a mangrove forest in Sai Kung,Hong Kong,*** main morphological features are described in this paper along with electron microscope(EM) *** comparisons are also made among several allied *** addition,a newly recorded species for china,Rhopalodia iriomotensis Kobayasi,Nagumo & Tanaka,is discussed *** several key features observed in this species resemble those of some Pro-tokeelia species,we suggest that it may actually represent a Protokeelia species.

pH-Dependent Stability of EGX, A Multi-functional Cellulase from Mollusca, Ampullaria crossean

The cellulase activity and stability of EGX, a multi-functional cellulase previously purified from the mollusca Ampullaria crossean, was systematically studied under different pH. The pH induced con- formation and stability change of EGX have been investigated by using the intrinsic fluorescence, ANS fluorescence and CD spectrum. It has been found that the conformation and activity of this cellulase were strongly dependent on the pH. EGX was stable for both the enzyme activity and the conformation from pH 5.6 to pH 7.4. As shown by intrinsic and ANS fluorescence, no red shift of emission maximum occurred and a negligible intensity change was observed at pH 5.6–7.4. The activity of EGX remained about 80% in pH 5.6–7.4 and obviously decreased out of side the pH range. Urea-induced changes in EGX at pH 5.4 and pH 8.0 were measured by intrinsic fluorescence and CD spectrum. At pH 5.4, a significantly red shift of emission maximum occurred when the concentration of urea was 5 M compared to the concentration was 3 M at pH 8.0. The α-helix at pH 5.4 was 40.51% in the absence of urea and 31.04% in the presence of 4 M urea. At pH 8.0 the α-helix was 7.23% in the presence of 4 M urea. The data indicated that EGX was much susceptible to urea-induced unfolding at pH 8.0 and much stable at pH 5.4. The greater pH dependent stability of EGX may allow the enzyme to adequately catalyze the hydrolysis of cellulosic materials under natural or industrial extreme conditions.
