Association of apolipoprotein E 4 polymorphism with cerebral infarction in Chinese Han population

AIM: To study the association between ApOE polymorphisms and cerebral infarction through a case-control study among the Chinese Han population. METHODS: First-ever cerebral infarction patients (n=226) whose ages ranged from 40 to 60 years old were recruited from department of Neurology, Zhongshan hospital, Shanghai, and zhejiang Chinese Traditional Medicine hospital, zhejiang, china. Unrelated healthy controls (n=201) were selected from the general population in the same area with similar age and sex distribution. ApOE was amplified by one-stage pCr using the forward primer: 5'-GGC ACG GCT GTC CAA GGA GCT-3' and reverse primer: 5'-GAT GGC GCTGAG GCC GCG CT-3'. The pCr product was digested directly with 5 U of CfoI and separated by a 20 % polyacryla- mide (acrylamide: bis-acrylamide=29:1) nondenaturing gel. rESULTS: Both cerebral infarction patient and control groups were in Hardy-Weinberg equilibrium. The allele frequency of ApOE*2, ApOE*3, and ApOE*4 was 4.6 %, 81.9 %, and 13.5 % respectively in the patients with cerebral infarction; 5.7 %, 87.3 %, and 7.0 % respectively in the healthy control group. Compared with ApOE3/3 subjects, ApOE4/4 carriers had a 2.1-fold risk of cerebral infarction (odds ratio 2.1, 95 % confidence limits 1.3 to 3.4). The allele frequency of ApOE*4 in the cerebral infarction patient group was significantly higher than that in the control group (13.5 % vs 7.0 %; p=0.002). CONCLUSION: ApOE 4 is a risk factor for cerebral infarction among the Chinese Han population.

BACKGrOUnd:Hepatocellular carcinoma (HCC) is characterized by a multi-cause,multi-stage and multi-focus process of tumor *** prognosis is poor and early diagnosis is of utmost *** study was undertaken to investigate the dynamic expression of oncofetal antigen glypican-3 (GpC-3) and GpC-3 mrNA in hepatocarcinogenesis and to explore their early diagnostic value for ***:A hepatoma model was induced in male Sprague-Dawley rats with 0.05% 2-fluorenylacetamide and confirmed by hematoxylin and eosin staining and gamma-glutamyltransferase (GGT) *** rNA was purified and transcribed into cDNA by reverse *** of the GpC-3 gene were amplified by nested rT-pCr,and confirmed by ***-3 was analyzed by immunohistochemistry,Western blotting or ***:positive GpC-3 expression showed as brown granule-like staining localized in the *** examination of hepatocytes revealed three morphological stages of granule-like degeneration,atypical hyperplasia (precancerous),and cancer formation,with a progressive increase of liver total rNA and GGT *** incidence of liver GpC-3 mrNA and GpC-3,and serum GpC-3 was 100%,100% and 77.8% in the HCC group,100%,100%,and 66.7% in the precancerous group,83.3%,83.3%,and 38.9% in the degeneration group,and no expression in the liver or blood of the control group,*** was a positive correlation between liver GpC-3 mrNA and total rNA level (r=0.475,pr liver GpC-3 (r=1.0,p1) or serum GpC-3 (r= 0.994,p1).CONCLUSION:Abnormal oncofetal antigen GpC-3 and GpC-3 mrNA expression in hepatocarcinogenesis may be promising molecular markers for early diagnosis of HCC.

The visual spatial attention mechanism in the brain was studied in 16 young subjects through the visual search paradigm of precue-target by the event-related potential (Erp) technique, with the attentive ranges cued by different scales of Chinese character and region cues. The results showed that the response time for Chinese character cues was much longer than that for region cues especially for small region cues. With the exterior interferences, the target stimuli recognition under region cues was much quicker than that under Chinese character cues. Compared with that under region cues, targets under Chinese character cues could lead to increase of the posterior p1, decrease of the N1 and increase of the p2. It should also be noted that the differences between region cues and Chinese character cues were affected by the interference types. Under exterior interferences, no significant difference was found between region cues and Chinese character cues; however, it was not the case under the interior interferences. Considering the difference between the exterior interferences and the interior interferences, we could conclude that with the increase of difficulty in target recognition there was obvious difference in the consumption of anterior frontal resources by target stimuli under the two kinds of cues.

AIM: To investigate the inhibitory effect of tea polyphenols (Tp)on the transforming growth factor-pi (TGF-β1) expression in rat model of cyclosporine A (CsA)-induced chronic nephrotoxicity. METHODS: The rat model of CsA-induced chronic nephrotoxicity was used, 4 groups of rats were respectively treated with vehicle (0.1 mL·lg-1-d-1 sc), Tp (80 mg·kg-1·d-1 ig), CsA (15 mg·2kg-1·d-1sc) and Tp plus CsA (CsA 15 mg·kg-1·d-1 sc+Tp 80 mg·kg-1·d-1 ig). At the end of day 28 of treatment, serum and urine are analyzed for creatinine clearance, kidney tissue for pathologic analysis. Comparison of Clinical Efficacy of Three Methods for Cervical ripening followed by Surgical Evacuation in Early Missed Abortion

Objective To compare clinical efficacy of three cervical preparations for surgical evacuation in first trimester missed abortion. Methods Two hundred and ten women with early missed abortion were equally randomized into three groups. Three cervical preparations, Dilapan-dilator method （group A, n= 70）, mifepristone method （group B, n= 70）, and misoprostol method （group C, n= 70）, were used 24 h before curettage for terminating missed abortion, respectively. Clinical outcomes and complications for these three methods were compared. results No differences were found in rates of successful treatment among the three groups （p〉O.05）. The incidence of fitting number 8 Hegar dilator was slightly high in group A, but it was not different compared with that in group B or group C （p〉0.05）. Bleeding in previous 24 h in group C was significantly greater than that in group A or group B （p〈0.01）. The cases of blood loss （〉50 ml） during evacuation in group B were more than those in group A （p〉0.05）. Women in group B had less abdominal pain than those in group A or group C （p〈0.01）. Side effects in group C were more than those in group A on nausea （p〈0.01）, vomiting （p〈0.05）, and diarrhea （p〈0.01）. Group B had the highest acceptability among the three groups （p〈0.05）. Conclusion Three methods have the similar clinical efficacies in cervical ripening for surgical treatment of early missed abortion. Based on individual characteristics, different approach would be chosen to avoid some adverse events and improve the clinical application of Dilapan-dilator.

Aim:To determine whether glycine could downregulate interleukin 1 receptorassociated kinase-4(IrAK-4)expression to interfere with lipopolysaccharides(LpS)signal transduction and blunt transplantative liver ischemia-reperfusioninjury(I/rI).Methods:SD rats were randomly divided into two groups:donoranimals of the glycine group(n=40)were given glycine(1.5 mL;300 mmol/L,iv)1h before harvest,and the control group were treated with 1.5 mL physiologicalsaline(n=40).Orthotropic liver transplantation was then performed according tothe Kamada *** animals in each group were followed up for 7 d aftersurgery to assess *** remaining animals in each group were dividedinto 3 subgroups(n=10)at 1h,2h and 6 h after portal vein *** ofLpS,serum aspartate aminotransferase(AST),alanine aminotransferase(ALT)and total bilirubin in portal circulation,as well as IrAK-4 and TNF-α expression,NF-κB transcriptional activity and morphological study of liver tissues ***:reperfusion resulted in a significant elevation of LpS concen-trations in each group persisting to the end of our ***,glycine,whichled to improved survival rate and liver function,significantly alleviated liver pa-renchyma cell damage by downregulating 1rAK-4,TNF-α expression and NF-κBtranscriptional activity compared with the control ***:Glycinecan attenuate hepatic I/rI by downregulating IrAK-4 to interfere with LpS signaltransduction.

This work characterizes the mitochondrial proteomic profile in the failing heart and elucidates the molecular basis of mitochondria in heart failure. Heart failure was induced in rats by myocardial infarction, and mitochondria were isolated from hearts by differential centrifugation. Using two-dimensional gel electrophoresis and matrix-assisted laser desorption/ionization-time of flight mass spectrometry, a system biology approach was employed to investigate differences in mitochondrial proteins between normal and failing hearts. Mass spectrometry identified 27 proteins differentially expressed that involved in energy metabolism. Among those, the up-regulated proteins included tricarboxylic acid cycle enzymes and pyruvate dehydrogenase complex subunits while the down-regulated proteins were involved in fatty acid oxidation and the OXpHOS complex. These results suggest a substantial metabolic switch from free fatty acid oxidation to glycolysis in heart failure and provide molecular evidence for alterations in the structural and functional parameters of mitochondria that may contribute to cardiac dysfunction during ischemic injury.
