Aim:To investigate the effects of tanshinone ⅡA（Tan ⅡA）on the regulation ofthe production of endothelin（ET）-1（including large ET-1）,mrNA levels of ET-1,endothelin-converting enzyme-1（ECE-1）,endothelin-A receptor（ETA）andendothelin-B receptor（ETB）induced by TNF-α in rat brain microvascular endo-thelial cells（BMVEC）.Methods:The ET-1 release（including large ET-1）into theculture medium was determined by enzyme *** levels of ET-1,ECE- 1,ETA,and ETBmrNA were measured by *** receptor bind-ing was also ***:The induction of ET-1 release by TNF-α from cul-tured BMVEC was dose-dependently reduced by Tan IIA,but large ET-1 levelsprogressively increased in response to Tan ⅡA;the mrNA expression of ET-1 *** ⅡA also caused a decrease in ETAreceptor mrNA and ECE-1expression in a dose-dependent *** receptor binding was unal-tered in BMVEC stimulated with TNF-α alone or a combination of TNF-α and TanⅡ***:These findings suggest that Tan ⅡA may inhibit ET-1 produc-tion in TNF-α-induced BMVEC through the suppression of ECE-1 synthesis.

This study aimed to develop a cell culture model of Huntington disease and observe the effect of sodium butyrate on this cell culture *** 1 of both a wild type and a mutant IT15 gene from the genomic DNA of a healthy adult and a patient with Hunt- ington disease was amplified and cloned into the eukaryotic expression vector *** neuroblastoma SH-SY5Y cells were tran- siently transfected with these recombinant plasmids in the absence and presence of sodium butyrate(0.1,0.2,0.5,1.0 mmol/L).The MTT assay was used to measure cell *** results indicated that the N-terminal fragment of mutant huntingtin formed perinuclear and intranuclear aggregates and caused a decrease of SH-SY5Y cell *** butyrate inhibited the decrease of SH-SY5Y cell via- bility caused by the N-terminal fragment of mutant *** suggests that sodium butyrate has a protective effect on this cell cul- ture model of Huntington disease.

AIM: To investigate the influence of L-methionine-deprived total parenteral nutrition with 5-FU on gastric cancer and host metabolism. METHODS: N-methyl-N'-nitro-nitrosoguanidine (MNNG) induced gastric cancer rats were randomly divided into four groups: Met-containing TpN group (n=11), Met-deprived TpN group (n =12), Met-containing TpN+5-FU group (n=11) and Met-deprived TpN+5-FU group (n=12). Five rats in each group were sacrificed after 7 days of treatment and the samples were taken for examination. The remaining rats in each group were then fed separately with normal diet after the treatment until death, the life span was noted. rESULTS: The tumors were enlarged in Met-containing group and shrank in Met-deprived group markedly after the treatment. The DNA index (DI) of tumor cells and the body weight (BW) of rats had no significant change in the two groups, however, the ratio of tumor cells'S phase was increased. The ratio of G2M phase went up in Met-containing group, but down in Met-deprived group. In the other two groups that 5-FU was added, the BW of rats, and the diameter of tumors, the DI of tumor cells, the S and G2M phase ratio of tumor cells were all decreased, particularly in Met-deprived plus 5-FU group. pathological examination revealed that the necrotic foci of the tumor tissue increased after Met-deprived TpN treatment, and the nucleoli of tumor cells enlarged. In MetTpN+5-FU group, severe nuclear damage was also found by karyopyknosis and karyorrhexis, meanwhile there was slight degeneration in some liver and kidney cells. The serum free Met and Cysteine decreased markedly (p1), while other amino acids, such as serum free serine and glutamine increased significantly (prats died of multiple organ failure caused by cancer metastasis. The average survival time was 18.6 days in Met-containing TpN group, 31 days in Met-deprived TpN group, 27.5 days in Met-containing TpN+5-FU group, and 43 days in Met-deprived TpN+5-FU group (p&

BACKGrOUnd: Calcitonin gene-related peptide (CGrp) and nerve growth actor (NGF) cam improve spatial learning and memory abilities of rats with cerebral ischemia/reperfusion; however, the effect of combination of them on relieving learning and memory injury following cerebral ischemia/reperfusion should be further studied. OBJECTIVE: To study the effects of exogenous CGrp and NGF on learning and memory abilities of rats with focal cerebral ischemia/reperfusion. DESIGN: randomized controlled animal study. SETTING: department of neurosurgery, the Second hospital of Xiamen; department of neurosurgery, the Second affiliated hospital of china Medical university; department of Neurobiology, Basic Medical College of china Medical university. MATErIALS: A total of 30 healthy male SD rats, aged 8 weeks, of clean grade, weighing 250-300 g, were provided by Experimental Animal department of china Medical university. All rats were randomly divided into sham-operation group, ischemia/reperfusion group and treatment group with 10 in each group. The main reagents were detailed as the follows: 100 g/L chloral hydrate, 0.5 mL CGrp (2 mg/L, Sigma Company, usa), and NGF (1× 106 U/L, 0.5 mL, Siweite Company, Dalian). METHODS: The experiment was carried out in the department of Neurobiology, Basic Medical College of china Medical university from February to July 2005. rat models of middle cerebral artery occlusion were established by method of occlusion, 2 hours after that rats were anesthetized and the thread was slightly drawn out for 10 mm under direct staring to perform reperfusion. rats in the ischemia/reperfusion group received intraperitoneal injection of 1 mL saline via the abdomen at two hours later, while rats in the treatment group at 2 hours later received intraperitoneal injection of 2 mg/L CGrp (0.5 mL) and 1×106 U/L NGF (0.5 mL) once a day for 10 successive days. In this paper,a novel method to automatically detect protein spots on a two-dimensional (2-D) electrophoresis gel image is proposed to implement proteomics analysis of complex *** the basis of the identifying spots results based on color variation and spot size features,morphological feature is introduced as a new criterion to distinguish protein spots from non-protein ***-sharpening,edge-detecting and morphological feature extraction methods were consequently combined to detect protein spots on a 2-D electrophoresis gel image subject to strong *** proposed method was applied to detect the protein spots of proteomic gel images from *** cell,human kidney tissue and human *** results demonstrated that this method is more accurate and reliable than previous methods such as pDQuest 7.2 and ImageMaster 5.0 software for detecting protein spots on gel images with strong interferences.

AIM: The most recent and innovative AquaLase liquefaction technology has offered an alternative to lens extraction. Many studies have investigated its functions and advantages. This article focuses on evaluating the vivo microscopic cornea changes after AquaLase liquefaction by using a laser confocal microscope. METHODS: In this perspective, randomized case study, 37 eyes of 35 patients submitted to cataract surgery were chosen to undergo AquaLase liquefaction cataract extraction. Each patient was assessed before the operation, on the 1(st), 7(th), and 30(th) postoperative days, and 6 months after the cataract extraction. The morphologies and quantitative comparisons of corneal cells and corneal nerves layer by layer were evaluated in vivo with the Heidelberg retina Tomograph III-rostock Cornea Module (HrT-III rCM) confocal microscope. ANOVA and post-Hoc Bonferroni test were carried out to compare the results pre- and post-operation. rESULTS: ANOVA results indicated no post-operation changes for epithelium and anterior stroma cells. Irregular segments of sub-basal nerve fiber were most pronounced seven days post-operation. In the mid and posterior stroma, keratocytes were obvious compared with the preoperative condition. Corneal endothelium cells became obviously swollen in cytoplasm and nucleus. The mid and posterior stroma cell density decreased from the 1(st) to 7(th) postoperative days (p rneal endothelium cell density decreased (p nd did not revert to the preoperative level after six months (p rostructural abnormalities were identified in the corneal recovery process after AquaLase liquefaction. AquaLase liquefaction cataract extraction is safe for cornea.

Transcriptional regulation of the neuronal nitric oxide synthase gene(nNOS)is particularlycomplex as 12 distinct transcripts derived from different first exons are expressed in a tissue- and cell-specific *** exon 1f mrNA is relatively highly expressed in nervous system and relies upon exon1f-specific promoter *** conventional and real-time reverse transcription-polymerase chain reaction,we found exon 1f mrNA was the major transcript of the nNOS gene in human neuroblastoma *** analyzed a 1090 bp fragment of 1f promoter by TrANSFAC-TESS and Match softwares andluciferase assay,and found an important positive transcriptional regulation region that contained a putativenuclear factor(NF)-κB binding ***,using electrophoresis mobility shift and chromatinimmunoprecipitation assays,we identified this site to be the NF-κB responsive element,a crucial positiveregulator in the activation of the nNOS 1f *** together,our study identified an NF-κB responsiveelement within nNOS 1f promoter and showed that it plays an important role in the transactivation of nNOS1f mrNA,the major transcript of nNOS in SK-N-SH cells.

To investigate the effect of signal transducers and activators of transcription 1 （STAT1） antisense oligonucleotides （ASON） on concentrations of TNF-α IL-8, NO secreted by alveolar macrophages （AMs） in bleomycin-induced rat pulmonary fibrosis, five adult female Wistar rats were intratracheally instilled with bleomycin. After 7 days, the rats were killed by right ventricle of heart exsanguinations under ketamine anaesthesia and bronchoalveolar lavage （BAL） was performed to obtain AMs. AMs were divided into four groups, treated with STAT1 ASON, STAT1 sense oligonucleotides （SON）, dexamethasone （DEX） and medium alone （control）, respectively. AMs and media were collected after culture for 36 h. The mrNA and protein expressions of STAT1 and ICAM-1 in AMs were detected by rT-pCr and ELISA, respectively. The concentrations of TNF-α IL-8, NO in cultured medium were detected. The STAT1 mrNA expression by AMs in the STAT1 ASON group was lower than those of AMs in the STAT1 SON group, the DEX group and the control group （p 〈 0.05）. Moreover, the STAT1 mrNA expression by AMs in the DEX group was also lower than those of AMs in the STAT1 SON group and the control group （p 〈 0.05）, but the STAT1 mrNA expression by AMs in the STAT1 SON group was not different from that of the control group （p 〉 0.05）. The protein expressions of STAT1 and ICAM-1 and the mrNA expression of ICAM-1 showed similar changes to the STAT1 mrNA expression by AMs. The concentrations of TNF-α IL-8, NO in cultured medium from STAT1 ASON group were lower than those from STAT1 SON, DEX and the control groups （p 〈 0.05）. Moreover, the concentrations of TNF-α IL-8, NO in cultured medium from DEX group were also lower than those from the control and STAT1 SON group （p 〈 0.05）, but no difference between STAT1 SON group and the control （p 〉 0.05）. The results suggest that STAT1 ASON could inhibit the secretion of TNF-α IL-8, NO in AMs, and STAT1 could become a target of treating pulmonary fibrosis. Cellular ＆ Molecul
