A series of high-throughput binary cloning vectors were constructed to facilitate gene function analysis in higher plants. This vector series consists of plasmids designed for plant expression, promoter analysis, gene silencing, and green fluorescent protein fusions for protein localization. These vectors provide for high-throughput and efficient cloning utilizing sites for λ phage integrase/excisionase. In addition, unique restriction sites are incorporated in a multiple cloning site and enable promoter replacement. The entire vector series are available with complete sequence information and detailed annotations and are freely distributed to the scientific community for non-commercial uses.

Microwave irradiation, as opposed to formalin exposure, has not routinely been used in the preparation of killed vaccines despite the advantages of decreased chemical toxicity, ability to kill cells quickly, ease of completion requiring only a standard microwave, and potential increased protein conservation during irradiation. We evaluated the potential of microwave irradiation versus formalin fixation of bacteria to improve Streptococcus agalactiae vaccine efficacy in 5 gr fish by intraperitoneal (IP) inj.ction and bath immersion (BI). There was no significant difference in the cumulative percent mortality (CPM) post-challenge between fish administered microwave-killed cells (MKC) or formalin killed cells (FKC) within the BI (p S. agalactiae antibody activity. Thirty days after vaccination and j.st prior to challenge, the optical density (OD) levels of the FKC and MKC groups in the IP trials were significantly higher (p < 0.0001) than that of the TSB group. None of the groups in the BI trial exhibited significantly different OD levels post vaccination. Fourteen days after the challenge, the OD levels of all groups in both trials increased significantly above their pre-challenge levels. Both the FKC and MKC IP groups (p < 0.0001) and only the FKC BI group (p < 0.0351) had significantly increased OD level above that of the corresponding post-challenge TSB group. These results indicate that the FKC vaccine provides marginally greater protection and increased antibody concentrations than the MKC vaccine by BI and the MKC vaccine may become a non-chemical alternative to FKC in vaccination.

An existing weakly nonlinear diffusive instability hexagonal planform analysis for an interaction-diffusion plant-surface water model system in an arid flat environment [11] is extended by performing a rhombic planform analysis as well. In addition a threshold-dependent paradigm that differs from the usually employed implicit zero-threshold methodology is introduced to interpret stable rhombic patterns. The results of that analysis are synthesized with those of the existing hexagonal planform analysis. In particular these synthesized results can be represented by closed-form plots in the rate of precipitation versus the specific rate of plant density loss parameter space. From those plots, regions corresponding to bare ground and vegetative Turing patterns consisting of tiger bush (parallel stripes and labyrinthine mazes), pearled bush (hexagonal gaps and rhombic pseudo-gaps), and homogeneous distributions of vegetation, respectively, may be identified in this parameter space. Then that predicted sequence of stable states along a rainfall gradient is both compared with observational evidence and used to motivate an aridity classification scheme. Finally this system is shown to be isomorphic to the chemical reaction-diffusion Gray-Scott model and that isomorphism is employed to draw some conclusions about sideband instabilities as applied to vegetative patterning.

Two new types of signaling compounds have been discovered in wildfire smoke due to their ability to stimulate seed germination. The first discovered were karrikins, which share some structural similarity with the strigol- actone class of plant hormones, and both signal through a common F-box protein. However, karrikins and strigolactones operate through otherwise distinct signaling pathways, each distinguished by a specific a/13 hydrolase protein. Genetic analysis suggests that plants contain endogenous compounds that signal specifically through the karrikin pathway. The other active compounds discovered in smoke are cyanohydrins that release germination-stimulating cyanide upon hydrolysis. Cyanohydrins occur widely in plants and have a role in defense against other organisms, but an additional role in endogenous cyanide signaling should also now be considered.

In this article,we present a nanoelectromechanical system(NEMS)designed to detect changes in the Casimir *** Casimir effect is a result of the appearance of quantum fluctuations in an electromagnetic *** experiments have used nano-or microscale parallel plate capacitors to detect the Casimir force by measuring the small attractive force these fluctuations exert between the two *** this new set of experiments,we aim to directly detect the shifts in the Casimir energy in a vacuum due to the presence of the metallic parallel plates,one of which is a superconductor.A change in the Casimir energy of this configuration is predicted to shift the superconducting transition temperature(T_(c))because of the interaction between it and the superconducting condensation *** our experiment,we take a superconducting film,carefully measure its transition temperature,bring a conducting plate close to the film,create a Casimir cavity,and then measure the transition temperature *** expected shifts are smaller than the normal shifts one sees in cycling superconducting films to cryogenic temperatures,so using a NEMS resonator in situ is the only practical way to obtain accurate,reproducible *** a thin Pb film and opposing Au surface,we observe no shift in T_(c)>12µK down to a minimum spacing of~70 nm at zero applied magnetic field.

A cohort of 404 patients referred for hereditary neuropathy with liability to pressure palsies was tested initially for the common PMP22 whole gene deletion. 94 whole gene deletions were detected, plus three partial gene deletions, and the remaining 307 patients were screened for PMP22 point mutations. Nine point mutations were identified (8.5% of all mutations), eight of which were in exon 5, suggesting a point mutation hotspot for individuals with this condition. Sequencing analysis of PMP22 exon 5 should therefore be included as a routine diagnostic test for gene deletion-negative patients.

Escherichia coli O157:H7 is a human pathogen that was first identified from a foodborne outbreak in 1982, and in the 25 years that followed, many new strains were identified and emerged in numerous outbreaks of human disease. Extensive research has been conducted to identify virulence factor genes involved in the pathogenesis of E. coli O157:H7 and many genome sequences of E. coli O157:H7 strains have become available to the scientific community. Here, we provide a comprehensive overview of the research that has been conducted over the first 25 years to identify 394 known or putative virulence factor genes present in the genomes of E. coli O157:H7 strains. Finally, an examination of the conservation of these 394 virulence factor genes across additional genomes of E. coli O157:H7 is provided which summarizes the first 25 years and 13 genomes of this human pathogen.
