In t.is work, we t.st.d whet.er a.mela.in-media.0}d redox rea.t.on involving a.sorbed NO a.d O2 ca. couple NO oxida.0}on wit. O2 reduct.on t. form rea.t.ve oxygen species (ROS) or rea.t.ve nit.ogen species (RNS) in vit.o a.0}pH 7.4. We mea.ured t.e forma.0}on of rea.t.ve species t.a.0}oxidize dihydrorhoda.ine123 (DHR) t. fluorescent.rhoda.ine123 in t.e presence a.d a.sence of sepia.mela.in. In sepa.a.0} experiment., we monit.red NO concent.a.0}on wit. 4,5-dia.inofluorescein (Da.) by mea.uring t.e highly fluorescent.NO-a.duct. Da.-2t. We a.0}empt.d t. det.ct.peroxynit.it. wit. 5 μM 3-met.yl-1,2-cyclopent.nedione (MCP), a.select.ve sca.enger of peroxynit.it. (IC50 = 3.6 μM for ONOO- vs. 63.8 μM a.d >> 100 μM for NO a.d O2- respect.vely). However, MCP it.elf oxidized DHR. We found t.a.0}in t.e a.sence of NO, mela.in it.elf oxidizes DHR, wit. no loss of Da.-fluorescence (i.e. no net.consumpt.on of NO). In t.e presence of NO, t.ere wa. a.~ 57% loss of Da. fluorescence, indica.0}ng t.a.0}NOx is formed a.0}t.e expense of NO. t.e da.0} provided good fit.(r2 = 0.94) t. a.La.gmuir a.sorpt.on isot.erm, wit. pseudo first.order ra.0} k' = 8.2 × 107) s-1 a.d a.sorpt.on coefficient.Ka. = 4.04 M-1.
