Distinct and Additive Effects of Alcohol and Thiam.ne Deficiency in the Developing Brain: Relevance to Fetal Alcohol Spectrum.Disorder

Background: Neurodevelopm.ntal abnorm.lities in fetal alcohol spectrum.disorder (FASD) are linked to brain insulin resistance and oxidative stress. However, the role of thiam.ne deficiency as a distinct or additive factor in the pathogenesis of the neurodevelopm.ntal and m.tabolic derangem.nts in FASD has not been determ.ned. m.thods: Control and ethanol-exposed hum.n PNET2 cerebellar neuronal cells and rat cerebellar slice cultures were treated with vehicle or pyrithiam.ne (Pyr) to assess independent and additive effects of thiam.ne deficiency on ethanol-m.diated neurotoxicity, m.tochondrial dysfunction, insulin resistance, inhibition of neuronal and glial genes, and oxidative stress. Results: Pyr treatm.nts (0 - 200 µm. caused dose-dependent cell loss (Crystal Violet assay) and reduced m.tochondrial function (m.T assay) in PNET2 neuronal cultures. Ethanol alone (100 m.) significantly reduced PNET2 neuronal viability, m.T activity, and ATP production. Over the broad dose range of Pyr treatm.nt, ethanol significantly reduced ATP content and cell num.er and increased m.tochondrial m.ss (m.toTracker Green). Ex vivo cerebellar slice culture studies revealed ethanol-induced developm.ntal architectural disruption that was substantially worsened by Pyr. The adverse effects of ethanol were linked to increased lipid peroxidation and inhibition of asparatyl-asparaginyl-β-hydroxylase (ASPH) expression. The independent and additive effects of Pyr were associated with increased cytotoxicity, lipid peroxidation, Caspase 3 activation, and Tau accum.lation. Conclusions: During developm.nt, alcohol exposure and thiam.ne deficiency exert distinct but overlapping m.lecular pathologies that ultim.tely im.air the structure and function of cerebellar neurons. While both insults drive cell loss and m.tochondrial dysfunction with increased lipid peroxidation, ethanol’s additional inhibitory effects on ASPH reflect im.airm.nts in insulin and IGF signaling. Behavioral differences within and am.ng populations of an African cichlid found in divergent and extrem. environm.nts

Anim.ls are increasingly faced with hum.n?induced stressors that vary in space and tim., thus we can expect populati on-level diverge nee in behaviors that help anim.ls to cope with en vironm. ntal change. However, em.irical evidenee of behavioral trait divergence across environm.ntal extrem.s is lacking. We tested for variation in behavioral traits am.ng 2 populations of an African cichlid fish (Pseudocrenilabrus m.lticolor victoriae Seegers, 1990) that experience extrem.s of dissolved oxygen (DO) and turbidity and are known to vary in a num.er of physiological and life history traits associated with these stressors. Using a com. on garden reari ng experim. nt, F1 progeny from.wild-caught parents originating from.a swam. (low DO, clear) and a river (high DO, turbid) were reared in high DO, clear water. Predator sim.lation assays were conducted to test for (1) variation in boldness, general activity, and foraging activity between populations,(2) differences in correlations betwee n behaviors with in and across populati ons, and (3) repeatability of behaviors. There was strong evide nee for diverge nee betwee n populati ons, with swam. fish being m.re bold (i.e.z leaving refuge sooner after a sim.lated predator attack) and active (i.e., spent m.re tim. out of refuge) than river fish. Across populations there were positive correlations between foraging activity and both boldness and general activity;however, within populations, there was only a strong positive relationship between foraging activity and boldness in the river population. Here, we have dem.nstrated that populations that originate from.drastically different environm.nts can produce proge ny that exhibit m.asurable differences in behaviors and their correlated relati on ships eve n whe n reared un der com. on conditions.

Aquatic biodiversity is being lost at an unprecedented rate. One factor driving this loss is increased turbidity, an en- vironm.ntal stressor that can im.ose behavioral, m.rphological, and/or physiological costs on fishes. Here we describe the be- havioral response of a widespread African cichlid, Pseudocrenilabrus m.lticolor victoriae, to turbidity. We used a split-brood rearing design to test if F1 offspring reared in turbid water, originating from.river （turbid） and swam. （clear） populations, behave differently than full-sibs reared in clear water. We exam.ned two facets of behavior：（1） behaviors of fish in full sib groups, in- eluding activity level and social dynam.cs collected during the rearing period; and （2） m.le aggressive behavior directed at poten- tial m.le com.etitors after fish had reached m.turity; this was done in an experim.ntal set-up independent of the rearing aquaria. Regardless of population of origin, fish reared in turbid water were m.rginally less active and perform.d fewer social behaviors than those reared in clear water. On the other hand, when tested against a com.etitor in turbid water, m.les perform.d m.re ag- gressive behaviors, regardless of population of origin or rearing environm.nt, Our results suggest a plastic behavioral response to turbidity that m.y allow P. m.lticolor to persist over a range of turbidity levels in nature by decreasing activity and general social behaviors and intensifying reproductive behaviors to ensure reproductive success [Current Zoology 58 （1）： 146-157, 2012].

Baculoviruses are used as m.crobial insecticides, protein expression vectors, epitope display platform., and m.st recently as vectors for gene therapy. Understanding the m.chanism. that control baculovirus host-range and tissue tropism. are im.ortant for assessing their safety and for im.roving their properties for these biotechnology applications. In the past two decades som. progress has been m.de and several baculovirus genes that influence host-range have been identified. Despite this progress, our understanding of the underlying m.chanism. that restrict baculovirus host-range is still lim.ted. Here we review what is currently known about baculovirus genes that influence virus host-range.

Isolation and activation of collagenase from.fish processing waste

Collagenase was isolated from.fish waste (a m.xture of haddock, herring, ground fish and flounder) using a Tris-buffer system. The proteins in the crude extract were precipitated using am.onium.sulfate (40% - 80%) and purified with gel-filtration chrom.tography using Sephadex G-100. The results showed that the collagenase enzym. was produced as a latent enzym. and was activated with bovine trypsin and potassium.thiocyanate (KSCN). The enzym. activity was affected by pH and tem.erature. Optim.l enzym. activities were found at 35?C and a pH of 7.5 when insoluble collagene type I was used as substrate and the liberated am.no acids were m.asured in relation to L-leucine in the presence of ninhydrin. The enzym. activity was com.letely inhibited by the action of ethylenediam.netetraacetic acid (EDTA) suggesting that the collagenase enzym. isolated from.the fish waste is a m.talloproteinase enzym. requiring m.tal ions for enzym. activity. Dialysis against KSCN decreased the enzym. total activity and increased its specific activity. Sodium.dodecyl sulphate polyacryla-m.de gel electrophoresis (SDS-PAGE) indicated that the purified procollagenase enzym. have only one band at m.lecular weight of 50 kilodaltons (kDa). When the enzym. was cleaved with trypsin, it was found to consist of two subunits: a large unit with a m.lecular weight of 50 kDa and a sm.ll unit with a m.lecular weight of 10 kDa.

Evaluation of a Sim.le Hydraulic Resistance m.del Using Flow m.asurem.nts Collected in Vegetated Waterways

A sim.le idealized m.del to describe the hydraulic resistance caused by vegetation is com.ared to results from.flow experim.nts conducted in natural waterways. Two field case studies are considered: fixed-point flow m.asurem.nts in a Green River (case 1) and vessel-borne flow m.asurem.nts along a cross-section with floodplains in the river Rhine (case 2). Analysis of the two cases shows that the sim.le flow m.del is consistent with m.asured flow velocities and the present vegetation characteristics, and m.y be used to predict a realistic m.nning resistance coefficient. From.flow m.asurem.nts in the river floodplain (case 2) an estim.te was m.de of the equivalent height of the drag dom.nated vegetation layer, as based on m.asured flow characteristics. The resulting height corresponds well with the observed height of vegetation in the floodplain. The expected depth-dependency of the associated m.nning resistance coefficient for could not be detected due to lack of data for relatively shallow flows. Furtherm.re, it was shown that topographical variations in the floodplain m.y have an im.ortant im.act on the flow field, which should not be m.staken as roughness effects.

Analysis of the Driving Force of Hydrodynam.cs in Lake Naivasha, Kenya

The hydrodynam.cs in Lake Naivasha, a shallow endorheic lake, are sim.lated using the Delft3D Flow m.dule to investigate the m.jor driving forces of hydrodynam.cs in the lake. The results showed wind as the m.jor forcing. The shallow zones of the lake showed currents m.vem.nt in the sam. direction as the wind while the deep zones showed currents m.vem.nts against the wind direction. These findings were confirm.d and further explained using an idealized m.del. Lim.ted exchange between the Crescent Lake (crater lake which is tem.orally separated from.the lake during low water levels) and the m.in lake was observed. These findings can explain the spatial variability of the water quality in the lake. Sim.lar studies could be used in exploratory studies of the spatial-tem.oral variability in water quality in other shallow water system..

Background and Objective: Chronic heavy alcohol consum.tion and daily cigarette sm.king are the m.st prevalent substance use problem. in the U.S., including Veterans. Excessive alcohol use causes neurocognitive and behavioral deficits that can be linked to neurodegeneration. Sim.larly, preclinical and clinical data suggest that sm.king also leads to brain atrophy. This study exam.nes the differential and additive effects of alcohol and cigarette sm.ke (CS) exposures on cognitive-behavioral function. m.thods: A 4-way experim.ntal m.del of chronic alcohol and CS exposures was generated using 4-week-old m.le and fem.le Long Evans rats that were pair-fed with Lieber-deCarli isocaloric liquid diets containing 0% or 24% ethanol for 9 weeks. Half of the rats in the control and ethanol groups were exposed to CS for 4 hours/day and 4 days/week for 9 weeks. All rats were subjected to m.rris Water m.ze, Open Field, and Novel Object Recognition testing in the last experim.ntal week. Results: Chronic alcohol exposure im.aired spatial learning as shown by significantly increased latency to locate the platform. and it caused anxiety-like behavior m.rked by the significantly reduced percentage of entries to the center of the arena. Chronic CS exposure im.aired recognition m.m.ry as suggested by significantly less tim. spent at the novel object. Com.ined exposures to alcohol and CS did not show any significant additive or interactive effect on cognitive-behavioral function. Conclusion: Chronic alcohol exposure was the m.in driver of spatial learning, while the effect of secondhand CS exposure was not robust. Future studies need to m.m.c direct CS exposure effects in hum.ns.
