Determination of risperidone in human plasma by HPLC-MS/MS and its application to a pharmacokinetic study in Chinese volunteers

This study presents a rapid, specific and sensitive liquid chromatography/tandem mass spectrometry (LC-MS/MS) assay for determination of risperidone (rIS) in human serum using paroxetine as an internal standard (IS). An Alltima-C18 column (2.1 mm×100 mm, 3.μm) and a mobile phase consisting of 0.1% formic acid-acetonitrile (40:60, v/v) were used for separation. The analysis was performed by selected reaction monitoring (SrM) method, and the peak area of the m/z 411.3.191.1 transition for rIS was measured versus that of the m/z 3.0.1→192.1 transition for IS to generate the standard curves. The assay linearity of rIS was confirmed over the range 0.2.～50.00 ng/ml and the limit of quantitation was 0.05 ng/ml. The linear range corresponds well with the serum concentrations of the analytes obtained in clinical pharmacokinetic studies. Intraday and interday relative standard deviations were 1.85%～9.09% and 1.56%～4.3.%, respectively. The recovery of rIS from serum was in the range of 70.2.%～84.50%. The method was successfully applied to investigate the bioequivalence between two kinds of tablets (test versus reference products) in 18 healthy male Chinese volunteers. The result suggests that two formulations are bioequivalent.

The blood-spinal cord barrier plays a vital role in recovery after spinal cord *** neurovascular unit concept emphasizes the relationship between nerves and vessels in the brain,while the effect of the blood-spinal cord barrier on the neurovascular unit is rarely reported in spinal cord injury *** models of spinal cord injury were established by heavy object impact and then immediately injected with plateletderived growth factor(80μg/kg)at the injury *** results showed that after platelet-derived growth factor administration,spinal cord injury,neuronal apoptosis,and blood-spinal cord barrier permeability were reduced,excessive astrocyte proliferation and the autophagyrelated apoptosis signaling pathway were inhibited,collagen synthesis was increased,and mouse locomotor function was *** vitro,human umbilical vein endothelial cells were established by exposure to 2.0μM *** 2.hours prior to injury,in vitro cell models were treated with 5 ng/mL platelet-derived growth *** results showed that expression of blood-spinal cord barrier-related proteins,including Occludin,Claudin 5,andβ-catenin,was significantly decreased and autophagy was significantly ***,the protective effects of platelet-derived growth factor could be reversed by intraperitoneal injection of 80 mg/kg chloroquine,an autophagy inhibitor,for 3.successive days prior to spinal cord *** findings suggest that platelet-derived growth factor can promote endothelial cell repair by regulating autophagy,improve the function of the blood-spinal cord barrier,and promote the recovery of locomotor function post-spinal cord *** for animal experiments was obtained from the Animal Ethics Committee,Wenzhou Medical university,China(approval ***2.18-0043.in July 2.18.

目的综合评估Elecsys血清异常凝血酶原(PIVKA-Ⅱ)检测试剂在中国人群中辅助诊断肝细胞癌(HCC)的临床价值。方法研究采用多中心病例-对照设计,连续收集首次确诊且未经治疗的HCC患者样本、良性肝病患者样本和干扰性样本进行Elecsys PIVKA-Ⅱ与甲胎蛋白(AFP)检测,并对受试者的多项临床信息进行汇总分析。计算特定阳性截断值下PIVKA-Ⅱ(2..2. ng/ml)和AFP(400 ng/ml)用于HCC诊断的效能。采用Kruskal-Wallis检验或受试者操作特征曲线进行统计学分析。结果最终在5家中心共纳入448例受试者,其中包括185例HCC病例。以良性肝病组为对照组时,PIVKA-Ⅱ的诊断灵敏度和准确率均高于AFP:84.86%对比3..81%和89.01%对比63.66%,而特异度略低。敏感性分析显示在该特定阳性截断值下,PIVKA-Ⅱ的临床性能在AFP阴性受试者亚组、不同病因亚组及多个巴塞罗那癌症分期的HCC患者亚组(含早期HCC)中的灵敏度均>80%。同时,PIVKA-Ⅱ的受试者操作特征曲线下面积亦略高于AFP(0.92.0对比0.8809)。结论Elecsys PIVKA-Ⅱ在中国人群中的临床性能良好且稳定,有潜力改善临床AFP阴性HCC诊断和早期HCC监测中的漏诊现状以进一步提升中国人群HCC精准辅助诊断效率。

Artificial intelligence(AI) has played a significant role in imitating and producing large-scale designs such as e-commerce banners. However, it is less successful at creative and collaborative design outputs. Most humans express their ideas as rough sketches, and lack the professional skills to complete pleasing paintings. Existing AI approaches have failed to convert varied user sketches into artistically beautiful paintings while preserving their semantic concepts. To bridge this gap, we have developed Smart Paint, a co-creative drawing system based on generative adversarial networks(GANs), enabling a machine and a human being to collaborate in cartoon landscape painting. Smart Paint trains a GAN using triples of cartoon images, their corresponding semantic label maps, and edge detection maps. The machine can then simultaneously understand the cartoon style and semantics, along with the spatial relationships among the objects in the landscape images. The trained system receives a sketch as a semantic label map input, and automatically synthesizes its edge map for stable handling of varied sketches. It then outputs a creative and fine painting with the appropriate style corresponding to the human’s sketch. Experiments confirmed that the proposed Smart Paint system successfully generates high-quality cartoon paintings.

In order to predict the main species of veterinary drugs in the environment, a liquid chromatogra-phy-tandem mass spectrometry （LC-MS/MS） method is described in this paper. The method uses the reversed-phase column of C 8 column, the HLB solid phase extraction cartridge, the 0.1% formic acid acetoni-trile-water solution mobile phase with gradient elution to determine ciprofloxacin （CPFX）, enrofloxacin （ErFX） and florfenicol （FF） in animal wastewater and soil. The flow rate was 0.3. mL/min. The detection mode was multiple reaction monitoring （MrM） with positive ionization mode for CPFX and ErFX and negative ion mode for FF, respectively. The injection volume was 10 μL. The quantification limit of the method （MQL） was 2.ng/L for FF and 40ng/L for CPFX and ErFX in animal wastewater samples. In soil samples, MQL was 40μg/kg for FF and 50μg/kg for CPFX and ErFX. The correlation coefficient was higher than 0.99. recoveries of CPFX, ErFX and FF at fortified levels were higher than 67%, with relative standard deviation of 0.72.5.81%.

Prognostic value of serum soluble Fas in patients with locally advanced unresectable rectal cancer receiving concurrent chemoradiotherapy

Objective: This study was designed to detect the changes of serum soluble Fas (sFas) levels in patients with locally advanced unresectable rectal cancer (LAUrC),and to explore its prognostic value of ***: Soluble samples were obtained from LAUrC subjects,treated by concurrent chemoradiotherapy,before treatment and one month after *** donor serum samples were used as *** concentration was measured by enzyme-linked immunosorbent assay (ELISA).results: The sFas levels before treatment and one month after treatment were both significantly higher in LAUrC subjects than in healthy controls [(8.79±1.3.) and (7.74±1.3.4} vs.(5.53.1.13. ng/L,Pre treatment and one month after treatment were significantly lower in the response group (complete and partial responses) than in the non-response group (stable and progressive diseases) [(8.50±1.2.) vs.(10.17±1.2.) ng/L,P2.) vs.(8.90±1.13. ng/L,Prespectively].The one-year survival rate was 54.2. and 82.6% in those with sFas levels >8.79 ng/L and ng/L before treatment (P2.,respectively,50.0% and 87.0% in those with sFas levels >7.74 ng/L and ng/L one month after treatment (Pr in LAUrC subjects than in healthy *** chemoradiotherapy can reduce sFas levels in LAUrC *** monitoring of sFas may provide prognostic information for LAUrC patients.
