Oleanolic acid improved intestinal immune function by activating and potentiating bile acids receptor signaling in E. coli-challenged piglets

Background Infection with pathogenic bacteria during nonantibiotic breeding is one of the main causes of animal intestinal *** acid(OA)is a pentacyclic triterpene that is ubiquitous in *** previous work demonstrated the protective effect of OA on intestinal health,but the underlying molecular mechanisms remain *** study investigated whether dietary supplementation with OA can prevent diarrhea and intestinal immune dysregulation caused by enterotoxigenic Escherichia coli(ETEC)in *** key molecular role of bile acid receptor signaling in this process has also been *** Our results demonstrated that OA supplementation alleviated the disturbance of bile acid metabolism in ETEC-infected piglets(P<0.05).OA supplementation stabilized the composition of the bile acid pool in piglets by regulating the enterohepatic circulation of bile acids and significantly increased the contents of UDCA and CDCA in the ileum and cecum(P<0.05).This may also explain why OA can maintain the stability of the intestinal microbiota structure in ETEC-challenged *** addition,as a natural ligand of bile acid receptors,OA can reduce the severity of intestinal inflammation and enhance the strength of intestinal epithelial cell antimicrobial programs through the bile acid receptors TGR5 and FXR(P<0.05).Specifically,OA inhibited NF-κB-mediated intestinal inflammation by directly activating TGR5 and its downstream c AMP-PKA-CREB signaling pathway(P<0.05).Furthermore,OA enhanced CDCA-mediated MEK-ERK signaling in intestinal epithelial cells by upregulating the expression of FXR(P<0.05),thereby upregulating the expression of endogenous defense molecules in intestinal epithelial *** In conclusion,our findings suggest that OA-mediated regulation of bile acid metabolism plays an important role in the innate immune response,which provides a new diet-based intervention for intestinal diseases caused by pathogenic bacterial infections in pig

AIM:To investigate the types of bacteria in patients with eye infections in Suzhou and their drug resistance to commonly used antibacterial ***:The clinical data of 155 patients were retrospectively collected in this study,and the pathogenic bacteria species and drug resistance of each pathogenic bacteria were ***:Among the 155 patients(age from 12 to 87 years old,with an average age of 57,99 males and 56 females)with eye infections(160 eyes:74.in the left eye,76 in the right eye and 5 in both eyes,all of which were exogenous),71(4..81%)strains were gram-positive bacteria,23(14.84.)strains were gram-negative bacteria and 61(39.35%)strains were ***-positive bacteria were highly resistant to penicillin and erythromycin(78.87%and 4..4.%respectively),but least resistant to vancomycin at ***-negative bacteria were highly resistant to cefoxitin and compound sulfamethoxazole(100%and 95.65%respectively),but least resistant to meropenem at *** of the resistance of gram-positive and gram-negative bacteria to some drugs revealed statistically significant differences(P<0.05)in the resistance of both to cefoxitin,cotrimoxazole,levofloxacin,cefuroxime,ceftriaxone and ceftazidime,and both had higher rates of resistance to gram-negative bacteria than to gram-positive *** distribution of bacterial infection strains showed that Staphylococcus epidermidis was the most common strain in the conjunctiva,cornea,aqueous humor or vitreous body and other eye ***,Fusarium and Pseudomonas aeruginosa were also among the most common strains of conjunctival and corneal ***:Gram-positive bacteria are the dominant bacteria in eye infections,followed by gram-negative bacteria and *** the resistance of gramnegative bacteria to multiple drugs,monitoring of bacteria should be strengthened in eye bacterial infections for effective prevention and control to reduce complications caused by eye infections.

Hot-extrusion and cold-rolling were conducted on Mg-Ce binary alloys to explore the effect of cerium(Ce)on microstructure,texture and mechanical properties of Mg *** addition of Ce results in significant grain refinement both in as-cast and hot-extruded *** basal texture is also weakened after extrusion and an inclined basal texture is formed with Ce *** strength and elongation of the Mg-Ce alloys are improved simultaneously due to such grain refinement and texture weakening *** cold-rolling,plenty of twins are found in the pure Mg and AZ31 plates while grains in the Mg-0.3Ce plate deform more uniformly without lamellar twin ***,Mg-0.3Ce alloys own strong and continuous strain hardening because Ce atoms and Mg-Ce precipitated phases serve as obstacles for dislocation slip.

Growing evidences indicate that dysfunction of autophagy contributes to the disease pathogenesis of amyotrophic lateral sclerosis(ALS)and frontotemporal dementia(FTD),two neurodegenerative *** GGGGCC·GGCCCC repeat RNA expansion in chromosome 9 open reading frame 72(C9orf72)is the most genetic cause of both ALS and *** to the previous studies,GGGGCC·GGCCCC repeat undergoes the unconventional repeat-associated non-ATG translation,which produces dipeptide repeat(DPR)*** there is a growing understanding that C9orf72 DPRs have a strong ability to harm neurons and induce C9orf72-linked ALS/FTD,whether these DPRs can affect autophagy remains *** the present study,we find that poly-GR and poly-PR,two arginine-containing DPRs which display the most cytotoxic properties according to the previous studies,strongly inhibit starvation-induced ***,our data indicate that arginine-rich DPRs enhance the interaction between BCL2 and BECN1/Beclin 1 by inhibiting BCL2 phosphorylation,therefore they can impair autophagic clearance of neurodegenerative disease-associated protein aggregates under starvation condition in ***,our study not only highlights the role of C9orf72 DPR in autophagy dysfunction,but also provides novel insight that pharmacological intervention of autophagy using SW063058,a small molecule compound that can disrupt the interaction between BECN1 and BCL2,may reduce C9orf72 DPR-induced neurotoxicity.

New formula for predicting the plastic buckling pressure of steel torispherical heads under internal pressure

Thin-walled torispherical heads under internal pressure can fail by plastic buckling because of compressive circumferential stresses in the head ***,existing formulas still have limitations,such as complicated expressions and low accuracy,in determining buckling *** this paper,we propose a new formula for calculating the buckling pressure of torispherical heads based on elastic-plastic analysis and experimental ***,a finite element(FE)method based on the arc-length method is established to calculate the plastic buckling pressure of torispherical heads,considering the effects of material strain hardening and geometrical *** buckling pressure results calculated by the FE method in this paper have good consistency with those of BOSOR5,which is a program for calculating the elastic-plastic bifurcation buckling pressure based on the finite difference energy ***,the effects of geometric parameters,material parameters,and restraint form of head edge on buckling pressure are ***,a new formula for calculating plastic buckling pressure is developed by fitting the curve of FE results and introducing a reduction factor determined from experimental ***,based on the experimental results,we compare the predictions of the new formula with those of existing *** is shown that the new formula has a higher accuracy than the existing ones.
