In this work,we proposed a ratiometric silver nanoclusters(AgNCs)fluorescent assay by designing a bifunctional-blockeraided hybridization chain reaction(HCR).Hairpin probe 1(HP1)containing two special DNA fragments(5′-CAC CGC T-3.and 5′-ATT TGC CTT TTG GGG ACG GATA-3.)at two terminals creates a red-emitting AgNC nucleation sequence(rNS,5′-CAC CGC TAT TTG CCT TTT GGG GAC GGATA-3.).We found that the presence of a toehold fragment(5′-TGCCC-3.)in HP1 could silence the *** the addition of a target nucleic acid,HCR of HP1 and hairpin probe 2(HP2)could be initiated,resulting in the formation of long chain of DNA duplexes with multibranched *** the toehold fragment in HP1participated in generating duplexes,a strong emission of rNS-templated AgNCs was observed at 670 *** significantly,a bifunctional blocker was introduced not only to reduce the background red-emitting fluorescence but also to play as an internal green-emitting AgNCs nucleation *** the one hand,the blocker could increase the signal-to-noise-ratio of the constructed biosensor,and on the other hand,the blocker also helped to prepare ratiometric HCR-AgNCs assay with self-calibrating ability to strengthen its *** with the traditional HCR-AgNCs sensors,the developed ratiometric assay based on the bifunctional-blocker-aided HCR has higher reliability,which is important for the fabrication of biosensors in various fields for practical biosensing applications.

Background: Hepatic ischemia-reperfusion injury (HIRI) stands as an unavoidable complication arising from liver surgery, profoundly intertwined with its prognosis. The role of lysine methyltransferase SET domain bifurcated 1 (SETDB1) in HIRI remains elusive, despite its confirmation as a potential therapeutic target for diverse diseases. Here, we investigated the mechanism by which SETDB1 regulated HIRI. Methods: RNA sequencing data were used to identify the expression and potential targets of SETDB1 through bioinformatics analysis. To elucidate the impact of SETDB1 on HIRI, both an in vivo model of HIRI in mice and an in vitro model of hepatocyte hypoxia/reoxygenation were established. Biochemical and histological analyses were used to investigate the influence of SETDB1 on liver damage mediated by HIRI. Chromatin immunoprecipitation and coimmunoprecipitation were implemented to explore the in-depth mechanism of SETDB1 regulating HIRI. Results: We confirmed that hepatocellular SETDB1 was up-regulated during HIRI and had a close correlation with HIRI-related inflammation and apoptosis. Moreover, inhibition of SETDB1 could mitigate HIRI-induced liver damage, inflammation, and apoptosis. Through our comprehensive mechanistic investigation, we revealed that SETDB1 interacts with apoptosis-signal-regulating kinase 1 (ASK1) and facilitates the methylation of its lysine residues. Inhibition of SETDB1 resulted in reduced phosphorylation of ASK1, leading to a marked suppression of downstream c-Jun N-terminal kinase (JNK)/p3. signaling pathway activation. The therapeutic effect on inflammation and apoptosis achieved through SETDB1 inhibition was nullified by the restoration of JNK/p3. signaling activation through ASK1 overexpression. Conclusions: The findings from our study indicate that SETDB1 mediates lysine methylation of ASK1 and modulates the activation of the ASK1–JNK/p3. pathway, thus involved in HIRI-induced inflammation and apoptosis. Precipitation projections over the Tibetan Plateau(TP)show diversity among existing studies,partly due to model *** to develop a reliable projection remains ***,based on the IPCC AR6–assessed likely range of equilibrium climate sensitivity(ECS)and the climatological precipitation performance,the authors constrain the CMIP6(phase 6 of the Coupled Model Intercomparison Project)model projection of summer precipitation and water availability over the *** best estimates of precipitation changes are 0.24,0.25,and 0.45 mm d^(−1)(5.9%,6.1%,and 11.2%)under the Shared Socioeconomic Pathway(SSP)scenarios of SSP1–2.6,SSP2–4.5,and SSP5–8.5 from 2050–2099 relative to 1965–2014,*** corresponding constrained projections of water availability measured by precipitation minus evaporation(P–E)are 0.10,0.09,and 0.22 mm d^(−1)(5.7%,4.9%,and 13.2%),*** increase of precipitation and P–E projected by the high-ECS models,whose ECS values are higher than the upper limit of the likely range,are about 1.7 times larger than those estimated by constrained ***,there is a larger increase in precipitation and P–E over the eastern TP,while the western part shows a relatively weak difference in precipitation and a drier trend in P–*** wetter TP projected by the high-ECS models resulted from both an approximately 1.2–1.4 times stronger hydrological sensitivity and additional warming of 0.6℃–1.2℃ under all three scenarios during 2050–*** study emphasizes that selecting climate models with climate sensitivity within the likely range is crucial to reducing the uncertainty in the projection of TP precipitation and water availability changes.

Non-destructive and accurate inspection of gallium nitride light-emitting diode(Ga N-LED)epitaxial wafers is important to Ga N-LED ***,the conventional electroluminescence inspection,the photoluminescence inspection,and the automated optical inspection cannot fulfill the complex technical *** this work,an inspection method and an operation system based on soft single-contact operation,namely,single-contact electroluminescence(SC-EL)inspection,are *** key component of the SC-EL inspection system is a soft conductive probe with an optical fiber inside,and an AC voltage(70Vpp,100 k Hz)is applied between the probe and the ITO electrode under the LED epitaxial *** proposed SC-EL inspection can measure both the electrical and optical parameters of the LED epitaxial wafer at the same time,while not causing mechanical damage to the LED epitaxial ***,it is demonstrated that the SC-EL inspection has a higher electroluminescence wavelength accuracy than photoluminescence *** results show that the non-uniformity of SC-EL inspection is 444.64%,which is much lower than that of photoluminescence *** addition,the obtained electrical parameters from SC-EL can reflect the reverse leakage current(Is)level of the LED epitaxial *** proposed SC-EL inspection can ensure high inspection accuracy without causing damage to the LED epitaxial wafer,which holds promising application in LED technology.

Vitamin E(VE) is an essential lipid-soluble vitamin that improves the fish flesh quality. However, the underlying molecular mechanisms remain unclear. This study aimed to investigate the effects of VE on growth performance and flesh quality in sub-adult grass carp(Ctenopharyngodon idella). A total of 450fish(713.53.± 1.50 g) were randomly divided into six treatment groups(three replicates per treatment)and fed for nine weeks with different experimental diets(dietary lipid 47.8 g/kg) that contained different levels of VE(5.44, 52.07, 96.85, 141.71, 185.66, and 23..12 mg/kg diet, supplemented as dl-a-tocopherol acetate). Notably, the treatment groups that were fed with dietary VE ranging from 52.07 to23..12 mg/kg diet showed improvement in the percent weight gain, special growth rate, and feed efficiency of grass carp. Moreover, the treatment groups supplemented with dietary VE level of 141.71,185.66, and 23..12 mg/kg diet showed enhancement in crude protein, lipid, and a-tocopherol contents in the muscle, and the dietary levels of VE ranging from 52.07 to 141.71 mg/kg diet improved muscle pH24h and shear force but reduced muscle cooking loss in grass carp. Furthermore, appropriate levels of VE(52.07 to 96.85 mg/kg diet) increased the muscle polyunsaturated fatty acid content in grass carp. Dietary VE also increased the mRNA levels of fatty acid synthesis-related genes, including fas, scd-1, fad,elovl, srebp1, pparγ, and lxra, and up-regulated the expression of SREBP-1 protein. However, dietary VE decreased the expression of fatty acid decomposition-related genes, including hsl, cpt1, acox1, and ppara,and endoplasmic reticulum stress-related genes, including perk, ire1, atf6, eif2a, atf4, xbp1, chop, and grp78, and down-regulated the expression of p-PERK, p-IRE1, ATF6, and GRP78 proteins. Recent advancements in constructed wetlands(CWs)have highlighted the imperative of enhancing nitrogen(N)removal ***,the variability in influent substrate concentrations presents a challenge in optimizing N removal strategies due to its impact on removal efficiency and *** we show the interplay between influent substrate concentration and N removal processes within integrated vertical-flow constructed wetlands(IVFCWs),using wastewaters enriched with NO_(3.^(-)-N and NH4þ-N at varying carbon to nitrogen(C/N)ratios(1,3.and 6).In the NO_(3.^(-)-N enriched systems,a positive correlation was observed between the C/N ratio and total nitrogen(TN)removal efficiency,which markedly increased from 13.46±2.23.to 87.00±2.3.%as the C/N ratio escalated from 1 to ***,in NH4þ-N enriched systems,TN removal efficiencies in the A-6 setup(3..69±4.83.)were marginally 1.25 to 1.29 times higher than those in A-3.and A-1 systems,attributed to constraints in dissolved oxygen(DO)levels and *** community analysis and metabolic pathway assessment revealed that anaerobic denitrification,microbial N assimilation,and dissimilatory nitrate reduction to ammonium(DNRA)predominated in NO_(3.^(-)-N systems with higher C/N ratios(C/N 3..In contrast,aerobic denitrification and microbial N assimilation were the primary pathways in NH4þ-N systems and low C/N NO_(3.^(-)-N systems.A mass balance approach indicated denitrification and microbial N assimilation contributed 4.12-47.12%and 8.51e3..96%in NO_(3.^(-)-N systems,respectively,and 0.55e17.3.%and 7.83.3..55%in NH4þ-N systems to TN *** enhance N removal,strategies for NO_(3.-N dominated systems should address carbon source limitations and electron competition between denitrification and DNRA processes,while NH4þ-N dominated systems require optimization of carbon utilization pathways,and ensuring adequate DO and alkalinity supply.
